Glucose utilization, serum insulin, human growth hormone, and free fatty acids were studied in 18 unipolar depressed patients and 14 normal controls, using the intravenous glucose tolerance test. Patients were kept drug-free for at least two weeks before testing, and physical activity and diet were carefully controlled. Previously described abnormalities in the balance between glucose and free fatty acids were not seen, but endogenous depression was associated with lowered glucose utilization rate with insulin resistance. Changes in kinetics of membrane transport in endogenous depression which could account for this finding are discussed.
Elevations in serum immunoglobulins, frequently monoclonal in nature, are known to occur in patients with the adult form of Gaucher's disease. We describe amyloidosis in a 46-year-old woman of Italian ancestry with Gaucher's disease, who also had 3100 mg/dl of monoclonal IgA. She died of restrictive cardiac disease. A 50-year-old sister, also with Gaucher's disease, had 1300 mg/dl of polyclonal IgM but no evidence of amyloidosis. A glucosyl sphingosine-Sepharose affinity column provided no evidence that the large amount of immunoglobulin present has specific affinity for glucosyl ceramide. This, to our knowledge, is the first report of amyloidosis complicating Gaucher's disease.
Lecithin can be prepared on a relatively large scale, free of colored impurities, by a simple two-column procedure. Commercial crude egg lecithin is partially purified by a single-step passage through an alumina column. It is then purified by a two-step passage through a prepacked, commercial silica gel column. The lecithin is prepared in solvent-free form for weighing by lyophilization from cyclohexane. Toxic solvents (chloroform and methanol) are avoided by the use of ethanol, isopropanol, hexane, and water. The elutions are easily monitored by a flow cell in an ordinary spectrophotometer set at 215 nm. Study of the column parameters has made it possible to use heavy loads with a relatively small column and minimal solvent.
Analogs of ceramide which inhibit galactocerebrosidase also demyelinate or inhibit myelination in organ cultures of rat cerebellum. The potency of the analogs in culture correlated with their effectiveness as inhibitors of cerebrosidase, but not with their effectiveness as inhibitors of galactosyl transferase. The most effective compound was the decanoyl amide of 3-phenyl-2-amino-1,3-propanediol with erythroconformation. Stimulators of cerebrosidase also demyelinated cultures. With both groups of compounds, myelin sheaths became distorted, then broke into lipid droplets. Axons were preserved, but neurons showed some nuclear changes and granularity. Metabolic studies with the most effective inhibitor showed that glucose incorporation into cerebroside and other alkali-stable lipids was initially depressed compared to proteins and total lipids.

Normal human skin fibroblasts were grown in the presence of N-hexyl-O-glucosyl sphingosine (HGS), an inhibitor of aryl glucosidase and glucocerebrosidase. Tests of the cells with aryl glycosides showed that beta-glucosidase activity in the cells was drastically reduced while other enzyme activities (alpha-glucosidase, beta-galactosidase, and N-acetyl-beta-hexosaminidase) were normal or elevated. Exposure of cells to HGS for 28 days resulted in increased values for cell weight per plate, glucocerebroside concentration, and galactosyl-galactosylglucosyl ceramide concentration. The concentrations of total lipid, cholesterol, and protein were unchanged, as was the fatty acid distribution within the glycolipids. Chemically, the inhibitor-treated cells exhibited a model form of Gaucher's disease. Although many membranous cytoplasmic inclusions were induced by HGS, they were unlike the characteristic inclusions seen in individuals with the genetic disorder. Skin fibroblasts from a Gaucher patient showed no abnormalities in composition or appearance.
An improved method for isolating cerebrosides from natural sources is described. The method is particularly suited to large scale work and can be adapted to the isolation of sphingolipids that are less polar than the gangliosides. It is based on the use of sodium sulfate to absorb the water from chloroform-methanol tissue extracts, the use of triiodide to cleave the ether linkage of plasmalogens, and the use of alkaline methanolysis to cleave the ester linkages of the glycerolipids. The final separation of the lipids is done with a silica gel column.

Glucocerebrosidase activity was reconstituted in vitro from a soluble glycoprotein factor (P) and a particle-bound factor (C). The physiological significance of the system is discussed.
