Cytoskeleton-associated antigens from African trypanosomes are recognized by self-reactive antibodies of uninfected mice.

Serum from uninfected mice of different strains, as well as from germ-free animals, contains antibodies which react specifically with at least two trypanosomal proteins, I/6 and MARP1. These antibody populations are highly specific for the respective proteins, are of similar affinity as hyperimmune antibodies, and consist of IgM as well as IgG isotypes. Hyperimmune antibody raised against the cross-reacting trypanosomal protein I/6 detects a 60 kDa protein in mouse 3T6 cells, which is a component of the fibroblast cytoskeleton.

Paired plateletpheresis with the CS 3000 plus blood cell separator using the new 30 ml low-volume collection chamber and the A-35 collection chamber.

Eight blood donors underwent paired plateletpheresis using the CS 3000 Plus blood cell separator. For each donor, two samples were run: one with the TNX-6 separation and the new 30-ml low-volume collection chamber (LVCC) and one with TNX-6 separation and the A-35 collection chamber. With the LVCC, 4.8 +/- 0.5 x 10(11) thrombocytes could be harvested, whereas 4.4 +/- 1.4 x 10(11) platelets (PLT) were collected with the A-35 chamber. The separation efficiencies were 51.7 +/- 6.6% for the LVCC and 43.8 +/- 10.7% for the A-35 chamber. The leukocyte contamination of the platelet concentrates was not statistically different.

T-cells and psychopathology in schizophrenia: relationship to the outcome of neuroleptic therapy.

Until recently, attention has been focused on the pathogenetic aspects of immunological findings in schizophrenic patients. There has been no mention of the relationship between such findings and schizophrenic symptoms. To study the probable relationship between T-cells and T-cell subgroups in the course and prognosis of schizophrenia, immunological and psychopathological parameters were correlated in 55 patients suffering from schizophrenia (ICD-9: 295.0-295.6) or schizoaffective psychosis (ICD-9: 257.7) before neuroleptic treatment. The correlations were performed for a second time in 24 of these patients after clinical improvement at a reinvestigation. Positive correlations of the Brief Psychiatric Rating Scale and the Scale for Assessment of Negative Symptoms with the numbers of CD3+ and CD4+ T-cells (total T- and T-helper cells), which were enhanced compared with controls, were found at the reinvestigation after clinical improvement, whereas no significant correlation could be detected at the pretreatment investigation. These results show that the cellular immune parameters are related to the course of the psychopathological symptoms in schizophrenia and, possibly, are a marker of the therapeutic outcome or neuroleptic treatment.

[Preliminary results of the 2nd multicenter study. Thrombocytapheresis with the Fresenius AS-104 blood cell separator and ring study of cell counting. Section of Therapeutic and Preparative Hemapheresis of the DGTI].

Only multicenter studies on cell separators give valid data to compare different cell separators. The aim of the 2nd multicenter study was to evaluate the separation protocol, software version V 4.61, of the Fresenius AS-104 cell separator for efficiency and deviation from predicted yields. Plateletpheresis data from twelve hemapheresis centers, using identical apheresis protocols and cell counting methods, were registered and statistically analyzed. Additionally, the counting methods of the centers were controlled by a ring study with biweekly external cell count trials. To get a comparison, the apheresis data, which depend on the center effects, were corrected by the values from the ring study. Preliminary results of 380 runs are 45.2 +/- 8.1% for the separation effectivity, 1.95% deviation from the predicted yield, whereby 90% of all runs deviated less than 20% from the predicted yield. 50% of products had a WBC contamination below 6 x 10(6).

German multicenter studies on plateletpheresis from healthy donors. German Hemapheresis Study Group.

It is reported on two German Multicenter Plateletpheresis Studies with the Fresenius AS 104 cell separator accompanied by a Multicenter Counting Study. 15 centers participated, a total of 1884 separations were evaluated. It could be shown that multicenter studies help to give a more objective opinion about a cell separator and the separation protocol used if it is possible to improve the counting methods and to eliminate individual center effects by an accompanying Multicenter Counting Study. The AS 104 proved to be a good cell separator for platelet collection regarding platelet yields (3.1 x 11" from 3.5 I citrated blood), separation efficiency (about 45%), yield prediction (SD = not equal to 15%) and leukocyte contamination (median WBC 47 x 10(6)).

