Adsorption of human salivary mucin MG1 onto glow-discharge plasma treated acrylic resin surfaces.

It has been suggested that altering the surface properties of acrylic resin material may change the nature of the adsorbed pellicle affecting denture retention and microbial adherence. This study aimed at evaluating the adsorption of salivary high molecular-weight mucins, a major component of denture pellicle, onto modified acrylic resin surfaces. (Poly) methylmethacrylate specimens were treated by glow discharge plasma technique, using hydrophilic 2-Hydroxyethylmethacrylate monomer or oxygen (O(2)) gas and hydrophobic Hexamethyldisiloxane monomer, at different discharge powers. Acrylic samples were incubated with high-molecular weight mucin, MG1 purified from saliva, the adsorbed fractions were transferred to nitrocellulose membranes by slot-blot technique, stained by periodic acid-Schiff and colour intensities were analysed by a colour densitometer. Higher amounts of mucins were adsorbed on all the surfaces modified by glow-discharge plasma treatment. Within the limitations of this study, it was concluded that glow-discharge plasma altered the surfaces of acrylic resin denture base materials and significantly increased the adsorption of high molecular-weight mucins at varying levels depending on plasma parameters.

Adherence of Candida albicans to glow-discharge modified acrylic denture base polymers.

An important aetiologic factor in the pathogenesis of denture-induced stomatitis, is the presence of numerous yeasts, usually Candida albicans, on the fitting surfaces of dentures. In the present study, effect of glow-discharge plasma, a technique applied to increase surface wettability of acrylic resins, on candidial adherence was evaluated. The durability of glow-discharge modification with saliva coating was also evaluated. Samples including control and experimental groups were prepared by using heat compression mould technique. To create a hydrophobicity gradient, experimental groups were exposed to a radiofrequency glow discharge in an O2 atmosphere under different discharge powers. To characterize the wetting properties, an expression of surface hydrophobicity, contact angle measurements were performed by the sessile drop method. The organism used was C. albicans (ATTC10321). Acrylic samples were coated with unstimulated whole saliva collected from a healthy man. The fungal suspension was poured on saliva-inoculated samples and incubated at 37 degrees for 2 h. The samples were then fixed with glutaraldehyde and Gram stained. Adhered candidial cells were examined by light microscope. Diffuse Reflectance FTIR (DRIFT) and scanning electron-microscope examinations were also performed to evaluate the surface composition and roughness of the test groups. Glow-discharge plasma was found to be an effective means of increasing surface wettability even with salivary pellicle. Amounts of candida cells adhered were significantly higher in all the plasma treated surfaces than the unmodified control group (P < 0.001). It was concluded that improving the surface wettability of acrylic resins by glow-discharge plasma in O2 atmosphere increased the adherence of the C. albicans.

The effect of sustained and local administration of epidermal growth factor on improving bilateral testicular tissue after torsion.

Epidermal growth factor (EGF) modulates Leydig cell proliferation, steroidogenesis, spermiogenesis, and Sertoli cell activity. It plays an important role in repairing ischemia-reperfusion injury in different tissues. The aim of this study was to evaluate the effects of sustained and local administration of EGF on improving bilateral testicular tissue after torsion. A total of 57 Wistar albino rats were used. For the EGF transport system, 1x2 cm gelatin films containing 2 microg EGF were used. Torsion was created by rotating the right testis 720 degrees in a clockwise direction for 4 h in all groups except the control group. Then, in the torsion group, bilateral orchiectomy was performed. After returning the torsioned ipsilateral testes to their normal state, the bilateral testes were wrapped by 1x2 cm unloaded gelatin films in the gelatin (G7 and G21) groups and, by 2 microg EGF loaded gelatin films in the EGF 7 and EGF 21 groups. The testes were removed on the seventh and 21st days, respectively, for biochemical and histological examination. Histologically, Johnsen's spermatogenesis criteria and mean seminiferous tubule diameter (MSTD) measurements were used. The EGF7 group did not show significant loss of Sertoli cells, while in the G7 group the number of these cells decreased. The ipsilateral ischemic testis of the EGF21 group showed Leydig cell hyperplasia, and the contralateral non-ischemic testes in this group were similar to the control group. In the G21 group, the bilateral testes showed Sertoli cell only syndrome in some sections, and most of the cells were undergoing apoptosis. The mean spermatogenesis scores and MSTD in the EGF7 and EGF21 groups were higher than in the G7 and G21 groups ( P<0.05). Malondialdehyde levels were significantly lower in the EGF groups than in the G groups ( P<0.05). Glutathione peroxidase (GSH-Px) levels in the G21 group were significantly higher than in the EGF21 group. Our study shows that local and sustained EGF release after testicular torsion improves bilateral testicular injury. EGF administration may be a new treatment choice for bilaterally injured testis after detorsion without removing the twisted testis.

