Lignin peroxidase: toward a clarification of its role in vivo.

The extracellular lignin peroxidase from the white-rot basidiomycete Phanerochaete chrysosporium is thought to play an important role in lignin biodegradation. However, the majority of lignin-derived preparations actually experience overall polymerization at the hands of the enzyme in vitro. It has now been found that, in the presence of H2O2 at pH 4.0, the monomeric lignin precursor coniferyl alcohol is polymerized quantitatively by a lignin peroxidase preparation which is uncontaminated with MnII-dependent peroxidases. 13C NMR spectrometry of the resulting dehydropolymerisates from 13C-labeled monolignols confirms that the frequencies of different interunit linkages are very similar to those engendered through the action of horseradish peroxidase with H2O2. Indeed, lignin peroxidase does not ultimately seem to be a prerequisite for lignin degradation in vivo, yet its activity can still accelerate the conversion of lignin-derived preparations by P. chrysosporium to CO2. Consequently, lignin peroxidase can provisionally be expected to fulfill two important functions. On the one hand, the enzyme may detoxify lower molecular weight phenolic compounds released from lignins during their fungal decomposition. On the other hand, through the introduction of suitable functional groups, lignin peroxidase could indirectly enhance the susceptibility of macromolecular lignin structures toward depolymerization by another enzyme.

Formation of cis-coniferin in cell-free extracts of Fagus grandifolia Ehrh bark.

American beech (Fagus grandifolia Ehrh) bark exclusively accumulates cis-monolignols and their glucosidic conjugates; no evidence for the accumulation of trans-monolignols has been found. The glucosyltransferase from this source exhibits a very unusual substrate specificity for cis, and not trans, monolignols. This is further evidence that cis monolignols are involved in lignin formation in these plant tissues. Preliminary evidence for the existence of a novel trans-cis monolignol isomerase was obtained, in agreement with our contention that this isomerization is not photochemically mediated.

Exclusive accumulation of Z-isomers of monolignols and their glucosides in bark of Fagus grandifolia.

In addition to Z-coniferyl and Z-sinapyl alcohols, bark extracts of Fagus grandifolia also contain significant amounts of the glucosides, Z-coniferin, Z-isoconiferin (previously called faguside) and Z-syringin. The corresponding E-isomers of these glucosides do not accumulate to a detectable level. The accumulation of the Z-isomers suggests that either they are not lignin precursors or that they are reservoirs of monolignols for subsequent lignin biosynthesis; it is not possible to distinguish between these alternatives. The co-occurrence of Z-coniferin and Z-isoconiferin demonstrate that glucosylation of monolignols can occur at either the phenolic or the allylic hydroxyl groups.

Lignin degradation by peroxidase in organic media: A reassessment.

The effect of horseradish peroxidase/H(2)O(2) in organic medium (dioxane/aqueous buffer, pH 5, 95:5) on the depolymerization of synthetic (dehydrogenatively polymerized) lignin was reinvestigated. In contrast to previous claims [Dordick, J. S., Marletta, M. A. & Klibanov, A. M. (1986) Proc. Natl. Acad. Sci. USA 83, 6255-6257], our results demonstrated that vigorous depolymerization of this substrate did not occur. Further, during this treatment ferulic acid was not a significant biodegradation product.

Synthesis of stereospecifically deuterated desoxypodophyllotoxins and 1H-nmr assignment of desoxypodophyllotoxin.

[4 beta- 2H1]Desoxypodophyllotoxin [3], [4 alpha- 2H1]desoxypodophyllotoxin [4], and [4, 4- 2 H2]desoxypodophyllotoxin [9] were prepared from podophyllotoxin [1] via its chloride [5]. A complete assignment of the 1H-nmr spectrum of desoxypodophyllotoxin [2] was made on the basis of the spectra of the deuterated compounds [3] and [4].
