Tentoxin sensitivity of chloroplasts determined by codon 83 of beta subunit of proton-ATPase.

Tentoxin is a naturally occurring phytotoxic peptide that causes seedling chlorosis and arrests growth in sensitive plants and algae. In vitro, it inhibits activity of the beta subunit of the plastid proton-adenosine triphosphatase (ATPase) from sensitive species. Plastid atpB genes from six closely related, tentoxin-sensitive or -resistant Nicotiana species differ at codon 83, according to their response to the toxin: glutamate correlated with resistance and aspartate correlated with sensitivity. The genetic relevance of this site was confirmed in Chlamydomonas reinhardtii by chloroplast transformation. The alga, normally tentoxin-resistant, was rendered tentoxin-sensitive by mutagenesis of its plastid atpB gene at codon 83. Codon 83 may represent a critical site on the beta subunit that does not compete with nucleotide binding or other catalytic activities.

Expression of active human interferon-beta in transgenic plants.

Tobacco plants were transformed with the human gene for interferon-beta (IFN-beta). Transformation was determined by the polymerase chain reaction (PCR), and expression was determined by Western blot analysis, by purifying the IFN from the transgenic plants, and by bioassays indicating its activity in human cells. Plants expressing IFN-beta were self-pollinated. IFN-beta-expressing progeny plants were selected and produced active IFN-beta, indicating stable transformation.

Interferon-gamma potentiates antibody affinity in mice with a genetically controlled defect in affinity maturation.

Interferon-gamma (IFN-gamma), the tetrapeptide tuftsin and the synthetic nonapeptide from interleukin-1 beta (IL-1 beta) (amino acids 163-171) have previously been shown to act on macrophages and/or T cells and to enhance antibody titres to T cell-dependent antigens. The ability of these immunomodulatory agents to potentiate antibody affinity in addition to antibody titre has been studied in a line of mice that fail to demonstrate normal maturation of antibody affinity (low N/M mice). The results presented here confirm that each of the agents potentiate antibody levels following simultaneous injection with a T cell-dependent antigen but demonstrate that only IFN-gamma is able to enhance antibody affinity in these mice. The observation that IFN-gamma can enhance both antibody affinity and antibody levels suggests that it could be an important adjuvant for vaccine use.

Vocational disability and rehabilitation in multiple sclerosis.

Multiple sclerosis represents a significant cause of vocational disability among young and middle age adults. As a group, M.S. patients are frequently well educated, highly trained and possess intact families but have had to leave their former occupations. While many unemployed M.S. patients are willing and able to work, they are not now receiving the vocational rehabilitation services they need to reenter the work force. Physicians frequently see M.S. patients, but they do not often refer those patients for vocational rehabilitation services. Unemployment associated with multiple sclerosis is a significant social complication of the disease. While the physician cannot cure or halt the progress of the disease, he or she can assist the M.S. patient in obtaining the specialized help necessary to re-enter the job market.

The status of automation and data processing in the United States blood banking community.

A survey of the extent of automation and data processing in the blood banking community was conducted by questionnaire. The likelihood that a facility had some type of automation was related to the volume of blood products produced or transfused. Facilitates that collected blood or collected and transfused blood were more likely to have automation for ABO and Rh testing than those that only transfused blood. Automation for many other blood bank tests is unavailable. Many facilities had data processing capability, which is used primarily for accounting.