Psychomotor disturbances in psychiatric patients as a possible basis for new attempts at differential diagnosis and therapy. V. Evaluation of psychomotor training programs in depressed patients.

Parts I-III of this series used psychometric assessment of motor performance in psychiatric patients and indicated a "psychotic-motor syndrome" (PMS) in schizophrenic and affective psychoses, which was not found in "neurotic"/reactive or healthy persons. Part IV yielded signs of concomitant brain dysfunction in these patients, demonstrated by EEG mapping as well as other (SPECT/PET) neuroimaging methods. Apart from this "basic science" interest into the pathophysiology of endogenous psychoses we engaged in the development of motor training programs using the PMS as "target" syndrome. We hypothesized, that motor training would not only improve disturbed motor behaviour, but ameliorate other symptoms of psychopathology also. These assumptions were supported in the first two independent studies involving n = 45 and n = 31 ICD-9 mono- and/or bipolar endogenous depressed patients, respectively (the studies on schizophrenic patients being reported finally as part VI of this series, along with the final version of our modified motor test battery). Examples of the motor training programs are provided in this paper, although the final version of the complete programs will be published separately for space reasons and for better availability for routine clinical use.

Identification and characterization of two repetitive non-variable antigens from African trypanosomes which are recognized early during infection.

The present paper describes two repetitive proteins representing common antigens of African trypanosomes which are non-variant and which are recognized early in infection by the host immune system. These antigens were identified by their ability to immunoreact with bovine serum taken during the early phase of a cyclic trypanosomal infection. Screening of a cDNA library from T. b. gambiense with such early infection serum identified a protein which contains a repetitive motif consisting of 68 amino acid repeat units (GM6). Immunofluorescence and immunogold electron microscopy revealed that GM6 is located on fibres which connect the microtubules of the membrane skeleton with the flagellum. A second repetitive antigen detected by this serum is MARP1 (microtubule-associated repetitive protein 1), a protein previously characterized in this laboratory as a high-molecular weight component of the membrane skeleton, which consists of more than 50 tandemly repeated, near-identical 38 amino acid repeat units. Beta-galactosidase fusion products of both proteins demonstrated a strong immunoreactivity with sera from T. b. brucei and T. congolense-infected cattle. The result from this preliminary immunological evaluation indicates a high immunodiagnostic sensitivity (90%) of the two recombinant antigens which make them interesting candidates for immunodiagnosis of trypanosomiasis in cattle.

Comparison of plateletpheresis using Cobe Spectra System and Fresenius AS 104.

To evaluate separation efficiency and concentrate purity, each of 50 donors underwent platelet (PLT) collection on both the Cobe Spectra and Fresenius AS 104 with the constancy of processed blood volume, ACD/blood ratio 1:10 and a blood flow of 50 ml/min. For the Fresenius AS 104 two protocols - V 4.11 and V 4.4 - were evaluated. Both systems provided high quality PLT collection: PLT yield 3.91 +/- 0.74 x 10(11) (Cobe); 4.16 +/- 1.12 x 10(11) (V 4.11); 4.66 +/- 0.89 x 10(11) (V 4.4). The mean cell contamination was low with all procedures, resulting in less WBC contamination with the Fresenius AS 104, whereby the higher efficiency obtained by the protocol version V 4.4 correlated with an increased of WBC contamination.

Leukocytapheresis: evaluation of a new procedure for the blood cell separator Fresenius AS 104.

