Smallpox diagnosed 400 years later: results of skin lesions examination of 16th century Italian mummy.

Results are presented on virological examination conducted on WHO request of a Naples infant mummy with smallpox-like lesions. Electron microscopy of lesions confirmed the findings of Italian researchers: well-preserved virus-like structures, whose size and morphology were identical to those of orthopoxviruses have been revealed. It was shown that the virus in mummy skin had lost its viability. Viral antigenic activity could not be detected in EIA or RPHA, nor was determined its DNA in the test of DNA molecular hybridization.

Monkey pox in humans: current results.

The presented studies were performed at the WHO Collaboration Center for Smallpox in Moscow in the framework of the WHO monkey pox project. The authors recommend improved methods for rapid detection of orthopoxvirus antigen, namely passive haemagglutination (PHA) using dried stable red blood cells and ELISA in order to provide more rapid and efficient laboratory diagnosis under field conditions. Independent serologic diagnosis of monkey pox by ELISA-adsorption (ELISA-A) was proved of value for epidemiological studies and for detection of inapparent infections. The application range of the latter technique and its limitations were also determined.

Antigenic properties of vaccinia virus and of the virus recombinant strains expressing heterologous genes.

Immunologic properties of vaccinia virus (VV), strain LIPV, and VV recombinant strains containing the gene of hepatitis B virus surface antigen and the TK gene of herpes simplex virus (HSV) have been studied. Production of antibodies against the majority of VV structural proteins, including nucleocapsid internal proteins was demonstrated in rabbits. Insertion of heterologous genes into the VV genome was without effect on the spectrum of antibodies produced against VV virion proteins. The data obtained in volunteers indicate that not only virus-neutralizing antibodies but also antibodies against most VV structural proteins are preserved in humans over many years. Reimmunization of volunteers with VV recombinant stimulates synthesis of antibodies against virion proteins whereas the spectrum of antibodies remains unchanged. Humans and rabbits did not differ in the spectrum of antibodies to VV virion proteins.

Properties of rabies virus (MNIIVP-74 strain) adapted to Japanese quail embryo cell culture.

The Pasteur strain of fixed rabies virus was adapted to primary cell cultures of Japanese quail embryos and designated as MNIIVP-74. In the course of adaptation the virus pathogenicity for rabbits by the intracerebral route decreased considerably and the pathogenicity for rabbits and adult white mice by extraneural routes was completely lost. After inoculation of Japanese quail embryo cell cultures, a titer of the virus in the culture fluid at 4 days was 6.25--7.0 lg LD50/ml (by the intracerebral inoculation of adult white mice). Viral antigen could be detected by immunofluorescence in the cytoplasm of approximately 60 per cent of the cells. Virus multiplication was accompanied by intensive interferon production. In cultures of BHK-21/13S cells the titer of the virus reached was 5.75 lg LD50/ml at 24 hours and about 30 per cent of the cells were affected. The MNIIVP-74 virus showed a high immunogenic activity in rabbits, guinea pigs and mice.
