[Comparative hepatic toxicity of isoniazid, rifampicin and ethambutol].

Isoniazid, rifampicin and ethambutol administered in equimolecular dosages (50, 250 and 100 mg/kg, respectively) for 4 days caused liver affection. This was manifested by elevated activity of serum aminotransferases and alkaline phosphatase, inhibition of lipid peroxidation of hepatocyte membranes and antioxidative system dysfunctions. Isoniazid and rifampicin were proved to be most hepatotoxic. The combined use of antituberculous drugs increases their membrane-damaging action, especially when isoniazid and rifampicin are used together, but mostly when isoniazid in combination with rifampicin and ethambutol is applied.

[Effectiveness of tocopherol and anti-hypoxic agents in liver damage caused by antitubercular agents].

Two-week administration of isoniazid, rifampicin (50 mg/kg and pyrazinamide (1,5 g/kg) to white rats brings about liver affection characterized by a higher activity of alanine and aspartate aminotransferases, lipid peroxidation activation and bile production inhibition. With the liver affected by antituberculous drugs, protective action is provided by acetate tocopherol an antioxidant, and piracetam riboxin and pyriditol, antihypoxic agents.

[Effectiveness of legalon and essentiale in a tetracycline-induced liver lesion].

It was shown in the experiments with albino rats that legalon and essentiale had a pronounced hepatoprotective effect in tetracycline affections of the liver. They retarded the increase in the activity of alanine aminotransferase, aspartate aminotransferase and alkaline phosphatase in blood serum, prevented suppression of cholopoiesis by tetracycline and inhibited lipid peroxidation. The drugs may be used for prevention and treatment of tetracycline affections of the liver.

[Synergistic effect of rifampicin on hepatotoxicity of isoniazid].

It was shown in experiments on male albino rats that rifampicin potentiated the hepatotoxicity of isoniazid. As compared to the use of isoniazid alone, its combination with rifampicin resulted in a higher activity of transaminases and alkaline phosphatases and a higher rate of inhibition of biliary secretion and synthesis and excretion of bile acids, bilirubin and cholesterol with bile. Moreover, an increase was observed in the level of lipid peroxidation products of the hepatocyte membranes in liver homogenates and blood, which was indicative of an increased intensity of lipid peroxidation. The increased hepatotoxicity of isoniazid was evident from a more pronounced decrease in the number of sulfhydryl groups accompanied by an increase in the number of disulfide ones in the liver and blood. The authors suggest that potentiation of isoniazid hepatotoxicity under the action of rifampicin was due to its inducing activity with respect to the microsomal oxidation enzymes.

[Effect of ethyl alcohol on bile production and lipid peroxidation in tetracycline-induced liver lesions].

The experiments with male albino rats showed that in toxic doses tetracycline induced affections of the liver evident from activation of transferases and alkaline phosphatases in the blood serum, inhibition of bile secretion, synthesis and secretion of bile acids and excretion of bilirubin and cholesterol with the bile. Moreover, the content of the products of lipid peroxidation in liver homogenates increased. Acute intoxication with ethyl alcohol also induced hepatic disorders. However, they were less pronounced. The intoxication was accompanied by inhibition of cholopoiesis associated with increasing free radical oxidation of lipids. On combined use of tetracycline and ethyl alcohol no potentiation or summation of their hepatotoxicity was observed.

[Use of bioflavonoids to prevent the harmful action of tetracycline on the liver].

It was shown in experiments on male albino rats that the plant bioflavonoids flamin, flakumin and tanaflon had hepatoprotective properties in liver function impairment caused by tetracycline intoxication. This was evident from a significant improvement of biliary secretion, synthesis and secretion of bile acids and bile secretion of bilirubin and cholesterol not only in the animals untreated with tetracycline but also in the controls. A decrease in the activity of alanine aminotransferase, aspartate aminotransferase and alkaline phosphatase in the serum was indicative of the pronounced hepatoprotective effect of the bioflavonoids. They inhibited lipid peroxidation in the hepatocyte membranes evident from a decrease in the content of malonic dialdehyde and diene conjugates in liver homogenates. The antioxidant properties were especially pronounced in tanaflon. Correlation between lipid peroxidation, activity of the transferases and alkaline phosphatase and cholopoiesis both in tetracycline intoxication and in the prophylaxis and treatment with flamin, flakumin and tanaflon was shown.