For granulocyte separation from peripheral blood with the Fresenius blood cell separator AS 104 a new procedure and a new tubing set were developed and evaluated. Separation is done in cycles of 400 ml of whole blood processed, with continuous blood flow of 50-55 ml/min, speed 750 rpm. HES-450 is added for sedimentation acceleration together with citrate, ratio 1:10. At the end of each cycle granulocytes were pumped out of the separation chamber into the collection container. After processing 13 cycles of 400 ml whole blood/HES-citrate-volume a total yield of 9.20 +/- 2.80 x 10(9) white cells were collected in volumes of 125 ml. 47 +/- 9% of the leukocytes collected were granulocytes, this corresponds to collection of 1.05 +/- 0.09 x 10(9) granulocytes per liter of processed blood. The efficacy of granulocyte collection is 22.5 +/- 5.6%, viability is more than 95%. With this procedure it is possible to collect granulocytes with the blood cell separator Fresenius AS 104 in therapeutical doses.

Evaluation of a monocyte enrichment procedure using the blood cell separator Fresenius AS 104.

The small number of circulating monocytes in peripheral blood makes it difficult to collect enough cells for studies on the metabolism and analysis of receptor functions of this cell type for diagnostic aims and treatment. Therefore, a new procedure to enrich monocytes from peripheral blood using the cell separator Fresenius AS 104 was developed and evaluated. The separation is done using the tubing set 'P1Y-Gran-collect', which is a closed system with a single-stage separation chamber. The procedure is running continuously, while the cells are harvested cyclically. After processing of 4.9 l whole blood an average yield of (2.47 +/- 2.05) x 10(9) monocytes could be collected with an efficiency of 46 +/- 20%. The monocyte enrichment procedure using the Fresenius AS 104 provides a sufficient number of cells for the next steps: purification and concentration of the cells for drug monitoring, cell culturing, receptor analysis and further in vitro diagnostics.

Hemotherapy in neonatology.

The intensive care of very immature preterm infants is of special significance in the treatment of neonates with blood and blood components. A new, practicable transfusion concept has been developed for this group with its high and often acute need of hemotherapeutic measures: 30-ml packs containing packed red cells of blood group 0 Rh neg, free of 'immune' anti-A/B and anti-CMV-free, or deep-frozen fresh plasma of blood group AB. This concept has the advantages of simplified pretransfusional serology, reduced blood requirement with simplified storage, fresher stored blood due to more rapid turnover, reduced risk of infection through reduction in number of donors per patient, implying a reduced strain on patients and personnel, and the opportunity provided by the reduced thawing time and the eliminated risk of error to commence therapy immediately after the decision.

[Effect of shift work on somatoform disorders].

An overview of the literature shows that shift work is linked with an enhanced risk of psychiatric disorders. A number of authors have reported anxious and depressive symptomatology in shift workers, but there are no data about the frequency of somatoform disorders associated with this type of work. Psychosocial stress and low socio-economic status, both often found among shift workers, are regarded as risk factors for somatoform disorders. The connection of shift work and somatoform disorders is discussed with examples of four case reports. It is suggested that shift work, which is known to be associated with an increased incidence of cardiovascular diseases, may promote the occurrence of vegetative symptoms, such as feelings of weakness and dizziness, and eventually leads to intensified self-observation, anxiousness and hypochondriasis with predominantly somatic symptoms.

NMR studies of an oligoproline-containing peptide analogue that binds specifically to the H-2Kd histocompatibility molecule.

T lymphocytes expressing variable cell surface antigen receptors recognize "processed" forms of antigen, presented on the surface of other cells by molecules of the major histocompatibility complex (MHC). Naturally processed antigenic peptides can be replaced by synthetic ones. The synthetic peptide AYPPPPPTLA (P5) is an active competitor to the antigenic peptide HLA A24 170-182 (sequence RYLENGKETLQRA) that is recognized by A24 specific T cells in association with the H-2Kd class I MHC molecule. In P5 the five prolines were designed to play the role of a rigid spacer between the residue Y and the T-L unit, so as to mimic the role of Y171, T178, and L179 in the HLA A24 antigenic peptide, since these residues have proven to be the most important with respect to the binding of the HLA A24 peptide with the H-2Kd MHC molecule. Nuclear magnetic resonance studies allow us to demonstrate that in aqueous solution P5 adopts at least three long-lived conformations that can be classified with respect to the Y2-P3-P4 amide bonds as trans-trans, cis-trans, and cis-cis. Among these, the trans-trans form is present in 67% of the molecules while the two others share the remaining 33%.
