[Effect of amino acids on cell proliferation and apoptosis in organotypic tissue culture of young and old rats].

There was studied in brain cortex, spleen and liver organotypic tissue culture the effect of effective concentrations (0.05 ng/ml) of 20 amino acids on the development of proliferative activity and apoptosis in peripheral fields of explants of rats of various age: 1 month and 18 months old. In the young tissues, the most frequent activity (3 times) was found while using the histidine, asparagine acide, valine, threonine, proline, leucine, tyrosine, methionine, and the less frequent (1 time)--while using lysine, glutamine, cysteine. Almost half number of the amino acids (11) was active in the old tissue cultures, as compared to the young tissues (19). The summary effectiveness of the amino acids was greater in young tissues (42), as compared to the old tissues (11). The data obtained suggest, that the bioregulator peptides should be necessary used to correct the less sensitivity of the tissues to the amino acids in old age.

[The tissue-specific effect of synthetic peptides-biologic regulators in organotypic tissues culture in young and old rats].

The synthetic peptides--cardiogen, bronchogen, prostamax and pancragen were used to investigate their effect on organotypic tissue culture of explants from heart, lungs, prostatic gland and pancreas of young (3-week old) and aged (18-month old) rats. The peptide effective concentration of 0.05 ng/ml showed a stimulating effect in appropriate tissue cultures as compared to the control explants in all groups of rats. So these bioregulatory peptides can be used in clinic practice for the stimulation of reparative processes in the appropriate tissues while ageing.

[Modulating effect of amino acids on cerebella of rats of various age in organotypic tissue culture].

There was studied in cerebella organotypic tissue culture the effect of effective concentrations (0.05 ng/ml) of 20 amino acids on the development of of proliferative activity in peripheral fields of explants of rats of various age: 3 days and 18 months old. The amino acids glycine, asparagine, lysine, arginine, proline, glutamine acid, cysteine, valine and isoleucine acid are active, inducing a more intensive growth zone in explants in 3-day old rats, as compared to control. The inhibitory effect was not observed. In the cerebella explant of old rats there was observed the inhibitory effect of 7 amino acids due to the apoptosis. This effect might be take in account by the synthesis of bioregulator peptides, contained these amino acids.

[The modulating action of amino acids upon development in an organotypic culture].

The effect of L-amino acids in concentration 0.05 ng/ml was investigated in organotypic tissue culture of spleen, liver and brain cortex at the age of 1 and 21 days. The high molecular and low hydrophobic amino acids: asparagin, lysine, arginine, and glutamine, are active inducing both a less intensive growth zone in explants of all three tissue types in 1-day old rats and more intensive growth zone in spleen, liver explants in 21-day rats as compared to control. Another group of low molecular and high hydrophobic amino acids is active inducing a more intensive growth zone in brain cortex explant in 21-day rats, as compared to control. The data obtained suggest a modulating role of amino acids in the tissues at different stages of maturation.

[The study of peptide effect in the mechanisms of angiogenesis and forming of microenvironment of the tumors in old rats].

The experiments were performed in rats with transplanted sarkoma M-1. The injections of synthetic peptide vilon at the doses 0.5 mcg sigificantly increased the apoptosis of tumor cells in the experiment as compared to the control group. So vilon has possessed the oncomodulating action on the transplanted carcinoma. The injections of epigene lead to the inhibition of the sarcoma growth due to the development of the hemorragic necrosis and apoptosis increased. The results obtained suggested that vilon and epigene are the perspective preparates in the cancer therapy.

The neurite-stimulating influence of components of medicinal leech salivary gland secretions in organotypic culture of spinal ganglia.

The effects of components from medicinal leech (Hirudo medicinalis) salivary gland secretions and the therapeutic agent Piyavit on the growth of chick embryo neurites in organotypic culture were studied. Native destabilase and bdellin A at concentrations of 0.01, 0.02, 0.05, and 0.1 ng/ml, bdellin B at a concentration of 0.05 ng/ml, and eglin at a concentration of 0.1 ng/ml had neurite-stimulating activity, evident on the third day of organotypic culture of spinal ganglia. The stimulatory activity of destabilase was lost after revere-phase chromatography. The neurite-stimulating activity of the extract of the therapeutic agent Piyavit (200 ng/ml) in organotypic ganglion culture appeared to result from the neurite-stimulating salivary gland components within this agent, suggesting that Piyavit could be used for the treatment of neurodegenerative disorders.

The neurite-stimulating activity of components of the salivary gland secretion of the medicinal leech in cultures of sensory neurons.

The effects of components of the salivary gland secretion (proteases and protease inhibitors) of the medicinal leech (Hirudo medicinalis) on the growth of neurites of sensory neurons from chick embryos (10-11 days old) were studied in organotypic cultures. Destabilase and high-molecular-weight bdellin B, (0.01, 0.02, 0.05, and 0.1 ng/ml), bdellastasin (0.02 and 0.05 ng/ml), and eglin C (0.1 ng/ml) had neurite-stimulating effects on day 3 of cultivation of spinal ganglia. Identification of the neurite-stimulating activity of these components of medicinal leech salivary gland secretions creates the basis for creating new therapeutic agents for the treatment of neurodegenerative diseases.

[Reciprocal relation of proliferative activity in central and peripheral zones of splenic organ culture exposed to vilon in rats of various ages].

