Antiviral activity of liposomal preparations of antibiotic geliomycin.

A liposomal preparation with maximally possible content of incorporated geliomycin was obtained. Its cytotoxicity was studied in a culture of human embryonal diploid fibroblasts. Antiviral activity was studied on a model of cytomegaloviral infection in vitro by the capacity to plaque formation. Liposomal geliomycin was 10-fold less toxic for human cells than the solution of the antibiotic in DMSO and exhibited antiviral activity towards cytomegaloviral infection at a concentration of 0.042 microg/ml.

[In vitro antiviral activity of fullerene amino acid derivatives in cytomegalovirus infection].

The in vitro effect of 5 water soluble fullerene C60 amino acid derivatives (FAD) on the development of cytomegalovirus infection was studied in the schemes of the therapeutic, prophylactic and virucidal action. The following compounds as FAD were used: fullerene conjugated with Na salt of gamma-aminobutyric acid (C60-ABA-Na), 2 derivatives based on Na salts of fullerene-gamma-aminobutyric acid and fullerene-omega-caproic acid (C60-ABA-OH-Na and C60-ACA-OH-Na respectively) and 2 derivatives based on methyl ethers of the above mentioned fullerene amino acids (C60-ABA-OH-CH3 and C60-ACA-OH-CH3). All the FAD were able to inhibit the development of the virus cytopathogenic action in the cell culture. However, the compounds had different antiviral properties. C60-ABA-OH-Na, C60-ABA-CH3 and C60-ACA-CH3 showed marked antiviral activity in the prophylactic scheme. 50-Percent inhibition of the virus cytopathogenic action (ID50) was observed when concentrations of the compounds were 0.31, 5 and 25 mcg/ml respectively. C60-ACA-OH-Na inhibited the development of cytomegalovirus infection in the cell culture only in the scheme of the therapeutic action (ID50 4 mcg/ml). C60-ABA-Na had the highest antiviral effect. In a concentration of 0.22 mcg/ml it inhibited the cytomegalovirus plague-forming capacity by 50% in both the prophylactic and the virucidal schemes. The chemotherapeutic index of the compound was within the limits of 2500 to 5450.

[Activation of transcription of ribosome genes following human embryo fibroblast infection with cytomegalovirus in vitro].

Cytomegalovirus (CMV) infection of human diploid embryo fibroblasts in vitro causes a massive cell death on the 3-4th day of infection with a high primary infection coefficient (1-5 U/cell). Cytopathological effects of viral infection on the 3-4th day includes diminishes of the cell size, changes in their form, compaction of the nuclear chromatin, and disorganization and inactivation of the nucleolus. However, the early stages of the viral infection progression (the 1st-2nd day) are accompanied by unequivocal activation of rDNA (ribosomal gene) and the bulk of chromatin transcription. There are several features to support this conclusion: In the early CMV-infected cell 1) the nucleolar size is increased; 2) the number of intranucleolar foci binding the specific RNA-polymerase I transcription factor (UBF) is augmented; 3) the Ag-NOR staining is enhanced; 4) 3H-uridine incorporation to the nucleoli is activated; 5) the ultrastructure of the nucleolus is changed. Altogether, these data argue in favor of activation of rDNA transcription in human fibroblasts in vitro at the initial stages of infection.

[Gout: current views. Stage oriented treatment].

Gout is a common disease arising due to abnormal purin metabolism and excessive accumulation of uric acid in the blood (hyperuricemia) and manifesting with attacks of acute gouty arthritis. In long duration of gout uric acids accumulate in the bones and periarticular tissues as tophuses. Repeat attacks lead to development of chronic gouty arthritis. Purins restriction diet is an important component of gout treatment. Treatment of acute arthritis should be started early, in initial pains before the development of the attacks. Gouty arthritis in the presence of continuous hyperuricemia, tophyses and urolithiasis is treated with allopurinol. Its intake should be long and controlled by the blood level of uric acid. Balneotherapy is recommended for patients with chronic gouty arthritis associated with cardiovascular diseases, urolithiasis.

[The prognosis and control of the efficacy of heparin therapy in patients with diabetic glomerulosclerosis].

