[Regulatory effects of human mononuclear cells treated with diucifon may be dependent on membrane-associated form of interleukin-2].

The immunomodulating activity of diucifon--activated and glutaraldehyde fixed human mononuclear cells (MNC-DGA) was studied in the test--system of autologous and allogenic mononuclear cells proliferative response. It was shown that MNC-DGA had the same stimulating activity as the diucifon treated unfixed cells. The immunomodulating capacity of MNC-DGA was fully abolished in the case MNC-DGA. So, the interaction between interleukin 2 receptor (Tac--antigen) and the membrane form of the mediator, expressed on MNC after diucifon treatment may play an important role in MNC-DGA immunomodulating activity. The regulating activity of MNC-DGA was revealed both in autologous and allogenic systems. The speculation is that construction of new immunomodulating drugs as complexes of cell-like carrier and immunomodulators fixed on their surface may be perspective.

[Modulating effect of mast cells on the proliferation of cells from various lymphoid organs of rats].

There are some reasons for suggesting that mast cells (MC) can modulate lymph cell (LC) activity. The blast-transformation test was used to study the effect of serum MC of Wistar and ACI rats on the proliferation of splenic, thymus and lymph node LC in vitro. Rat MC modulate lymphocyte proliferation. Depending on their concentrations, serum MC increased spontaneous and T-mitogen induced proliferation of splenic and lymph node cells. T-mitogen thymocyte proliferation was also increased. The promoting effect was maximal when the MC proliferative response was induced by suboptimal doses of mitogens. No genetical restriction was found for development of the MC promoting effect.

[The therapeutic use of diucifon-activated immunocytes in adoptive transfer in mice with burn trauma complicated by Pseudomonas aeruginosa infection].

The effectiveness of the therapeutic use of diucyphone-stimulated syngeneic spleen cells was studied in mice with a burn trauma complicated by P. aeruginosa infection. For this purpose CBA mice were subjected to a burn of degree IIIa, covering 30% of their body surface, the wound was infected with P. aeruginosa strain, and 24 hours later spleen cells were taken from some of these animals. The spleen cells, incubated for 3 hours with or without diucyphone at a concentration of 10-100 micrograms/ml, were washed and reinfused intravenously to the remaining animals. The injection of diucyphone-treated spleen cells was shown to greatly increase the survival rate of the animals, to accelerate healing processes and to enhance the proliferative response of spleen cells to mitogens (phytohemagglutinin, concanavalin A) and interleukin-2. A conclusion was made on the efficacy of the therapeutic use of diucyphone-activated immunocompetent cells in cases of immunodeficiency induced by burn infection.

[Cells-boosters as the modulators of immunogenesis].

In the course of mouse immune response to ram erythrocytes, cells capable of enhancing immune and proliferative response (enhancing cells, or EC) are shown to appear in the spleen. The EC effect is antigen-unspecific. Cycloheximide (CH), an inhibitor of protein synthesis, speeds up EC development, which is associated with a greater immune response. In vitro CH treatment of mouse spleen cells (SC), for 3 hours at 37 degrees C, CH taken in a concentration of 1-3 micrograms/ml, resulted in in vitro induction of EC, capable of enhancing proliferative response to PHA and recombinant IL-2 in normal SC and increasing immune response to ram erythrocytes. Glutaric aldehyde fixation of EC was not reflected in their immunostimulating activity, either in vivo, or in vitro. The data obtained point to the significance of CH-induced changes in membrane-associated structures in mediating EC effect. It is assumed that the agents, altering the expression of functionally-significant structures (membrane-linked mediators and auxiliary intercellular interaction molecules) on cell surface, may be used for extracorporeal treatment of lymphoid cells to be eventually re-introduced into the body to control immunologic disorders.

[Clinico-immunologic characteristics of acute toxic-allergic reactions to drugs and current methods for their treatment].

The paper is concerned with clinicoimmunological and allergological characterization of 29 cases of acute toxic allergic reactions (ATAR) to drugs. Four degrees of severity of disease were identified; the most severe form was toxic epidermal necrolysis caused by the use of sulfanilamides and pyrazolones in patients with acute respiratory virus and bacterial infections. Blood analysis in patients with III-IV degree of severity showed a sharp decrease in Ctot and C3 up to 0, an increase in the level of circulating immunocomplexes and "average molecules". Cellular immunity in III degree of severity was decreased. The main principles of therapy using extracorporeal immunocorrective methods were worked out. A leukocyte natural migration inhibition test was used for specific diagnosis of intolerance to medication.