There was studied in spleen organotypic tissue culture the effect of vilon on the development of of proliferative activity in central and peripheral fields of explants of rats of various age: 3 days, 3 weeks and 2 years old. Vilon stimulated cell proliferation both in young and old rats, in spite of all there are reciprocal relations of this process in central and peripheral fields in the spleen organotypic culture of rats of various age.

[The modulating role of essential and non-essential amino acids in organotypic tissue culture in rats of different ages].

The effect of amino acids L-lysin, L-asparagin, L-arginin, L-glutamate was investigated in organotypic tissue culture of spleen, liver and brain cortex of rats at different age. The amino acids in concentration 0.05 ng/ml are active inducing a less intensive growth zone, as compared to control, in 1-day and in older rats--an intensive growth zone, as compared to control in 21-day rats. The data obtained suggest a modulating role of amino acids in the tissues at different stages of maturation.

[The effect of ouabain on sensory neuron neurite growth in organotypic culture].

Ouabain, a known inhibitor of Na+, K(+)-ATP, taken in a wide range of concentrations, was investigated in organotypic tissue culture of dorsal root ganglia cells of 10-11 day old chick embryos. Ouabain inhibited neurite growth in a dose-dependent manner. The Hill coefficient was defined as 1, and the Kp value was estimated as 1 x 10(-10) M. At inhibitor concentrations exceeding 1 x 10(-9) M, the growth of neurites was totally inhibited. It is assumed that Na+, K(+)-ATPase may play an important role in regulation of the process of neurite growth in sensory neurones.

[Combined effect of vilon and cyclophosphane on tumor transplants and lymphoid tissue explants in mice and rats of various age].

The experiments were performed in mice with transplanted Lewis lung carcinoma. The injections of synthetic peptide vilon at the doses 1 mg/kg significantly increased the survival of mice. So vilon has possessed the oncomodulating action on the transplanted carcinoma. The synchronous injection both of vilon and cyclophosphan at the doses 100 mg/kg decreases the survival of mice. There was also studied in spleen organotypic tissue culture the effect of vilon and cyclophosphan on the development of explants of rats of various age: 1 day and 2 years old. Vilon stimulated apoptosis both in young and old rats, but the inhibitory effect of cyclophosphan was abolished in the presence of vilon in culture media at the dose 5 ng/ml. The results obtained suggested that perspective preparates in the cancer therapy--vilon and cytostatic drugs-, must be used not synchronously.

[Functional morphology of organotypic culture of spleens from rats of various ages exposed to vilon].

A synthetic dipeptide vilon (Lys-Glu, 20 ng/ml) was added to a cultural medium of explants taken from rats of 3 days, 3 weeks and 2 years of age. The peptide induced morphologic stability of the tissue and activated the regeneration and functional activity of cells. The stronger effect on the explants taken from the old rats suggests that vilon is a candidate for geriatric research and practice.

[Tissue-specific action of peptides in tissue culture of rats of various ages].

There was studied the effect of polypeptide preparations isolated from the cerebral cortex (Cortexin), epiphysis (Epithalamin), liver (Hepalin), thymus (Thymalin) and small synthetic peptides--Cortagen, Epitalon, Livagen, Vilon on the development of explants from the cerebral cortex, brain subcortical structures, liver and thymus of rats of various age. With respect to the applied concentrations of the investigated peptides, they exerted tissue-specific effects in the organ-typical culture of animal tissues.

[Regulating role of various amino acids in development of apoptosis in organotypic culture of the nervous and lymphoid tissue].

The effect of aminoacids L-arginin, L-lysine, L-asparagin was investigated in organotypic tissue culture of brain cortex and spleen of 1-day old rats. The aminoacids in concentrations 0.05 and 0.1 ng/ml are active, inducing a less intensive growth zone as compared to the control, excluding the effect of asparagin on lymphoid tissue. Method of fluorescent staining shows a negative correlation between growth zone size and apoptotic cell number. The nerve and lymphoid tissue express apoptosis in response to aminoacids.

[Neurite-stimulating effect of Hirudo medicinalis salivary gland secreting factors in organotypic culture of the dorsal root ganglia].

Effects of destabilise, bdellin, bdellin A, eglin were investigated in organotypic tissue culture of dorsal root ganglia (DRG) of 10-11-day old chick embryos. Native destabilase and bdellin A, bdellin B and eglin are more active inducing a more intensive neurite growth in DRG as compared with the control. A neurite-stimulating effect of the drug "pyjavit" seems to be associated with destabilase, bdellins and eglin neurite-stimulating activity.

[Transplantation of embryonic myocardium in animal experiments].

The work presents the results of experimental transplantation of a not-divided embryonic myocardium culture into the mesentery of the adult rats' intestine for the terms of about 30 days. The influence of Cimitidine--cardiolen on the development of this culture was investigated. The grafts were shown to be taken and to function well and the use of cardiolen reliably improves the development of the culture.

[Neurite-stimulating activity of secretions from the Hirudo medicinalis salivary gland in cultured sensory neurons].

Components of leech (Hirudo medicinalis) salivary gland secretion (high molecular mass bdelin B, bdellastasin, eglin C, destabilize) were used to investigate their neurotrophic effect on organotypic culture of dorsal root ganglia of the 10-11-day old chicken embryos. Destabilize and high molecular mass bdelin B in concentrations of 0.01, 0.02, 0.05 and 0.1 ng/ml, bdellastasin in concentrations of 0.02 and 0.05 ng/ml, eglin C in concentrations of 0.1 ng/ml exhibit neurite-stimulating effect as compared to the control.
