Effect of papaya seed extract on contractile response of cauda epididymal tubules.

AIM: To evaluate the administration of Carica papaya seed extract on the contractility of cauda epididymal tubules in male rats. METHODS: Adult male albino rats were administered intramuscularly papaya seed extract at a dose of 0.5 mg/kg/day for 7 days. Animals were killed, cauda epididymal tubules of 5 cm length were isolated and the contractile response to different concentrations of adrenalin (1-500 microg/25mL) was examined. In another group of animals, the contractile response was assayed 3 months after withdrawal of the treatment. RESULTS: Papaya seed extract brought about a significant decrease in the contractile response of epididymal tubules as compared with the control. After three months of papaya withdrawal, a nearly normal pattern of contraction was regained. CONCLUSION: Papaya seed treatment reversibly reduces the contractile response of cauda epididymal tubules.

Role of ascorbic acid on mercuric chloride-induced genotoxicity in human blood cultures.

Efforts are made to find therapeutic agents capable of minimizing genotoxicity of various natural and man-made compounds. The genotoxicity induced by mercury compounds remains controversial. Therefore we have investigated the genotoxic effect of mercuric chloride (MC; HgCl(2)) at three concentrations (1.052, 5.262 and 10.524 microM) and role of L-ascorbic acid (vitamin C) at a concentration of 9.734 microM on MC-treated short-term human leucocyte cultures. We assessed the proliferative rate index (PRI), sister chromatid exchange (SCE) and chromosomal aberrations (CAS) in control and MC-treated cultures with and without vitamin C supplementation. The results showed that MC has no effect on cell-cycle kinetics, but the frequency of SCE/cell was significantly higher in a dose-dependent manner than control values. HgCl(2) also significantly induced C-anaphases (abnormal mitosis) in blood cultures. These effects were prevented by the addition of vitamin C to MC-treated cultures. The data indicate the mutagenic activity of MC and the protective role of vitamin C on mercury-induced genotoxicity in human blood cultures is probably due to its strong antioxidant and nucleophilic nature.

Effect of papaya seed extract on microenvironment of cauda epididymis.

AIM: To evaluate the effect of aqueous Carica papaya seed extract on microenvironment of cauda epididymis. METHODS: Adult male albino rats were intramuscularly administered with 0 (control) or 0.5 mg papaya seed extract/kg body weight for 7 days. Cauda epididymal tubular content was collected by micropuncture technique; epididymal luminal fluid and sperm pellets were separately analyzed. RESULTS: The results revealed that the extract treatment caused significant reduction, as compared with control, in total protein and sialic acid contents in both epididymal fluid and sperm pellet. As compared with control, significantly lowered acid phosphatase activity was recorded in sperm pellet but was higher in epididymal fluid after the treatment. The extract treatment also caused significant reduction in level of inorganic phosphorus in the epididymal fluid. CONCLUSION: It is concluded that the aqueous papaya seed extract alters cauda epididymal microenvironment.

Three siblings with Harlequin Ichthyosis in an Indian family.

The Harlequin fetus is a distinct genetic entity with a strikingly grotesque appearance. Three siblings (two males, one female) with Harlequin Ichthyosis, a rare form of congenital disorder, born to the same parents are presented. The genetic aspects are discussed and the relevant literature is reviewed.

Incomplete gonadal dysgenesis.

A 21-year-old phenotypic male with ambiguous external genitalia and hypospadias was referred for cytogenetic studies. Exploratory laparotomy revealed presence of a small atropic uterus, unilateral gonadal dysgenesis with fallopian tubes on both sides and a cryptorchid testis on the left side. No gonad could be traced on the right side. Chromosomal analysis from peripheral whole blood culture revealed a 46,XY cell line. No mosaicism was detected. Endocrine studies showed elevated levels of serum FSH and LH with low borderline testosterone level and failure to respond to HCG stimulation. The presence of fallopian tube next to testis suggests absence of anti-Mullerian hormone secretion by Sertoli cells. The absence of Wolffian duct derivatives indicates insufficient secretion of testosterone by Leydig cells.