Cytotoxicity of 5-fluorouracil entrapped in gelatin microspheres.

Gelatin microspheres were prepared by water/oil emulsion polymerization and by cross-linking with glutaraldehyde. For the microsphere preparation procedure, two different gelatin (5 or 10% w/v) and three different glutaraldehyde (5, 0.5 or 0.1% v/v) concentrations were used. The influence of preparation compositions on microsphere recovery, particle size and morphology, swelling and degradation, 5-fluorouracil loading and release, and cytotoxicity were investigated. The concentrations of gelatin and glutaraldehyde influenced the size and surface properties of microspheres. The decrease in gelatin concentration and the increase in glutaraldehyde concentration resulted in the formation of smaller (140.82-71.47 microm for gelatin microspheres with a 5% gelatin content; 297.67-97.44 microm for gelatin microspheres with a 10% gelatin content) microspheres with smoother surface properties. Swelling values were decreased as the amount of glutaraldehyde was increased. In particular, for microspheres with a high glutaraldehyde content (5% v/v), only about 15% were degraded in 12 days, whereas for those with 0.5% (v/v) glutaraldehyde, almost 97% degradation occurred in the same period. The most rapid 5-fluorouracil release was observed from uncross-linked microspheres (about 88% in 4 h), whereas a particular slower release (about 36% in 4 h) profile was obtained for the highly cross-linked ones. Cytotoxicity tests of free and entrapped 5-fluorouracil were carried out with MCF-7 breast cancer cell line. Free 5-fluorouracil produced an immediate effect, whereas entrapped 5-fluorouracil showed a prolonged cytotoxic effect.

Macroporous poly(3-hydroxybutyrate-co-3-hydroxyvalerate) matrices for bone tissue engineering.

Macroporous poly(3-hydroxybutyric acid-co-3-hydroxyvaleric acid) (PHBV) matrices were prepared after solvent evaporation and solute leaching. PHBV solutions with different concentrations were prepared in chloroform: dichloromethane (1:2, v/v). In order to create a matrix with high porosity and uniform pore sizes, sieved sucrose crystals (75-300 or 300-500 microm) were used. PHBV foams were treated with rf-oxygen plasma to modify their surface chemistry and hydrophilicity with the aim of increasing the reattachment of osteoblasts. Surface characteristics, pore sizes and their distribution on PHBV surface were studied by scanning electron microscopy (SEM) and Scion Image Analysis Program. Void volume, pore sizes and density of foams were found to be significantly affected by foam preparation conditions. Stability of PHBV foams in aqueous media was studied. Their weight and density were unchanged for a period of 120 days and then a significant decrease was observed for the rest of the study (60 days). Osteoblasts were seeded onto the foams and their proliferation inside the matrices was also determined by SEM. After 29 and 60 days of incubation, growth of osteoblasts on matrices was observed.

EGF containing gelatin-based wound dressings.