[Staged analysis of the lowered proliferative response of human lymphocytes to mitogens].

The current concepts of the mechanisms of initiation and regulation of the proliferative response of lymphoid cells underlie a series of research performed by the authors. Using the modern methods an attempt was made to analyze comprehensively the causes of its lowering in patients with congenital immune deficiency (ataxia-telangiectasia, the Louis-Bar syndrome). It was established that irresponsiveness of the patients' cells to mitogens is not associated with the inability of these cells to interact with mitogen, to produce the growth factor of T cells or adsorb it on their membrane. It is suggested that the derangement is likely to be connected with the blockade of the postmembrane component of the proliferation trigger. It is also assumed that the analysis of the low proliferative response employed by the authors can be successfully used for other pathologies.

[Properties of T-cell growth factor obtained from diucifon-stimulated human lymphocytes].

The properties of T-cell growth factor (TCGF), obtained by diucifon (Dc) stimulation of human mononuclear cells (MNC) (TCGF-Dc) have been studied. Taking into account the fact that Dc alone does not, like other TCGF inductors, cause proliferation, differences between TCGF-Dc and TCGF were suggested. Partial purification of supernatant from cells, activated by Dc was performed on Sephadex G-100 column. TCGF-Dc biological activity in these fractions was determined in the system of mitogen activated human MNC and mice thymocytes, as well as in the system of concanavalin A transformed cells. 2 peaks of TCGF-Dc activity have been revealed that are indicative of TCGF-Dc molecular mass heterogeneity. In contrast to TCGF, low molecular mass TCGF-Dc (8000-12000) and TCGF-Dc from the whole supernatant were capable of absorbing on intact human MNC. TCGF-Dc may be constantly present on MNC membrane, but TCGF-Dc fixation is not sufficient for proliferation induction, the receptor activation is necessary as well. Receptors to TCGF-Dc were suggested to consist of fixing and triggering sites.

[Correlations between nonspecific suppressor activity of mononuclear cells and IgE levels in patients with pollinosis].

The total IgE level and some indicators of nonspecific suppressor activity (NSA) (the amount of theophylline-sensitive sells and the activity of Con A-induced spontaneous and short-lived suppressors) were studied in 36 pollinosis patients with high sensitivity to timothy dust, confirmed by the data of the allergological anamnesis, the data of skin and provocative tests with timothy allergen, and by the presence of allergen-specific IgE antibodies. The IgE level ranged from 23 to 2925 KU/1. Preincubation of mononuclear cells with theophylline did not lead to a significant diminution of the number of E-RFC. The majority of patients demonstrated the activity of all the above-indicated suppressors upon the use of mitogens at several concentrations. Correlation of the NSA with the IgE magnitudes showed disagreement in all the cases with the exception of the test with short-lived suppressors (a variant of the test culture with Con A) where in one group of patients (11 examinees), a direct relationship was found, whereas in the other one (10 examinees), a reverse relationship. It is concluded that the test forms of NSA of lymphoid cells do not play an essential role in the maintenance of the total IgE level in pollinosis patients.

[The immunological indices of guinea pigs modelling Marburg hemorrhagic fever].

Presents data on changes in nonspecific immunity (interferon, tumor necrosis factor, natural killers), spontaneous and mitogen-induced lymphocyte proliferation in the course of development of experimental Marburg fever. Demonstrates the effects of the said factors and interleukin-2 on the course of the fever and their role in a lethal outcome of the disease.

[Experimental study of possible treatment of Marburg hemorrhagic fever with desferal, ribavirin, and homologous interferon].

The paper presents data on the effects of the drugs desferal, ribavirin and homologous (guinea pig) interferon on the course of experimental Marburg fever. Neither ribavirin nor interferon were effective. A moderate response to desferal was noted. It is suggested that tumor necrosis factor may contribute to development of Marburg hemorrhagic fever.
