[Cerebrospinal fluid sorption in patients with heroin addiction].

The sorption of cerebrospinal fluid (CSF) was attempted as a special detoxifying procedure in a group of sixty heroine addicts. CSF contents of cells, total protein, nucleic acids and interleukin-1 (IL-1), as well as acridine orange (AO) binding to CSF cells were determined before and after the procedure. The treatment provided immediate clinical improvement for 70% of the patients. Clinical effect was accompanied by decreased of CSF cells, diminished nucleic acids and protein amounts, along with increased DNA-AO binding and accumulation of IL-l. These data are interpreted in terms of intensive apoptosis of CSF cells and increased acute phase of aseptic inflammation-like events induced by the procedure of liquor sorption.

[Effect of amphotericin B on the energy metabolism of tissue forms of Candida albicans].

Dehydrogenase activity of the tissue form cells of C. albicans during the infection process in albino mice with and without amphotericin B treatment was studied. The strength of the metabolic reactions resulting in accumulation of ATP was evident from the activity of 4 main enzymes, i.e. succinate dehydrogenase, lactate dehydrogenase, alcohol dehydrogenase and glucose-6-phosphate dehydrogenase. The enzymatic activity was determined by the tetrasol method based on formation of diphormazan. Investigation of the fungal cells 10 minutes after the infection showed that preliminary intravenous or intraperitoneal administration of amphotericin B did not change the activity of the tissue forms. The cytochemical characteristics of the fungal cells remained the same as that in the untreated animals. Six hours after infection of the animals treated with amphotericin B administered intravenously the fungus vegetation decreased from 52 to 38 per cent, while in the animals treated with amphotericin B administered intraperitoneally it was suppressed completely. Simultaneously the energy metabolism was also suppressed, the activity of alcohol dehydrogenase being suppressed most significantly. The activity of this enzyme in the cells of C. albicans isolated from the animals treated with the antibiotic administered intraperitoneally was 14 times lower than that in the cells of the culture isolated from the control animals.

Amphotericin B↗

[Effect of amphotericin B on the enzyme system of Candida albicans].

The mechanism of amphotericin B action was studied with the aid of cytochemical methods providing determination of the activity of the 4 main enzymes characterizing the cell energetics, i. e. succinate dehydrogenase, lactate dehydrogenase, alcohol dehydrogenase and glucose-6-phosphate dehydrogenase inside the cell. With an increase in the concentration of amphotericin B in the medium the activity of all the 4 enzymes decreased, the percentage of the inactive cells being regularly increased. Changes in the fermentative activity of C. albicans as dependent on the incubation time with the antibiotic were studied. Only the respiration activity decreased in 2 hours. As a result of a 4-hour exposure to the polyen in the cells of C. albicans the activity of the lactic acid fermentation, respiration through succinate dehydrogenase and activity of the pentose shunt decreased 1.5--2 times. In 24 hours of incubation the activity of the above decreased 80--90 per cent as compared to the activity of the initial culture.

[Neuronal activity in the prefrontal cortex in rats with various typological features during emotional affect].

Unit activity in the right and left prefrontal cortex was recorded in male Wistar rats after testing by the emotional resonance technique. Rats were divided in two groups by their reaction to the suffering cry of a partner. Rats from the group A ("altruists") escaped partner's crying, and those from the group E ("egoists") did not. Activity of neurons was analyzed in hungry rats, after feeding, during intracranial emotionally positive and negative stimulation, and during crying of the rat partner. Some differences in neural activity between A and E groups were revealed. In the hungry state the rate of neuronal discharges was higher in the A group. In both groups of animals the positive emotional stimulation was accompanied by more intensive neuronal reaction that the negative stimulation, but in the E group increase in the rate of neuronal discharges in both hemispheres was significantly more pronounced. Negative stimulation produced in both groups a significantly greater activation in the left hemisphere than in the right one while during the positive stimulation the neural activity was more intensive in the left hemisphere. The neuronal reaction to partner's crying was significantly higher in the A group in both hemispheres, while the neuronal activity in E group did not significantly change.

[Direct current self stimulation in rats].

In experiments on 16 male albino rats a phenomenon of self-stimulation of the hypothalamic structures has been produced by a DC anode and cathode, with 20 to 100 microamperes intensity. DC self-stimulation consists in regular succession of approaching the pedal, staying on the pedal and leaving it, i. e. a succession of states of motivational drive, reinforcement and avoidance. In a number of cases the conditioned reflex of self-stimulation is not extinguished for two to three days after switching off the current.

