Kinetics and free-radical mechanisms of ageing and carcinogenesis.

Kinetic investigations play an important role in gerontology. Basis research on mathematical models of the ageing process in living organisms is described and illustrated by empirical data. The role of free-radical processes in the development of pathological states, including carcinogenesis, and in ageing is considered. The role of free-radical reactions in the conversion of known carcinogens in the organism and the interaction of their active radical forms with the most important biomacromolecules is discussed. It is shown that antioxidants of low toxicity, which inhibit radical processes, are anticarcinogenic, and data are given on their experimental utilization as possible 'geroprotectors'.

Potential anticancer agents--nitroxyl derivatives of Rubomycin.

Two new of the spin-labeled analogues of Rubomycin were studied. It was shown that spin-labeled derivatives were less toxic than the parent compound and had a broad-spectrum antitumor activity. Spin-labeled derivative was clearly less cardiotoxic in the rat model than Rubomycin (Daunorubomycin).

Assessment of lethal and cytostatic effects of alkylnitrosoureas on the cells of experimental tumors.

The action of a number of active anticancer agents belonging to the alkylnitrosoureas (MNU, DMNU, ADEKO, BCNU, CCMU, MeCCNU), as well as the effectiveness of a combined use of DMMU and ADEKO with cyclophosphamide was assessed on experimental tumors transplanted by inocula of different size. A mathematical model of the connection between survival of tumor-bearing animals with the kinetics of tumor growth was proposed to analyze the treatment effect. The contribution of lethal and cytostatic factors to the resultant effect of the drugs was evaluated. A combination of DMNU and cyclophosphamide was found to possess an additive effect. The synergism of the therapeutic action shown in a combined application of ADEKO and cyclophosphamide was stipulated by the potentiation of the lethal effect of these drugs on tumor cells.

Intensity of proliferation and sensitivity of experimental tumors to the 1-methyl-1-nitrosourea.

The kinetics of tumor growth and cell proliferation of eleven generations of spontaneous mammary carcinoma in F1(CBA2 X C57B1/6) mice was studied. Factors responsible for different sensitivity of tumors to 1-methyl-1-nitrosourea were analyzed. The process of tumor transplantation was accompanied by the reduction in latent period and the increase in the mean specific rate of tumor growth. The rise in tumor growth rate in consequent generations was associated with the increase of the number of DNA synthesizing cells. The fall in the intensity of the DNA synthesis was observed along with the tumor growth within one passage. The sensitivity of tumors to 1-methyl-1-nitrosourea was correlated reciprocally only with the initial labeling index of tumors and the specific rate of tumor growth.

Antitumor activity of 1,2-bis diazoacetyl ethane.

1,2-Bis diazoacetyl ethane (diazane) was selected for a detailed study as the most active compound in a large group of mono- and bi-functional diazoketones subjected to screening. In this paper data on the toxicity and cumulative and antitumor effects of diazane are reported. Diazane markedly inhibits the growth of certain experimental tumors and increases the mean survival time of animals with leukemias, even after a single injection. The drug is characterized by a high level of accumulation in the body.

Toxicity, antitumor activity, and pharmacokinetics of spin-labeled thioTEPA analogs.

Two spin-labeled analogs of thioTEPA, containing an iminoxyl radical, have been shown to have significant antitumor activity in several experimental tumor systems, combined with a marked decrease in toxicity compared with the parent compound, thioTEPA. Walker 256 carcinosarcoma, Guérin carcinoma, and Schweitz erythromyelosis showed complete regression using one of these analogs. Pharmacokinetic studies have shown that these compounds possess particular affinity towards tumor tissue.
