Identification of 5-HT3 recognition sites in the ferret area postrema.

The 5-HT3 receptor antagonist [3H]zacopride was used to identify 5-HT3 recognition sites in the ferret area postrema. Specific binding was determined by the inclusion of the 5-HT3 receptor antagonist BRL 43694 in the incubation media, and was shown to be much higher in the area postrema than in other brain regions. The increased binding in the area postrema may reflect either a greater number of binding sites, a higher affinity for such sites, or both. The results indicate that 5-HT3 recognition sites are present within the area postrema and may afford an antiemetic site of action for zacopride and other 5-HT3 receptor antagonists.

Ketotifen can antagonise changes in sensitivity of cerebral dopamine receptors: behavioural correlates in rodent and primate.

Rats preselected as "low responders" to the hyperactivity-inducing action of (-)N-n-propylnorapomorphine [(-)NPA] responded to an infusion of dopamine into the nucleus accumbens (25 micrograms/24 hr for 13 days) with hyperactivity during the infusion and long-term increased sensitivity to (-)NPA administered after the infusion. Both effects were antagonised by ketotifen (intraperitoneal infusion, 1 mg/kg/day), administered during the period of infusion of dopamine. Animals subject to infusion of dopamine also showed enhanced hyperactivity to L-DOPA (plus benserazide): this enhanced response was also antagonised by ketotifen, given acutely as a single dose (1 mg/kg i.p.). When haloperidol was given concurrently with the infusion of dopamine, spontaneous locomotor activity was markedly increased after the infusion for a period of at least 7 weeks: this long-term change was antagonised by ketotifen (1 mg/kg/24 hr), given during the period of treatment with dopamine/haloperidol or by a single acute injection of ketotifen (0.1-1.0 mg/kg i.p.) on an established response. Ketotifen, in doses up to 20 mg/kg (i.p.) did not reduce spontaneous locomotor activity, cause catalepsy or antagonise amphetamine-induced stereotypy in normal rats. In the marmoset, an enhanced sensitivity to the locomotor-stimulant effects of L-DOPA was induced 5 to 8 weeks after a treatment for 4 days with the neurotoxin 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP), which led to the development of akinesia and severe depletion of dopamine in the striatum. Treatment with a single dose of ketotifen (1 mg/kg i.p.) 60 min before L-DOPA antagonised the enhanced locomotor responsiveness to the L-DOPA/benserazide regimen.(ABSTRACT TRUNCATED AT 250 WORDS)

An HPLC assay procedure of sensitivity and stability for measurement of acetylcholine and choline in neuronal tissue.

A method is presented for the sensitive and specific determination of acetylcholine and choline in neuronal tissue. The method is based on the separation of acetylcholine and choline by reversed-phase HPLC, passing the eluent into a post-column reactor containing choline oxidase and acetylcholinesterase covalently bound to vinyl sulphone bonded onto a hydroxyethyl methacrylate support, and electrochemical detection of the hydrogen peroxide formed. The limit of detection of the procedure is 1 pmol for acetylcholine and 500 fmol for choline. The excellent baseline stability of the method ensures the rapid and reliable processing of a large number of samples.

Home peritoneal dialysis. A major advance in promoting home dialysis.

The institution of a home peritoneal dialysis program has allowed us to increase the number of patients with end-stage renal failure entering our home dialysis program from 42.5% to 67.5% of the total population. This represents a 56.5% increase over the rate achieved by home hemodialysis alone. Twenty-four percent of the home peritoneal dialysis patients could have managed home hemodialysis, but 38 could not and this represents 48% of the total patients who entered our home dialysis program, for the period of the study. These patients would have required institutional dialysis which would not have been practical for 52.6% of them because of the distance they live from Toronto. The results of home peritoneal dialysis have compared favorably with home hemodialysis in the 2 concurrent but unmatched series in respect of training time, failure rate, need for in-hospital back-up and patient survival. A long-term study of matched patients randomized to either treatment group such as that described by Blumenkrantz will finally answer the question as to how valid is our contention that peritoneal dialysis compares favourably to hemodialysis for the treatment of end-stage renal failure.

Cognitive enhancing actions of DuP 753 detected in a mouse habituation paradigm.

The potential of DuP 753, an angiotensin II receptor antagonist, to improve cognitive performance was assessed in a mouse habituation paradigm. On repeated daily testing DuP 753 (10.0 ng kg-1 p.o.) enhanced the basal cognitive performance of mice (decrease in latency to move from the light to the dark compartment and increases in the rears, line crossings and percentage of time spent in the dark compartment) and overcame the cognitive impairment induced by the administration of scopolamine (0.25 mg kg-1 i.p.). These findings further implicate the modulation of mental function by angiotensin II.

Normal densities of 5-HT3 receptor recognition sites in Alzheimer's disease.

The present studies investigate the possible difference in 5-HT3 receptor recognition site density in amygdaloid and hippocampal homogenates from patients with Alzheimer's disease compared with age-matched controls. The 5-HT3 receptor recognition site radioligand [3H]-(S)-zacopride identified a comparable density of specific sites (defined by granisetron 10.0 microM) in tissues obtained from patients with Alzheimer's disease as compared to age-matched controls (amygdala, Bmax 41 +/- 4 and 43 +/- 9; hippocampus, Bmax 59 +/- 9 and 51 +/- 9 fmol mg-3 protein; tissue from Alzheimer's disease patients and age-matched controls, respectively, mean +/- s.e.m., n = 7-8). These results suggest that the density of 5-HT3 receptor recognition sites labelled by [3H]-(S)-zacopride are not altered in the amygdala and hippocampus of patients with Alzheimer's disease.