Polydactyly: a study of a five generation Indian family.

Preaxial polydactyly was observed in up to five generations of an Indian family living in a village in the Rajkot district (Gujarat). Among the 71 affected members, 45 were males and 26 were females. All these affected members showed preaxial polydactyly manifesting as a well formed, articulated extra digit of the hand or foot. Twenty other cases were also identified with polydactyly involving triphalangeal digits replacing the thumbs or duplication of the big toe(s). To the best of our knowledge, the present family is the largest in which several members have preaxial polydactyly of different types. No other abnormalities were apparent. The present study strongly suggests that preaxial polydactyly with a well formed extra digit, triphalangeal thumbs, and duplication of the big toe can be manifestations of the same autosomal dominant gene. It is likely that other factors are modifying the expression of this gene.

Translocation t(22;22)(p11.1;q11.1) and NOR studies in a female with a history of repeated fetal loss.

A 32-year-old phenotypic female with a history of nine consecutive abortions each in the first trimester was referred for cytogenetic studies. She was found to have 45,XX,t(22;22) (p11.1;q11.1) chromosomal pattern. The Ag-NOR banding technique showed that the NORs of both the acrocentrics involved in the translocation were deleted and the loss suffered from the elimination was compensated by the increased NOR activity as well as presence of dNOR on other acrocentric chromosomes.

Unilateral gonadal dysgenesis with both testis and fallopian tube on the same side in a 45,X/46,X inv (Y) mosaic male.

A 5-year-old male with ambiguous external genitalia, hypospadias and microphallus without an urethral orifice was referred for cytogenetic studies. Exploratory laparotomy revealed presence of an infantile uterus and unilateral gonadal dysgenesis with both testes and fallopian tube on the right side. The metaphase cells from peripheral blood culture showed both 45,X/46,X inverted Y (p11.2q11.23) cell-lines (98:2). The inverted Y was found to be of paternal origin. Maternal chromosomal pattern was normal 46,XX. The presence of a fallopian tube next to testis suggest absence of secretion of anti-Mullerian hormone by Sertoli cells. The absence of Wolffian duct derivatives suggest insufficient secretion of testosterone by Leydig cells.

Microdose vasal injection of sodium fluoride in the rat.

A single microdose (50 micrograms/50 microL) injection of sodium fluoride (NaF) into the vasa deferentia of adult male albino rats (Rattus norvegicus) caused arrest of spermatogenesis and absence of spermatozoa in the lumina of the seminiferous tubules of the testes, which consequently led to a decline in the sperm count in the caudae epididymides. Scanning electron microscopy of cauda and vas deferens sperm revealed deflagellation and tail abnormalities. This is probably related to the alterations in the internal milieu of these organs which rendered the spermatozoa immotile and consequently caused fertility impairment in the experimental animals. Thus microdoses of sodium fluoride were found to affect reproductive function and fertility rate.

A triple-X female with long arm deletion of one of the X-chromosomes associated with primary amenorrhoea: 47,XX, +del(X) (q27.3).

A 25-year-old phenotypic female with primary amenorrhoea was referred for chromosomal analysis. Earlier she had undergone hormonal therapy but showed no response. The secondary sex characters were of female type, with poor breast development. Laparoscopic findings revealed the presence of a very small uterus; the right ovary was found to be undeveloped and the left was absent. Cytogenetic study revealed a case of triple-X with deletion of the terminal region of the long arm of one of the X chromosomes [Xq27.3]. Among the 100 buccal mucosa cells analysed, 30 cells showed double Barr bodies. Hormonal studies using RIA technique revealed normal levels of prolactin (9.1 ng/ml), a high level of FSH and LH (135 and 61 mIU/ml) and low levels of estradiol and progesterone (12 pg/ml and 0.20 ng/ml respectively). To our knowledge, this may be the first report of a triple X with deletion of the X chromosome associated with primary amenorrhoea.

