Fluvirucins A1, A2, B1, B2, B3, B4 and B5, new antibiotics active against influenza A virus. II. Structure determination.

A series of structurally related antiviral antibiotics, fluvirucins A1, A2, B1, B2, B3, B4 and B5 have been isolated from the fermentation broths of five unidentified actinomycete isolates. Based on spectroscopic analysis, partial degradation experiments and 13C-enriched acetic acid-fed biosynthetic studies, their structures were elucidated to be 2.6,10-trialkyl-3(or 9)-aminoglycosyl-13-tridecanelactams.

Pumilacidin, a complex of new antiviral antibiotics. Production, isolation, chemical properties, structure and biological activity.

New antibiotic pumilacidins A, B, C, D, E, F and G were isolated from the culture broth of a strain of Bacillus pumilus. They are cyclic acylheptapeptide composed of a beta-hydroxy fatty acid, two L-leucine, two D-leucine, L-glutamic acid, L-aspartic acid and L-isoleucine (or L-valine). Pumilacidin components were inhibitory to herpes simplex virus type 1 and H+, K(+)-ATPase and demonstrated antiulcer activity in rat.

Lanthiopeptin, a new peptide antibiotic. Production, isolation and properties of lanthiopeptin.

A strain of Streptoverticillium cinnamoneum produced a peptide antibiotic named lanthiopeptin, which contained four unusual amino acids, erythro-beta-hydroxyaspartic acid, mesolanthionine, threo-beta-methyllanthionine and lysinoalanine. Lanthiopeptin showed antiviral activity against herpes simplex virus type 1 KOS strain infection in Vero cells by cytopathic effect reduction assay. The structure of lanthiopeptin is similar to that of ancovenin.

Effects of interferon on the lymphocyte subset in the blood and lymphoid organs in mice.

The effects of mouse interferon-alpha/beta (MuIFN) or poly(I):poly(C) injection on the lymphocytes in the blood, thymus, and spleen of C3H/HeN mice were examined. A single injection of MuIFN (2 X 10(6) to 2 X 10(7) IU/kg of MuIFN) or poly(I):poly(C) (0.12-1.2 mg/kg) caused a significant lymphocytopenia without any evidence of direct cytotoxic effects on lymphocytes in the blood or lymphoid organs, a significant decrease in the number of T cells both in the thymus and spleen along with a significant increase in the number of non-T cells in the splenic white pulp, and a differential decrease in the number of blood T-cell subsets ("suppressor/cytotoxic" subset was decreased continuously whereas "helper" subset only transiently). The differential effect on T-cell subsets could explain the complicated effects of IFN on the cell-mediated immune response.

Comparison of disappearance from blood and lymphatic delivery of human fibroblast interferon in rat by different administration routes.

Disappearance from the blood and lymphatic delivery of human fibroblast interferon (HuIFN-beta) in rats by the various administration routes were studied. Intravenous injection showed a rapid disappearance of HuIFN-beta from the blood circulation, however, the clearance rate was greatly decreased after 2 h and the half-lives were 15 min and 2.0 h. Intramuscular, subcutaneous and intraperitoneal injections maintained readily detectable levels in the blood for 12 h, and the half-lives were 2.3, 1.7 and 1.2 h, respectively. Lipid-surfactant mixed micelles greatly promoted the absorption of HuIFN-beta from the large intestine with extremely high selective lymphatic delivery, whereas intravenous and intramuscular injections showed no selective transfer into the lymphatics.

A method to potentiate enteral absorption of interferon and selective delivery into lymphatics.

The potentiated absorption of human fibroblast interferon (HuIFN-beta) with the aid of lipid-surfactant mixed micelles from the rat large intestine and lymphatic delivery were studied. The administration of HuIFN-beta with saline solution alone into the lumen of the large intestine indicated no detectable HuIFN-beta in the serum or the lymph for 5 h. Neither lipid (linoleic acid) nor surfactant ( HCO60 , polyoxyethylene hardened castor oil derivative) could promote the absorption of HuIFN-beta. However, the administration with linoleic acid- HCO60 mixed micelles enabled the great absorption of HuIFN-beta from the large intestine, and HuIFN-beta was delivered into the lymphatics with an extremely high selectivity.

Chicamycin, a new antitumor antibiotic. II. Structure determination of chicamycins A and B.

Structures of chicamycins A and B have been determined from a series of chemical degradation studies coupled with spectroscopic analysis. Chicamycin A is 2(S),11(R), 11a (S)-1,2,3,10, 11, 11a -hexahydro-2,8-dihydroxy-7,11-dimethoxy-5H-pyrrolo-[2,1-c] [1,4]-benzodiazepin-5-one, and chicamycin B is 2(S), 11a (S)-1,2,3, 11a -tetrahydro-2,8-dihydroxy-7 -methoxy-5H-pyrrolo-[2,1-c][1,4]-benzodiazepin-5-one which is the demethanol form of chicamycin A. The structure of chicamycin B is closely related to that of neothramycin , differing only in the position of a hydroxyl substituent on the pyrrolidine ring.

[Pharmacokinetics of human fibroblast interferon in rats].

Pharmacokinetic behaviors of human fibroblast interferon (HuIFN-beta) administered intravenously to rats were examined. After intravenous injection, plasma interferon level decreased in a biexponential fashion having the half-times of 15 min and 120 min respectively. Distribution levels of HuIFN-beta in the tissues at 30 min, though the actual concentrations were less than that of plasma, were observed according to the following order; kidney greater than adrenal greater than lung greater than spleen greater than liver greater than testis. Small amounts of HuIFN-beta were also detected in the thoracic lymph and bile but no HuIFN-beta was found in urine. The in vitro study indicated that the stability of HuIFN-beta was different in each tissue homogenates and body fluids. From the results obtained, we concluded that HuIFN-beta exhibited a low distributional character to the tissues when administered intravenously. It was also suggested that the kidney and the liver were the main catabolic sites of HuIFN-beta.
