Teratological study of hydroxypropylmethylcellulose acetate succinate in rabbits.

A teratological study was carried out in New Zealand White rabbits in order to examine the teratogenic potentiality of HPMCAS, a useful pharmaceutical excipient. HPMCAS was orally administered at dose levels of 625, 1,250 and 2,500 mg/kg/day for a period of 13 days from day 6 to day 18 of gestation. All pregnant females were sacrificed on day 29 of gestation and their fetuses were examined. The administration of HPMCAS during a period of organogenesis produced no embryotoxic and teratogenic effects as well as no influence on behavior, appearance and growth of animals.

Effects on offspring induced by oral administration of hydroxypropylmethylcellulose acetate succinate to the female rats in peri- and post-natal periods.

A perinatal and postnatal study was carried out in Slc: SD rats orally administered Hydroxypropylmethylcellulose acetate succinate (HPMCAS), a useful pharmaceutical excipient, at dose levels of 625, 1,250 and 2,500 mg/kg/day for a period from day 17 of gestation to day 21 after delivery. All pregnant rats were allowed to litter naturally, and the postnatal development of the offsprings was observed. In the administered group of 2500 mg/kg, the liver weight was significantly increased in males and showed a tendency to increase in females as compared with control. No significant differences between the control group and the administered groups were found in postnatal growth and differentiation, behavior and reproductive ability of male and female offsprings.

Production, purification and characterization of the plasminogen activator in teratocarcinoma stem cells induced with sodium butyrate.

Suspension cultures of pluripotent teratocarcinoma cells were induced with sodium butyrate to produce plasminogen activator (PA), generally regarded as a marker enzyme of differentiation of the teratocarcinoma. The induction of plasminogen activator was very efficient, resulting in production of sufficient enzyme to allow its purification. The activator was inactivated by rabbit anti-human melanoma plasminogen activator antiserum, indicating that it was a tissue-type activator (t-PA). The enzyme was purified by column chromatograph on phosphocellulose, zinc-chelate agarose, Con-A Sepharose and Sephadex G-150. The preparation at the final step of purification gave a single peak of enzyme activity at pH 7.3 +/- 0.1 on isoelectric focusing, and showed a molecular weight of approximately 77,000 on SDS PAGE.

An aetiological study on spondylolysis from a biomechanical aspect.

The authors report clinical studies on lumbar disorders (clinical symptoms, X-ray findings) in athletes in various sports. The sport items were divided into three groups according to the main dynamic load applied to the lumbar region. As a result, over 60% of the athletes suffered from "lumbago", and among them spondylolysis reached the high rate of 27%. Arising from these clinical observations, we performed biomechanical laboratory analyses on human cadaver material, axial compression and rotational bending. Our results suggest that the incidence of spondylolysis depends upon the extent and direction of the loads.

Isolation and characterization of deletion mutants affected in early genes of bacteriophage phi 80.

When Escherichia coli cells that had been irradiated with ultraviolet light were infected with bacteriophage phi 80, five major (pE, pB, pA, pC and pD) and two minor (pU and pV) proteins were found to be synthesized during early stages of infection. The genes coding for the five major proteins were mapped on the phi 80 chromosome using various deletion mutants which lacked the capacity to synthesize some or all the major proteins. The size and positions of all the deletions were determined by gel electrophoresis of EcoRi digests of phage DNA and by electron microscopy of heteroduplexes between DNAs of the deletion and wild-type phage. The five major proteins designated pE(25K), pB(40K), pA(45K), pC(34K) and pD(31K) were shown to be encoded in this order presumably by a single operon that was located at 60.2--67.4% on the phi 80 genome. These proteins were found to be involved in phage recombination. The absence of pE or pB resulted in a Red- phenotype and the absence of three proteins (pE, pB and pA) resulted in a Fec- phenotype. The exact positions of the genes for the minor proteins pU(29K) and pV(26K) have not been determined.

A study of lectin histochemistry in ameloblastoma.

The lectin histochemistry of ameloblastoma was investigated in order to understand the nature of the carbohydrate residues in this tumor, in accordance with the WHO subclassification. Mannose and glucose were generally common in almost all cells in both the follicular and plexiform types, especially stellate cells and cystic lesions of the follicular type. N- acetylglucosamine (GlcNac), N-acetylneuraminic acid (NANA) and galactose were much more abundant in stellate cells of the plexiform type than in those of the follicular type. Thus, there is a slight difference in lectin histochemistry of the stellate cells between the follicular and plexiform types of ameloblastoma.

Appearance of fracture caused by emphysema.

A case presenting radiographic appearance of a fracture line associated with emphysema which occurred in the removal of a mandibular third molar is reported. The cause of the fracture line in the radiograph is discussed together with the benefit of CT in diagnosis.