Effects of fluoride on the histoarchitecture of reproductive organs of the male mouse.

The effects of sodium fluoride (NaF) ingestion in two doses (10 and 20 mg/kg body weight) for 30 days on histology and histocytometry of reproductive organs of the adult male mouse were investigated. In order to study reversibility, treatment was withdrawn for one and two months. The testes, epididymides, vas deferens, prostate, and seminal vesicle were utilized for the study by standard hematoxylin-eosin staining and an ocular eye piece and micrometer scale. NaF treatment caused severe disorganization and denudation of germinal epithelial cells of seminiferous tubules with absence of sperm in the lumina. The Leydig cell and nucleus diameters were not affected. The caput epididymis showed fewer changes than the cauda. However, epithelial cell nuclear pyknosis and absence of luminal sperm were observed. A reduction in epithelial cell height, nuclear pyknosis, denudation of cells, and absence of sperm occurred in the cauda epididymis. The vas deferens epithelium showed nuclear pyknosis, clumped stereocilia, and cell debris but no sperm in the lumen and an increase in the lamina propria. The prostate and seminal vesicles were not affected by treatment. Withdrawal of treatment caused marked recovery in the histoarchitecture of these organs. The effects of NaF treatment are therefore transient and reversible.

Effects of acetylsalicylic acid on reproductive organs of adolescent male rats.

The effects of acetylsalicylic acid (ASA) at a dose of 800 micrograms/day per rat for 7 days on some androgenic parameters such as organ weights, succinate dehydrogenase, acid phosphatase, fructose, cholesterol and protein of testis, epididymis, vas deferens and accesory glands in adolescent male rats were investigated. The semen characteristics and scanning electron microscopy (SEM) study on sperm morphology of cauda epididymis were also carried out. The results revealed that the treatment manifested a marked effect in altering the metabolism of testis, cauda epididymis, seminal vesicle and vas deferens. The androgen antagonistic and antianabolic effects were by and large transient and reversible by ascorbic acid administration.

Effect of estradiol benzoate on rat testis and adrenal.

The effects of in vivo administration of estradiol benzoate at a dose of 50 micrograms/rat/day for 15 days on testosterone production of testis and adrenal in vitro were investigated in rats. The data revealed that the androgen synthesis of both the steroidogenic organs were reduced by the treatment. The decrease in androgen production by testis was related to its reduced sperm count and weight. On the contrary, accumulation of cholesterol occurred in treated rat testis indicating inhibition of its steroidogenesis. This data confirm our earlier reports on the direct effect of estrogen on testis.

Effect of lead on human semen.

Semen qualities were studied in workers with an average age of 30 years and occupationally exposed to lead in a printing press. Another sample with the same average age but not exposed to lead were taken as control subjects. The average lead content in blood and seminal plasma of the exposed group were 42.5 mcg/100 ml and 14.80 mcg/100 ml, respectively. Their sperm counts and percentage of motile sperm were significantly affected. Significantly higher percentages of abnormal spermatozoa were also observed in these semen samples. The levels of seminal acid phosphatase, succinic dehydrogenase, and fructose in them were also significantly found to be low compared with those from the unexposed subjects. Cytochemical study of sperm head DNA in the exposed groups showed a low surface reaction.

Structural changes in reproductive organs of male rats after estradiol benzoate treatment.

The effects of estradiol benzoate (E2B) at a dose of 50 micrograms/rat/day for 7, 15 and 24 days on histoarchitecture, histocytometry and relative weights of reproductive organs in mature albino rats were investigated. The results revealed that the organ weights, histology of the testis, epididymis, vas deferens and seminal vesicle of treated rats were considerably affected as a consequence of the treatment. The height of germinal/secretory epithelium and tubular diameter of these organs also declined, whereas the peritubular muscle layer thickness of epididymis and accessory gland was increased. In vas deferens, in contrast, the muscle layer was reduced. The induced effects of estrogen were related to the duration of the treatment and support our earlier data that estrogens have a direct effect on the testis.
