Thermal denaturation of Na- and Li-DNA in salt-free solutions.

Thermal denaturation of Na- and Li-DNA from chicken erythrocytes was studied by means of scanning microcalorimetry in salt-free solutions at DNA concentrations (Cp) from 4.5 x 10(-2) to 1 x 10(-3) moles of nucleotides/liter (M). Linear dependencies of DNA melting temperature (Tm) vs lgCp were obtained: [formula: see text] for Na- and Li-DNA, respectively. Microcalorimetry data were compared with the results of spectrophotometric studies at 260 nm of DNA thermal denaturation in Me-DNA + MeCl solutions at Cp congruent to (6-8) x 10(-5) M and Cs = 0-40 mM (Me is Na or Li, Cs is salt concentration). It was found that Eqs. (1) and (2) are valid in DNA salt-free solutions over the Cp range 6 x 10(-5)-4.5 x 10(-2) M. Protonation of DNA bases due to the absorption of CO2 from air in Na-DNA + NaCl solutions affects DNA melting parameters at Cs < 4 mM. Linear dependence of Tm on lga+ is found in Na-DNA + NaCl at Cs > 0.4 mM in the absence of contact of solutions with CO2 from air (a+ is cation activity). A dependence of [dTm/dlga+] on Li+ activity was observed in Li-DNA + LiCl solutions at Cs < 10 mM: [dTm/dlga+] increases from 17 degrees-18 degrees at Cs > 10 mM to 28 degrees-30 degrees at Cs congruent to 0.2-0.4 mM. Spectrophotometric measurements at 282 nm show that this effect was caused by protonation of bases in fragments of denatured DNA in neutral solutions. The Poisson-Boltzmann (PB) equation was solved for salt-free DNA at the melting point. The linear dependence of Tm vs lgCp was interpreted in terms of Manning's condensation theory. PB and Manning's theories fit the experimental data if charge density parameter (xi) of denatured DNA is in the range 1.8-2.1 (assuming for native DNA xi = 4.2). Specificity of Li ions in interactions with DNA is discussed.

[Interaction of immobilized DNA with silver ions].

The interaction of Ag+ with DNA immobilized in polyacrylamide gel was studied by means of the ion-exchange method. Ag+ ions are shown to bind to DNA bases, their charges being neutralized by phosphate groups. The binding sites of Ag+ and H+ are likely to be the same, but the strength of Ag+ binding is greater than that of H+. Ag+ ions like H+ are shown to cause the formation of compact structures in immobilized DNA, the amount of these structures being dependent on subtle differences in DNA samples. DNA samples, not forming compact structures under the influence of H+, do not form them under the influence of Ag+. This fact can indicate the similarity of the mechanisms of the compact structures formation in both cases. The results obtained are compared with the data available for the interaction of Ag+ with DNA in solution. The mechanism of the Ag+-DNA interaction is discussed.

[Thermal denaturation of DNA in concentrated salt-free solutions: comparison of microcalorimetric and spectrophotometric data].

Scanning microcalorimetry and spectrophotometry were used to study the dependence of melting enthalpy (delta Hm) and temperature (Tm) on DNA concentration in salt free solutions and on NaCl concentration in solutions with constant DNA concentration. This data is used to calculate the Manning's charge density parameter which is found to be equal 1.8. The linear dependence of Tm on the logarithm of DNA concentration in salt free solution was obtained. An approximate evaluation of dissociation degree in native DNA at different concentrations was made by comparison of straight lines in the Tm = f(lg CNaCl) and Tm = f(lg Cp) coordinates.

[Interaction of Cu2+ and Ca2+ ions with DNA immobilized in polyacrylamide gel].

Interaction of cupric and calcium ions with DNA entrapped in acrylamide gel was studied by means of ion-exchange method. Ion-exchangers on the basis of DNA were obtained by polymerization of acrylamide with and without warming-up. Samples of DNA with different degrees of denaturation were also used for preparation of ion-exchangers. It was demonstrated that ion-exchange capacity is determined by the number of phosphate groups of DNA entrapped in gel. Cu2+ and Ca2+ ion-exchange equilibrium on the macromolecules of immobilized DNA was studied at different temperatures. Ion-exchange equilibrium constant alpha Cu2+ Ca2+ was shown to vary from 1,9 to 8,2 an increase of selectivity for Cu2+ ions taking place under conditions favouring denaturation of DNA. Stoichiometric ratio between the quantity of the phosphate groups and the number of equivalents of Cu2+ and Ca2+ was observed in all the experiments. Lack of over-equivalent absorption of CuCl2 on the DNA molecules indicates that chloride ions do not participate in charge neutralization within the Cu2+.DNA complex. It means that Cu2+ ions interact with phosphate groups of DNA coordinate simultaneously with DNA bases only.