In case of bulk loss of tissue or non-healing wounds such as burns, trauma, diabetic, decubitus and venous stasis ulcers, a proper wound dressing is needed to cover the wound area, protect the damaged tissue, and if possible to activate the cell proliferation and stimulate the healing process. In this study, synthesis of a novel polymeric bilayer wound dressing containing epidermal growth-factor (EGF) -loaded microspheres was aimed. For this purpose, a natural, nontoxic and biocompatible material, gelatin, was chosen as the underlying layer and various porous matrices in sponge form were prepared from gelatin by freeze-drying technique. As the external layer, elastomeric polyurethane membranes were used. Two different doses of EGF was added into the prepared gelatin sponges (1 and 15 microg/cm2) to activate cell proliferation. EGF addition was carried out either in free form or within microspheres to achieve prolonged release of EGF for higher efficiency. The prepared systems were tested in in vivo experiments on full-thickness skin defects created on rabbits. At certain intervals, wound areas were measured and tissues from wound areas were biopsied and processed for histological examinations. The wound areas decreased upon low-dose EGF application but the difference between the affects of free EGF and microsphere loaded EGF was not so distinct. Upon increasing the dose of EGF by a factor of 15, it was observed that controlled release of EGF from microspheres provided a higher degree of reduction in the wound areas. Histological investigations showed that the prepared dressings were biocompatible and did not cause any mononuclear cell infiltration or foreign body reaction. The structure of the newly formed dermis was almost the same as that of the normal skin.

Bond strength and failure analysis of lining materials to denture resin.

OBJECTIVES: This study was conducted to investigate the bonding properties of five lining materials to a denture base resin. Two hard (chemical-cured resin: Kooliner "Coe Labs, USA" and light-cured VLC resin: Triad "Dentsply, USA") and three soft (chemical-cured resin: Express "Alcos,-USA", heat-temperature vulcanized 'HTV' silicone material: Molloplast-B "Regneri GmbH, Germany" and room-temperature vulcanized 'RTV' silicone material:Ufi Gel-P "Voco, Germany") liners were used. METHODS: Paladent 20 "Heraeus Kulzer GmbH, Germany", a conventional heat-cured polymethylmethacrylate (PMMA), was used as the denture base resin and as the control material. Bonding strength and adhesion properties of the liners to PMMA were compared by tensile test and scanning electron microscope (SEM) analysis. After curing, an aging process was applied and the samples were immersed and stored in distilled water at 37 +/- 1 degrees C and taken out at certain aging intervals (at 0, 15, 30 and 90 days) for examination. Specimens (168) were processed for tensile tests and other specimens (24) for fracture tests. The mean and standard deviation values were calculated. Changes in the mechanical properties and the SEM findings of the adhered surfaces were evaluated. RESULTS: Triad (a hard liner) has the closest tensile strength to the control, indicating the strongest bonding between the base and the liner. Also, during the aging process, formation of better adhesion was observed in SEM micrographs. From the SEM analysis it was found that, Molloplast-B (a soft liner) also has a very good filling capacity. SIGNIFICANCE: Among the hard lining materials, VLC resin to chemical-cured resin; and among the soft lining materials, HTV resin to RTV and chemical-cured ones should be preferred for relining procedures. Molloplast-B and Express as resilient liners were found to have adequate adhesive values for clinical use.

Stability of alpha-amylase immobilized on poly(methyl methacrylate-acrylic acid) microspheres.

Poly(methyl methacrylate-acrylic acid) microspheres were prepared and the acid groups were activated by using either carbodiimide (CDI) or thionyl chloride (SOCl2). alpha-Amylase was covalently bound on these activated microspheres. The properties of the immobilized enzyme were investigated and compared with those of the free enzyme. The relative activities were found to be 80.4 and 67.5% for carbodiimide and thionyl chloride bound enzymes, respectively. Maximum activities were obtained at lower pHs and higher temperatures upon immobilization compared to free enzyme. No change in Vmax and approximately 12-fold increase in K(m) were observed for immobilized enzymes. The enzyme activities, after storage for 1 month, were found to be 24.5 and 52.5% of the initial activity values for CDI and SOCl2 activated matrices, respectively. On the other hand the free enzyme lost its activity completely in 20 days. Immobilization, storage stability and repeated use capability experiments carried out in the presence of Ca2+ ions demonstrated higher stability, such as SOCl2 immobilized enzyme retained 83.7% and CDI immobilized enzyme retained 90.3% of the original activity of the enzyme. The immobilized enzymes that were used 20 times in 3 days in repeated batch experiments demonstrated that, in the absence of Ca2+ ions about 75% and in the presence of Ca2+ ions greater than 90% of the original enzyme activity was retained.

Estimation of monomer content in polymethyl methacrylate contact lens materials by Raman spectroscopy.

