Functional association of immediate early gene c-fos with nuclear matrix.

Accumulation of c-fos gene locus DNA in the nuclear matrix of hepatocyte nuclei was observed during induction of c-fos with cycloheximide. No enhanced association with the nuclear matrix was detected for inactive immunoglobulin gene locus. The use of endogenous DNAses allows isolation of nuclear matrix preparations enriched with transcribing chromatin.

Dual effects of endogenous DNases on transcriptionally active and inactive chromatin.

Experiments on resting hepatocytes with inactive c-fos gene and active albumin gene. We revealed that DNA of the transcribed gene is less susceptible to the influence of endogenous Ca2+/Mg(2+)-dependent DNases in matrix-associated and highly soluble chromatin fractions. In the fraction of low soluble chromatin active gene was more accessible for DNases. Our results indicate that activity of endogenous DNases can change in the transcribed gene locus.

[Complex use of benzodiazepines, lithium salts and haloperidol in the treatment of obsessive movements and tics of different origin].

Seventy-two children and adolescents with obsessive movements and tics of residual-organic etiology were treated by combined application of benzodiazepins (diazepam, phenazepam), lithium carbonate and haloperidol. The nature of obsessive syndromes and tics is considered in the light of the theory of generator mechanisms of neuropathological syndromes. The use of the studied combination due to its synergic action on the structures of the pathological system elicited a good therapeutic response while using lower doses of each drug (half to two-thirds of the conventional age-specific dosage). Reduction in drug doses in combined therapy decreases the risk of side effects and complications. The above treatment was followed by disappearance of essential and facultative symptoms in 21 patients, considerable improvement with a marked reduction of facultative symptoms in 22 cases, clear-cut improvement in combination with decreased facultative symptomatology in 14 patients, and insignificant improvement in 6 patients.

[Transport of basic amino acids into Yersinia pestis cells].

The transport of the basic amino-acids into the plague microbe cells has been shown to proceed by means of two transfer systems, the common transfer system and the system specific for lysine. Besides that, arginine can be transported into the plague microbe cells by the histidine transport system. The vaccine strain EV NIIEG was used as a model to study the amino-acid transport systems.

[Endogenous DNases as a tool for isolation of nuclear matrix: critical parameters of nucleolysis].

Degree of nucleolysis has critical significance for isolation of nuclear matrix (NM) specifically enriched in transcribed DNA sequences as demonstrated at the example of inactive (c-fos, c-myc, and Ck) and active (p53, albumin, and 28S rRNA) genes in resting hepatocytes. Optimal degree of nucleolysis features degradation of loop domains of chromatin with preserved relatively uniform molecular weight distribution of DNA. Deviation from these parameters leads to nonspecific fragmentation of chromatin in various gene loci and isolation of NM samples nonspecifically enriched or depleted of transcribed DNA sequences. Under optimal hydrolytic conditions, the transcribed chromatin is more resistant to endogenous DNase attack, which allows selective conservation of its association with the nuclear matrix.
