Increased tendency to reversible agglomeration in erythrocytes in outdated units has been previously demonstrated. This phenomenon occurred while post transfusion viability was still acceptable. In the present study erythrocytes of units with early agglomeration were investigated and compared with those with late agglomeration. There was no morphological characterization of these erythrocytes and their sialic acid content and the toluidine blue uptake were not lower than in late agglomeration. Thus, it seems that agglomeration reflects membranal changes, but is not directly related to the sialic acid content of the membrane or to its toluidine blue uptake.
A new type of direct-contact oxygenator has been tested satisfactorily on dogs in nine experiments. It is based on the principle of the "packed column" and has the advantage of a small hold-up (meaning a small priming volume), no formation of foam, and gentle flowing of the blood. The apparatus is simple enough to be commercially available at a low price and discarded after use. Both PO2 increase and PCO2 decrease were satisfactory and plasma hemoglobin values remained low after 75 minutes of perfusion. Preliminary in vitro experiments on ox and human blood showed a low degree of blood damage.
The murine plasmacytomas MOPC-104E and J558 secrete IgM-lambda and IgA-lambda, respectively, which are antibodies to alpha, 1-3 dextran, a constituent of dextran-S (DEX-S) extracted from Leuconostoc mesenteroides. A single i.p. injection of 10 microgram DEX-S into BALB/c mice from 7 days before and up to 3 days after the implantation of 5 x 10(3) plasmacytoma cells protected the BALB/c mice from developing the tumors. Immunization with other antigens, such as Escherichia coli lipopolysaccharide, inulin (alpha, 1-6 linkage), and dextran T-10 (no known alpha, 1-3 linkages) did not protect the mice from developing the tumors. Growth of LPC-1 was not affected by DEX-S. The mechanism of this growth inhibition is unknown. It does not appear to depend solely on binding of antigen to tumor cells, and the limits of the effective time intervals between antigen and tumor injection suggest dependence on the presence of antibody to alpha-1,3 dextran.
The effect of copper ions on the permeation of glycerol and its mono-, di-, and triacetate derivatives was studied in fresh and stored erythrocytes. Permeability was unchanged with storage. Acylated glycerols permeate the cells mainly by non-facilitated mechanism, as their transport is almost unaffected by copper ions.
We have previously reported increased tendency to agglomeration in CPD-stored erythrocytes. In the present study we investigated the deterioration pattern of fresh units, as detected by agglomeration, free hemoglobin levels, ATP levels, osmotic fragility, and certain enzymatic activities. When the negative test was converted to a positive one, the 24-h survival values of these units were determined by autotransfusion. A positive agglomeration occurred while viability was still above 70%. Fresh units had high ATP levels and negative agglomeration while old units had low ATP levels and positive agglomeration. However, in intermediate-storage units, no definite correlation between ATP and agglomeration was found. Agglomeration seems to reflect the degree of changes in stored erythrocytes. However, further studies are needed before this test may serve for prediction of post-transfusion viability.

Chickens were exposed to heat and dehydration, and the effects on their erythrocytes were examined. As chicken and camel (Camelus dromedarius) erythrocytes are of similar shape, the erythrocytes of the exposed chickens were compared with those of the camel. The salient findings were that chickens kept at 35 C without drinking water remained visibly unaffected. Their blood, however, showed hemoconcentration and increased erythrocyte osmotic fragility. Hemoconcentration could lead to an increase in body temperature of chickens in increased environmental temperatures.