3-week heparin treatment (25000 U/day) of 38 patients with diabetic glomerulosclerosis (DGS) produced positive results in 31 of them, was ineffective in 5 patients and induced complications in 2 cases. Prognostic criteria of heparin treatment efficacy basing on hemostatic changes on day 7 have been developed. Hemostasis was controlled most effectively in patients receiving heparin by a new graphic turbidimetric method capable of objective registration of clotting and fibrinolysis in one plasma portion. Because individual sensitivity to heparin widely varies and this variability manifests as early as first days of treatment, control over hemostasis system in heparin day dose 25000 U/day is advisable before treatment, on treatment day 3, 7 and on aftertreatment day 2.

[Changes in thrombocytic hemostasis in patients with diabetic glomerulosclerosis].

Indices of thrombocytic hemostasis were determined in 40 patients with diabetic glomerulosclerosis. It was found that patients suffering of diabetic glomerulosclerosis with chronic renal insufficiency showed signs of hypercoagulemia while changes of the thrombocytic hemostasis become of diverse values. There was a high correlation of hemostasis changes and clinical manifestations of diabetes mellitus and diabetic glomerulosclerosis. Three tests are recommended for treatment choice: hemolysate-aggregation test, determination of soluble complexes of monomer fibrin and fibrin-splitting product.

[Biological effect of brown coal dust in experimental studies].

For 4 months laboratory white rats were administered inhalations of polydisperse dusts of the brown coal from the Nazarovsky and Beregovsky coal deposits. Revealed were mainly identical, early marked reactions of the bronchi and lung system, which, judging from the quantitative indices, could be regarded as ++pre-fibrosis, as well as some signs of irritation on the cell elements, bronchi and lung vessels. The portion of lipids and general oxyproline in the dry lung substance in the sample group amounted to 16.3-17.2%, and 9.2% in the control group. The results in combination with the immune indices changes (complement, lysozyme, lymphocyte populations), histamine in the blood and non-specific shifts in the adrenal gland state and in the heart, proved the unfavourable action of ducts with low ash content and a minimal (no less than 1.3%) content of free silica dioxide. The reasons underlying brown coal's action in question are also thoroughly discussed.

[A comparative evaluation of methods for determining the activity of rheumatic diseases].

The blood concentration of soluble complexes of monomere fibrin (SCMF), fibrin/fibrinogen splitting products (FSP) and sigma-ESB were determined in 11 patients with rheumatic diseases showing an articular syndrome. The authors revealed the diagnostic significance of SCMF in arthritis of any etiology and FSP in rheumatic diseases with systemic manifestations.

[Hygienic assessment of working conditions for schoolchildren at video terminals during their academic studies].

Proceeding from the studies on the conditions of schoolchildren's work with video terminals during school lessons there was developed a complex of hygienic measures aimed at reduction of impact of factors specified by the work with video terminals and by anthropogenic pollution. Requirements to design of classrooms equipped by video terminals were listed and optimal and permissible ergonomic characteristics of the workplace established along with air temperature and humidity.

[The fibrinogen-fibrin-fibrinolysis system in rheumatoid arthritis].

The enzymatic, nonenzymatic and XIIa-dependent fibrinolysis was inhibited. In RA without systemic manifestations the main cause of appearance of soluble complexes of monomeric fibrins (SCMF) in the blood is a local activation of coagulation in the inflamed joint tissues. It is suggested that an increase of SCMF concentration in the blood in patients with RA is a reliable and dynamic index of the activity of the inflammatory process.

[Indices of the blood fibrinolytic system in osteoarthrosis and rheumatoid arthritis].

The fibrinolytic system of the blood was studied in 46 patients with osteoarthrosis (OA) associated with reactive synovitis or without it, 44 patients with rheumatoid arthritis (RA) without visceral manifestations. It was found that a slowing of enzymatic, nonenzymatic and XIIa-dependent fibrinolysis is typical of RA but not of OA. Products of fibrin breaking in the general blood flow were absent in patients with OA but may be observed in RA. Increase of the level of fibrin monomers (FM) in the blood are observed in OA with reactive synovitis. The concentration of FM in the blood in RA and OA with reactive synovitis is an informative and dynamic sign of the activity of the inflammatory process and efficacy of treatment.
