[A new approach to setting hygienic pesticide standards].

Studies are performed on tobacco-pickers, which are in contact with dimethoate and tomato-pickers, sprayed with methylparathion. The content of pesticides in washes from leaves and hands, as well as inhibitors and cholinesterase activity in serum and erythrocytes is measured. A correlation is found between the organophosphate pesticides content on vegetable material and hand wash and presence of inhibitors in blood. Proposal is made for defining the quantity of pesticides and its metabolites in hand wash when no effect is observed to be determined by coefficients, characterizing the quality of hydrolytic inhibitors in erythrocytes and serum and the rate of cholinesterase activity decrease. The control for observing the minimum reentry time for to be carried out by measuring the quantity of pesticides in hand wash.

Cholinesterase Inhibitors↗

[Dynamics of cholinesterase inhibition in methylparathion poisoning].

The inhibition of cholinesterase (ChEA) and acetylcholinesterase activity (AcChEA) in blood and brain of albino rats was followed up after repeated exposure to methylparathion (MP). An abrupt reduction of ChEA and AcChEA was observed by 30 th and 90 th min after the single peroral administration of 32 mg/kg MP. Furthermore, the reduction was most manifested with ChEA, where the activity was reduced with 71 per cent, AcChEA was reduced to 54 per cent. By 24 th h after the administration of the dose ChEA was significantly higher than the value in the control animals. That could be explained by a quick reactivation of enzyme activity. No clinical picture of intoxication was observed during that period. AcChEA in blood and brain remained strongly reduced, both by 24 th hour after the single MP quantity administered and during the following observation periods--till 14 th day. After the discontinuation of MP administration, a slow tendency to increase of AsChEA values was observed, being strongly decreased as compared the level of the enzyme in the control animals. The reduction of ChEA and AcChEA is associated with the appearance of non-identified metabolites of MP-inhibitors of ChEA with Rf 0. I and 0.65 during the first periods--to 90 min. On 14 th day, after 13-days administration, metabolites with Rf 0.15 and 0.4 were confirmed in chromatogram, besides the main spot with Rf--0.26 (corresponding to MP). Due to the considerable ChEA reduction, prior to the manifestation of the clinical picture of intoxication, ChEA determination in blood could be an early index for confirming the exposure to organophosphoric compound.

[Cutaneous resorption of actellic (O,2-diethylamino-6-methyl pyrimidin-4-yl-O,O-dimethyl phosphorothioate) in the presence of high temperature and humidity].

Studies were carried out on the combined effect of high relative humidity and temperature on stability and dynamics of skin actelic resorption. Actelic resorption with skin application was established to be uneven. The initial resorption of the substance by the skin, was followed by its increased concentration on skin surface, and a further decrease and maintenance with a general tendency to decrease of the concentration. Humidity accelerated resorption, whereas with the combination of high temperature and high relative humidity--an reverse tendency was observed.

[35S-cysteine incorporation into protein synthesis of the liver and testes in subacute oral poisoning with zineb, maneb and mancozeb].

35S-cysteine incorporation in the protein synthesis of liver and testes is studied in groups of 20 white rats each, following 14--day poisoning with dithiocarbamates, at doses 1/5 LD50, as follows: group one, cineb - o,180 g/kg, maneb - 0,140 g/kg, and mankozeb - 0,260 g/kg. Protein incorporation in liver and testes is studied after decapitation of the animals, within two hours of intraperitoneal 35S-cysteine injection at dose 15 micro C per rat. Liver and testes undergo appropriate treatment after the method of Munro et al./10/, as modified by Sheitanov /9/, and counting is done with VAZ--310. The results point to a varying degree of cysteine incorporation increase in the protein synthesis of liver and testes. It is attributed to a variety of dystrophic and necrobiotic changes in the tissues of either of the organs, taking place under the effect of dithiocarbamate preparations. An enhancement of regenerative processes in the above mentioned organs, associated with intensified protein synthesis, is by no means ruled out. Differences established in the intensity of amino acid incorporation correlate with the degree of protein synthesis disorders, mostly in terms of the effect of maneb preparations.

[Method for obtaining bile from rats in a chronic experiment].

The authors make a critical review of the methods, described in the literature for multiple samples of bile from rats under the conditions of a chronic experiment. They present their own method. The results from the follow-up of the dynamics in the bile cholinesterase activity in one of the rats canulated by this method are described.

[Metabolism and the dynamics of the urinary excretion of Sayfos in rabbits].

The splitting of Sayphos into metabolites inhibiting indoxyl acetate esterase activity is investigated. Also the dynamics of Sayphos excretion in the urine is studied. It is established that in the urine of unpoisoned rabbits, normally a compound with Rf 0.79 is contained, inhibiting indoxyl acetate esterase activity. Sayphos and metabolite I with Rf 0.08 are demonstrable both in the blood and urine at 2 hours after peroral introduction of the poison. Within 24 hours no traces of the pesticide and metabolite I are discovered in the blood, and in the urine--within 96 hours since the beginning of the experiment. Most of the indoxyl acetate esterase activity inhibitors (Sayphos and metabolite I), excreted via urine (mean 95%), are emitted up to the 24th hour inclusive.

[Determination of the minimal times for safe work following the Sayfos spraying of tobacco].

The semidisintegration period of Sayphos over tobacco leaves is determined--2.5 days at angular coefficient of the disintegration straight line K=0.12 1 ppm/ 1 mg/kg concentration of Sayphos over tobacco leaves is considered as harmless and accordingly safe for work. A table for minimal terms of safe work computation is also suggested. Because of the slow Sayphos decomposition in soil, the preparation should be employed but once--after the planting of tobacco.