[The effect of different activating influences on the self stimulation reaction].

A study was made of the influence on self-stimulation of non-painful sensory stimuli of different modalities, and of intra-brain stimulations of emotionally positive and neutral points with the wiew to elucidate the specificity of certain functional relations appearing during interaction between emotionally negative and positive conditions. The data obtained attest that the influence of various excitation sites on self-stimulation reactions depends not so much on the strength of the stimuli, as on the specific neurophysiological organization of emotionally negative zones in the brain. A reciprocal enhancement of excitation of self-stimulation zones points to a certain non-specificity of positively reinforced structures.

[Interhemispheric relations of prefrontal cortical neurons in rats during emotional stimulation of increasing intensity].

Unit activity of the prefrontal cortex of the right and left brain hemispheres of rats was recorded during intracranial stimulation of emotionally positive and negative brain structures. The neurons were divided according to their reaction to a change in food motivation: cells that decrease (M-neurons) and cells that increase their firing frequencies (R-neurons) after feeding. Three levels of stimulation current intensity were used. When stimuli of subthreshold intensity (evoking the behavioral reaction of smelling) were applied, the recorded neuronal activity was higher in the left hemisphere. During threshold emotionally positive or negative stimulation (producing approach behavior or freezing, respectively), activity of M-neurons was higher in the right hemisphere, whereas the left-side R-neurons were more active than the right-side ones. During strong emotionally positive stimulation producing self-stimulation, the firing frequency of both groups of neurons was higher in the left hemisphere. Strong emotionally negative stimulation that evoked behavioral avoidance to a greater extent activated the right hemisphere.

[Interhemispheric asymmetry in the functional organization of the prefrontal cortex in animals of different typologic groups].

Rats allocated to groups by the method of "emotional resonance": rats which did and did not escape crying of a partner (A- and E-groups, respectively). Unit activity in the right and left prefrontal brain cortex (PFC) was recorded in these rats. The recorded neurons neurons were divided in two groups according to their reaction to a change in the level of food motivation. The so-called D-neurons decreased their activity after feeding of animals after a 24-hour food deprivation and the other group (I-neurons) increased its firing rate rate in this situation. It was shown that hemispheric distributions of D- and I-neurons are different in selected rat groups. In E-rats the I-neurons substantially predominated in the left hemisphere, whereas the D-neurons were more frequently recorded in the right one. No such asymmetry was observed in A-group of rats. During intracranial stimulation of emotionally positive brain structures I-neurons increased their firing rate, predominantly, in the left hemisphere, whereas during intracranial emotionally negative stimulation activation of the D-neurons predominated at the right. Features of the observed functional interhemispheric asymmetry of prefrontal cortex in A- and E-groups of rats were explained by differences in the interaction between hemispheres and dissimilar activation control.

[Effect of dalargin on the processes of memory and learning in the rat].

In experiments on rats, the influence was studied of dalargin on the elaboration and preservation of various homogeneous and heterogeneous conditioned reflexes (CRs) elaborated in single and multiple pairings. The effect of dalargin on the processes of learning and memory was compared with the action of the peptide on the activity of hypothalamic neurones. Administration of dalargin delayed the elaboration of maze defensive CRs and practically did not affect the elaboration of two-way avoidance. The preservation of CR also deteriorated under the influence of dalargin. Administration of dalargin 10 min before the CRs testing did not prevent their reproduction. When using CRs elaborated in a single pairing, dalargin disturbed the preservation of the drinking CR and improved that of passive avoidance CR. Dalargin in this dose affected the emotional state of animals in the open field and did not significantly affect their motor activity. Dalargin suppressed impulse activity in 17 out of 22 tested neurones of the lateral hypothalamus, with maximum effect in 20-50 min after its administration. The obtained data show that the character of dalargin action on the elaboration of CR and mainly on its consolidation, depends on the character of the elaborated CR and is probably due to great extent to the effect of the peptide on the brain emotional mechanisms.

[Neuronal activity in self-stimulation zones of the hypothalamus in motivational and emotional states induced by electrical and natural stimuli].

Study of neuronal activity in hypothalamus areas of self-stimulation in rats showed two types of neurones. Some of them had an increased frequency at food and water deprivation and they had maximum activation at current stimulation, eliciting motivational behaviour. Others had maximum activation at current level, eliciting self-stimulation effects, some of these neurones had positive activating relation to satisfaction of food requirements. The obtained data are considered as morphofunctional substrate of motivational and emotional states.
