Presence of a distinct 24-hour melatonin rhythm in the ventricular cerebrospinal fluid of the goat.

Melatonin profiles in the cerebrospinal fluid (CSF) of conscious goats were examined under long-day (16L:8D) and short-day (8L:16D) environments. CSF melatonin, collected from the lateral ventricle, showed distinct 24 h rhythms with high concentrations being restricted to the dark phase, which averaged 1,320.6 pg/ml under 16L:8D and 660.6 pg/ml under 8L:16D. On the contrary, the nocturnal rise in CSF melatonin was totally absent in the pineal sympathetically denervated animals. Night interruption by 1 h lighting (about 400 lux at the height of goat's head) resulted in an abrupt decline of CSF melatonin to the basal level within 30 min and immediate recovery to the previous high level after reestablishment of the dark phase. The CSF/plasma ratio was 10.8-18.4 during the dark phase and 1.7-1.8 during the light phase. CSF and plasma melatonin levels were also examined after exogenous melatonin given either peripherally or intraventricularly. Continuous subcutaneous infusion of melatonin (5 micrograms/h) maintained melatonin levels in the plasma about 3 times higher than that in the CSF during its application. On the other hand, an intraventricular injection of 2 micrograms melatonin elevated plasma melatonin by 100 pg/ml within 1 min. These results indicate that turnover of CSF melatonin is fairly rapid and favor a hypothesis for direct access of pineal melatonin to the brain ventricular system in the goat.

Study on abnormalities in the appearance of finger and palm prints in children with cleft lip, alveolus, and palate.

The aetiology of abnormalities in the appearance of finger and palm prints in children with cleft lip, alveolus, and palate was examined by comparing the frequencies of abnormalities in hereditary cases and sporadic cases, and those in hereditary cases with those in their parents. The following results were obtained: Comparison of hereditary cases and sporadic cases. Abnormalities in the appearance of finger and palm prints were greater in hereditary cases than in sporadic cases. Abnormalities in the appearance of finger and palm prints were greater in parents of hereditary cases than in parents of sporadic cases. The rate of agreement in the appearance of finger and palm prints between parents and children was greater in hereditary cases than in sporadic cases. Comparison within hereditary cases. Abnormalities in the appearance of finger and palm prints were greater in affected children when one of the parents was affected (gamma = 1/2) than when both parents were normal (gamma = less than 1/2). Abnormalities in the appearance of finger and palm prints were greater in parents, one of whom was affected, than in normal parents. The rate of agreement in the appearance of finger and palm prints of parents, one of whom was affected, and their gamma = 1/2 children cases was higher than of normal parents and their gamma = less than 1/2 children. It was concluded that the above results could be explained by multifactorial inheritance.

[Clinical results of cefaclor treatment of infections of oral regions].

Cefaclor (CCL) is a cephem antibiotic for oral use. Thirty cases of infection of oral regions, in the First Department of Oral and Maxillofacial Surgery, School of Dentistry, the University of Tokushima, were treated with CCL at 750 mg/day. The results were as follows: The efficacy rate for all kinds of diseases was 80.0% in 3 days and 96.7% in 5 days and 86.7% as an overall assessment by the patients' doctors. With regard to the degree of symptoms, the efficacy rate for cases with moderate symptoms was less than that for mild cases in 3 days and according to the assessment by the patients' doctors. Some moderate cases required increased medication because no beneficial effect was observed. In sensitivity tests, CCL was shown to be effective on all 27 isolated clones of bacteria tested. CCL caused no detectable changes in laboratory data. No side effects were observed in any case. From these results, it is concluded that CCL is a highly effective antibiotic for use on infections of the oral regions.

Rec assay and mutagenicity studies on metal compounds.

We carried out rec assays on 127 metal compounds with Bacillus subtilis to check their DNA-damaging capacity and mutagenicity. Certain compounds of beryllium, cobalt, cesium, iridium, osmium, platinum, rhodium, antimony, tellurium, thallium and vanadium were newly found to be positive in addition to those of known positive metals such as arsenic, cadmium, chromium, mercury, molybdenum and selenium. Reverse mutation assays with Escherichia coli and Salmonella strains showed that compounds of rhodium (RhCl3), tellurium (Na2H4TeO6, Na2TeO3) and platinum (PtCl4, (NH4)2PtCl6) are potent mutagens.

Stimulation of in vivo calcification using collagen membranes cultured with osteoblastic cells in vitro: a preliminary report.

Guided tissue regeneration (GTR) is a useful modality in the management of periodontal disease and for bone augmentation around osseointegrated implants. This study evaluated the in vivo use of atelocollagen membrane (AC) on which osteoblastic cells (OBCs) were cultured in vitro, for application as a GTR membranous material. Osteoblastic cells isolated in our laboratory from mouse calvaria formed a thin film on the AC in vitro which was easily manipulated after 21 days in culture. The AC and OBCs complex material (ACOB) was subjected to freezing and thawing and implanted in mouse subcutaneous tissue for the study of histologic events surrounding the implanted ACOB. Histologic findings in the subcutaneous tissue showed calcification on the ACOB at 28 days postimplantation, while no such finding was evident at the control site, where only AC without OBCs were grafted. The present study suggests the possibility of membrane calcification for GTR through ACOB produced by OBCs on an AC in vitro.