Polymethyl methacrylate is the most commonly used contact lens material due to its excellent optical properties. However the presence of residual monomer in the structure alters its transparency as well as its biocompatibility, thus, there is a need to detect any remaining methyl methacrylate. Raman spectroscopy is a rapid, sensitive, and non destructive method as compared to other spectroscopic, chromatographic or polarographic methods. In this study, the spectra of some lens materials (prepared from methyl methacrylate, 2-hydroxy ethyl methacrylate, Hexamethyl disiloxane and polypropylene glycol with or without the addition of crosslinker ethylene glycol dimethylacrylate) were obtained by Raman spectroscopy. It was observed that an amount of crosslinker present in the structure proves effective for the unpolymerized monomer content. None of the samples, except those containing nexamethyl disiloxane, demonstrated any monomer residue.

Covalent immobilization of Aspergillus niger on pHEMA membrane: application to continuous flow reactors.

Poly(2-hydroxyethyl methacrylate) (pHEMA) membrane was prepared via photopolymerization and activated with epichlorohydrin. The conidia of Aspergillus niger strains (wild type 'NRRL-3' and genetically improved strain 'NRRL-3/2-2A') were covalently-immobilized on the membranes. Uniform growth of A. niger cells on membrane surfaces was verified by SEM. The glucose oxidase (GOD) activity of the immobilized cells was determined in a continuous flow membrane reactor (CFMR) by assaying for hydrogen peroxide produced. The activity was also determined in the culture fluids of A. niger strains, freely grown in batch cultures. The CFMR was run with 0.1 mol dm-3 glucose with a fixed flow rate of 20 cm3 h-1 for 60 h during which a 10% loss of the original activity was detected. The loss of the activity with the freely cultivated mycelia was about 50% after 30 h. The GOD activity of the improved strain NRRL-3/2-2A was about 20 times higher whether in immobilized or in free form. The GOD activity of the immobilized A. niger strains in the continuous flow membrane reactor was found to be 2.5 times better than their counterparts freely grown in batch cultures indicating that immobilization increases the activity and the stability of the microorganisms.

A comparative study of the performance of solid supported and soluble urease for the enzymatic hydrolysis of urea.

The performance of both free and solid supported urease for the enzymatic hydrolysis of urea was studied. Kinetic analysis of reaction rate data shows that the kinetic data were consistent with the proposed model. Statistical tests validated the model. Despite the deactivation of ureases, the immobilized enzyme could be the choice for a technological process aiming at the detoxification of blood in haemoperfusion columns.

Studies on contact lens materials.

The development of plastics with the optical properties of glass led promptly to their use as contact lenses and intra-ocular lenses to rectify certain visual defects. Research to improve these polymeric materials is continuous but there is not much in the literature since most of the findings are patented. In this work, polymethyl methacrylate, the most commonly used lens material was chosen as the base material and its co and terpolymers were prepared using 2-hydroxyethyl methacrylate, N-vinyl-2-pyrrolidinone, hexamethyl disiloxane, and polypropylene glycol. The transparency, refractive index, contact angle, density, equilibrium water content, and percent hydration properties were examined. Theoretical values were calculated for linear expansion and oxygen permeation from the density and hydration values.

Polymer coating for hemoperfusion over activated charcoal.

Plasma polymerization (glow discharge) was used to make a polymeric coat with hexamethyldisiloxane on the surface of activated charcoal granules. These samples were examined with ESCA and SEM for surface chemical analysis, coat thickness, and uniformity, and for changes in surface topography. The effects of the coat on absorptive capacity and release of fine particles were examined.

Wetting properties of saliva substitutes on acrylic resin.

The good wetting of the acrylic resin by saliva substitutes is of clinical importance in xerostomic patients. This study evaluated the wetting properties of different artificial saliva formulations that were mucin-based, carboxymethylcellulose-based, and concentrated ion-based on poly(methyl methacrylate) denture base resin, and compared these properties with natural saliva. The wetting properties of the test materials were examined by contact angle measurements. Ninety-six samples that measured 30 x 30 x 3 mm were examined. The wetting properties of mucin-containing and carboxymethylcellulose-containing substitutes on poly(methyl methacrylate) were significantly better than those of human saliva. Mucin-containing artificial salivas had the best wetting properties on the acrylic resin for the materials tested.
