[The properties of the extracellular alkaline phosphatase of the causative agent of melioidosis].

After growing P. pseudomallei VPA on solid medium extracellular alkaline phosphatase with a molecular weight of 93,000 AMU was isolated, and practically purified from the extract of this medium by precipitation with ammonium sulfate, subsequent gel chromatography and concentration on membrane filters. The optimum conditions for enzymatic reaction were found to be pH 9.0 and a temperature of 50 degrees C. The enzyme was resistant to freezing and to heating at a temperature of up 60 degrees C for 30 minutes, as well as to the action of pH 3.0-10.5, but became completely inactivated after heating at 90 degrees C for 10 minutes and incubation at pH 2.0 for 20 hours.

[Use of ganglioside-containing magnetic polyacrylamide sorbents for the immunoenzyme method of determining cholera enterotoxin].

A variant of EIA techniques for the determination of cholera enterotoxin is proposed. This method is based on the selective sorption of the toxin on ganglioside-containing magnetic granules with its subsequent detection by means of immune serum and antispecific immunoperoxidase conjugate. The proposed method permits the detection of 0.052 +/- 0.02 ng of protein of the purified toxin.

[Production of an erythrocytic ganglioside diagnostic reagent for detecting cholera enterotoxin and toxoid in indirect hemagglutination tests].

The studies described in this work have indicated that cholera enterotoxin and its components (cholerogenoid and subunit B) can be detected in amounts of 0.25, 0.28 and 0.6 microgram of protein per 1 ml, respectively, by means of erythrocytes sensitized with gangliozide-containing complex. The conditions for erythrocyte sensitization have been established. The methods of cholerogen titration in the passive hemagglutination test by means of the erythrocytic gangliozide diagnostic reagent and in Craig's skin test have been shown to correlate. Similarly, the passive hemagglutination test is supposed to be suitable for detecting other bacterial toxins interacting with gangliozides.

Immunobiological characteristic of antigens from the parasitic phase of Coccidiodes immitis.

The possibility of obtaining serologically active antigens from tissue spherules extracted from the lungs of albino mice infected with C. immitis has been demonstrated. Unlike the method of obtaining spherules in vitro, the accumulation of sufficient quantities of tissue spherules does not involve special difficulties. To obtain more pure and specific antigens from crude tissue spherules material, chromatographic separation on a column with Sephadex G-100 is recommended. Antigens obtained from the spherules in vivo have been found to be more specific and active in serological and allergic reactions in comparison with the corresponding antigens from the mycelial form or from spherules in vitro and can be used in the diagnosis of coccidioidosis as well as in the study of the mechanism of cellular immunity.

[The enzyme-antienzyme interactions of a number of Burkholderia pseudomallei hydrolases with monoclonal immunoglobulins].

Phosphatase, phospholipase C and a proteolytic complex, including casein- and hemoglobin-hydrolases, have been isolated from cell-free extracts of B. pseudomallei cultivation medium. A set of monoclonal antibodies (McAb) to the antigenic complexes of this infective agent has been obtained. Conditions for the study of the interaction of the enzymes and McAb have been worked out, and most of the 14 immunoglobulins under study have been shown to neutralize 2-3 hydrolases each. Three types of McAb, selectively inhibiting only the proteolytic complex, only casein-hydrolase and only phospholipase C, have been identified.

[Some properties of plasmocoagulase of the causative agents of glanders and melioidosis].

Criteria for the evaluation of the plasmocoagulase activity of natural isolates and mutant strains of the causative agents of glanders and melioidosis were worked out, which made it possible to subdivide them by this sign into pathogens with high, moderate and low activity. Plasmocoagulase produced by pathogenic Burkholderia was shown to be a thermolabile enzyme, comparatively stable with respect to the action of such chemico-biological agents as hydrogen peroxide and chloramine.

[Thermostable Pseudomonas pseudomallei hemolysin].

The chemical nature of thermostable P.pseudomallei hemolysin has been determined. This substance is glycolipid, synthesized by bacteria at the stationary phase of growth under the conditions of the prevalence of the source of carbon over the source of nitrogen in the nutrient medium. The hemolytic activity of thermostable hemolysin has a delayed character and is more pronounced in P.pseudomallei R-forms.
