[T-lymphocyte induced normalization of transformed malignant fibroblasts: lymphokine with normalization factor activity].

Normalizing influence of different lymphocyte populations and their soluble factors on L-929 transformed malignant fibroblasts (L cells) has been examined. It was demonstrated that splenic T-lymphocytes caused stable (heritable) normalization of receptor apparatus, biophysical and proliferative characteristics of L cells. Lymphokine with the activity of normalization factor (NF) was purified from 24-hour immunocyte conditioned medium. Stability of the normalization phenomenon was caused by NF induced synthesis of functionally analogous factor in L cells. The results obtained indicate the existence of non-cytotoxic mechanisms of tumour growth immunological control. The isolated lymphokine possessed also the activity of early embryonal cell differentiation factor. It is suggested that lymphokines with the activity of NF are physiological regulators of nonlymphoid cell differentiation.

[Effect of lymphocytes on the differentiation of in vitro transformed L-line mouse fibroblasts].

Possible participation of immune system cells in the differentiation of non-lymphoid tissues has been examined. Lymphocytes were tested on transformed fibroblasts of L mouse strain. The presence of lymphocytes in L cell culture enhanced their differentiation, assessed by morphological, proliferative and biophysical criteria. The induction of L-cell differentiation took place during contact and short-distant interaction both with mouse and human lymphocytes. The phenomenon had a stable character, as it was revealed in L cells separated from lymphocytes during passaging. The results obtained indicate real and potential abilities of the immune system to affect differentiation of proliferating non-lymphoid tissues.

[Effect of a factor (factors) secreted by human amnion cells on the proliferative activity of normal and transformed fibroblasts].

The effect of FL-medium obtained upon cultivation of the human amniotic cells (FL) on the proliferative activity of intact (from spleen of C3H mice) and transformed (L-cells) fibroblasts was studied. The effect depended on the intensity of cell division: in case of high mitotic activity of fibroblasts in the control cultures FL-medium inhibited their proliferation and in case of low activity stimulated their division. It is suggested that the human amniotic cells secrete a factor (s) regulating the mitotic activity of cells in the culture medium.

[Effect of fetoplacental factors on in vitro proliferation of lymphocytes and non-lymphoid cells].

The effect of extracts of placenta, amniotic fluid, serum of pregnant women and newborn infants on PHA-stimulated proliferative activity of lymphocytes and proliferation of non-lymphoid cells (L-cells) was studied. The inhibiting effect decreased in the series: placenta -- amniotic fluid -- serum of pregnant women. The effect on L-cells was similar to that on lymphocytes. It is suggested that the biological samples under study contained factors having non-specific immunosuppressive effect.

[Analysis of the cellular interaction of 2 transplantable lines in mixed culture].

A relationship between two transformed cell lines (L and HEp-2) has been studied at different population densities. L cells were inhibited at a low, but not at a high density. We noted specific cell structures in mixed culture at a low density. Crowded cells did not form such structures at the same conditions. Possible alterations of L cells are discussed.

[Placental proteins as regulators of immunological reactions in pregnancy].

The effect of two specific placental proteins, trophoblastic beta 1-glycoprotein (TBG) and chorionic alpha 1-microglobulin (CAG), on the immunological reactions was studied in vitro. TBG in physiological doses suppressed the proliferation of lymphocytes induced by plant mitogens and allogenic cells in the unidirectional mixed cultures, strengthened the effect of concanavalin A upon the induction of cells-suppressors in the culture and, in low concentrations, decreased the percentage of E- and EAC-rosette-forming cells. In none of the tests used CAG was effective. But when studying the effect of TBG and CAG mixture on PHA-induced proliferation of lymphocytes the inhibiting effect of TBG was weakened and, in some cases, completely relieved.

[Comparison of the action of the sera of pregnant women and of trophoblastic beta 1-glycoprotein in a number of immunological tests].

The effects of blood sera of pregnant women in the I, II and III trimesters (SPWI, SPWII, SPWIII) were compared with those of purified preparations of trophoblastic beta 1-glycoproteid (TBG) on phytohemagglutinin (PHA) induced proliferative response of lymphocytes of the intact donors and on the production of a factor inhibiting the migration of leucocytes (FIM-L). SPWI and purified TBG preparations in doses corresponding to its level in SPWI were shown to produce similar effects. With the increase of TBG concentration, however, the inhibiting effect of its preparations on the proliferation of lymphocytes increased much more intensively than that of SPWII and SPWIII containing the same dose of this protein. The stimulating effect of TBG on the production of FIM-L increased with the dose as well while SPWII and SPWIII did not exert any effect on the migration of leucocytes. A suggestion is put forward that SPWII and SPWIII contain factors modifying the effect of TBG.

[A model for the assessment of fibroblast morphology].

A model for comprehensive assessment of morphologic parameters of a cell (area, nucleus and cytoplasm area, degree of spreading), based on the use of a scanning device, has been developed. The model comprises an original method for staining the preparations, which permits assessment of the degree of cell spreading by microdensitometry, and updating the equipment and software. The programs help assess the integral and derivative parameters, such as the area of the interface zone of a cell, the shape factor, spreading, and nucleus to cytoplasm ratio. Using this model, the effect of peripheral blood mononuclears on the morphology of transformed murine fibroblasts L929 upon a contact exposure was assessed. Addition of mononuclears to fibroblast culture led to alteration of their morphology: increase of the mean cellular area, degree of spreading, and of shape factor and reduction of the nucleus/cytoplasm ratio. This was true for both the mean values and the population composition.

[Reactions between protease inhibitors in blood plasma and collagen].

The interaction of three macroglobulins and three serpines, as well as inter-alpha-trypsin inhibitor (ITI) with alpha 1- and alpha 2-chains of collagen I which were immobilized on nitrocellulose. All seven proteinase inhibitors were shown to have a certain affinity for collagen. The binding of alpha 2-macroglobulin and gestation-associated protein A with collagen chains is largely determined by the conformational state of these macrophages. alpha 2-Antiplasmin, proteinase alpha 1-inhibitor, antithrombin III and ITI with collagen yield complexes that are resistant to urea, sodium dodecylsulfate, and Trilon B.

[Chronic bacterial prostatitis as a manifestation of secondary immunodeficiency state].

The clinico-immunological examination of 57 patients with chronic bacterial prostatitis was carried out. The clinical analysis made it possible to divide the patients into 3 groups characterized by the presence of chronic inflammatory diseases of other organs which had appeared before (group 1) or after (group 2) the manifestation of the symptoms of prostatitis, aw well as by the absence of concomitant inflammatory diseases (group 3). At the same time these patients were found to have changes in their immune status, most pronounced in patients of groups 1 and 2. The clinico-immunological analysis of the patients with chronic bacterial prostatitis revealed the fact that chronic bacterial prostatitis was a chronic inflammatory process linked with changes in the immune system; these changes had the signs of secondary immunodeficiency and required immunocorrective therapy.
