[A parallel study of CT findings and histopathology of metastatic liver tumors].

The relation between the CT findings and the histopathology of metastatic liver tumors was investigated in nine autopsy cases of liver metastases. In hypervascular and necrotic tumors, large necrotic lesions demonstrated by CT were almost consistent with the pathological findings, but small metastatic nodules were better identified on plain CT than those on contrast-enhanced CT. Postmortem CT examination of the liver obscured metastatic nodules as compared with diagnostic CT. Diffuse liver involvement of lymphoma cannot be identified on CT examination. Plain CT is considered to be useful for screening liver metastases.

Topographical study on arteriosclerotic lesions at the bifurcations of human cerebral arteries.

We have studied the topographical distribution of arteriosclerotic lesions at the bifurcation of the internal carotid-anterior cerebral-middle cerebral arteries (internal carotid bifurcation) and of the middle cerebral artery-first temporal branch (first bifurcation of M.C.A.) in humans. The arteriosclerotic lesions showed a distinct pattern with a high incidence on the outer walls of the daughter vessels and at the inner curvatures in the bifurcations where wall shear stress was believed to be relatively low. However, there were differences in the distribution of the lesions between the internal carotid bifurcations and the first bifurcations of M.C.A.. The former are considered to be three-dimensionally unsymmetrical and curved, and the latter symmetrical and straight. The present study suggests that lower shear stress is of considerable importance in both the initiation and localization of arteriosclerotic lesions, and that study of the three-dimensional blood vessel architecture and blood flow patterns needs to be done to clarify the role of hemodynamic forces in atherogenesis.

Azuki bean (Vigna angularis) protease inhibitors: isolation and amino acid sequences.

Double-headed protease inhibitors I, IIa, and IIc (AB I, AB IIa, and AB IIc) have been purified from azuki beans "Takara" (Vigna angularis) by conventional chromatographic methods and their amino acid sequences have been determined. AB I, AB IIa, and AB IIc had molecular weights of 9,166, 8,661, and 8,756 daltons, consisting of 82, 78, 79 amino acid residues, respectively. The molecular weights of these inhibitors, determined by gel filtration at pH 8.0, were 18,000 for AB I and 17,000 for both AB IIa and AB IIc, indicating that the inhibitors are dimers. The inhibitors had isoelectric points of 4.7 (AB I), 6.8 (AB IIa), and 6.2 (AB IIc). AB I stoichiometrically inhibited both trypsin and chymotrypsin at a molar ratio of 1 : 1. On the other hand, AB IIa and AB IIc both inhibited trypsin at a molar ratio of about 1 : 2 and also inhibited chymotrypsin, though only weakly. Sequence comparison with other double-headed inhibitors indicated the reactive sites of AB IIa and AB IIc for trypsin to be Lys26-Ser27 and Arg53-Ser54, and those of AB I for trypsin and chymotrypsin to be Lys26-Ser27 and Tyr53-Ser54, respectively. The differences between AB IIa and AB IIc were that AB IIa lacked the C-terminal aspartic acid residue, and that Glu10 and Arg60 in AB IIa were replaced by Gln10 and His60 in AB IIc. A comparison between AB IIa and AB I revealed 25 variant amino acids among the 78 residues of AB IIa; further, Ab IIa lacked 4 amino acid residues in the C-terminal region of AB I.

[CT and pathologic studies on detecting hepatic involvement of malignant lymphoma].

In order to determine the value of CT in detecting lymphomatous involvement of the liver, 57 autopsied cases (48 cases of non-Hodgkin lymphoma and 9 of Hodgkin's disease) were analyzed in detail. Additionally, 90 recent cases of abdominal CT with liver study in the initial staging were reviewed and 6 cases in which the liver had been examined within 2 weeks before autopsy were compared for macroscopic and pathologic findings. The following results were obtained. Autopsy findings in 57 cases of malignant lymphoma. (1) Hepatic involvement was demonstrated in 56% of the patients with non-Hodgkin lymphoma and in 67% of those with Hodgkin's disease. Of these patients with hepatic involvement, intrahepatic nodular lesions over 1.0 cm in diameter were macroscopically identified in 22% of the cases of non-Hodgkin lymphoma and 33% of those with Hodgkin's disease. Lymphomatous involvements were seen in many organs in the cases of hepatic involvement. CT examinations of 90 cases for the initial staging showed no significant abnormality in the liver or spleen. Of the 6 cases in which the liver was examined by CT within about 2 weeks before autopsy, lymphomatous involvement was pathologically diagnosed in 5. Of these, only one having multiple nodules larger than the tip of a thumb was correctly diagnosed by CT. Hepatic involvement of lymphoma was thought to be the end stage, and minimal or early disease was detected in the region of Glisson's capsule. The above findings indicate that early hepatic involvement is not likely to be detected by CT at the present time.

Inhibition of DNA replication and membrane transport of some nutrients by clazamycin in Escherichia coli.

Growth of Escherichia coli in a nutrient medium was inhibited by 100 micrograms/ml of clazamycin and at this concentration, the viable cell number decreases slowly. Elongated cells were observed in the treated cultures. The bactericidal activity was abolished by high concentrations of either sucrose or sorbitol but not by chloramphenicol. Non-growing cells suspended in a medium devoid of both carbon and nitrogen sources were killed by clazamycin more rapidly than cells in a rich medium. Incorporation of radioactive thymidine, uridine, leucine and N-acetylglucosamine into cellular macromolecules was inhibited to a similar extent. Permeability of N-acetylglucosamine and leucine was blocked by clazamycin . On the other hand, membrane transport of thymidine was only slightly inhibited. Thymidine-derived radioactivity accumulated as dTTP in the cells suggesting that DNA synthesis was blocked at the polymerization step. DNA synthesis in toluene-treated cells was also sensitive to clazamycin while the repair DNA synthesis induced by bleomycin in these cells was not. DNA-repair deficient mutants of E. coli were as sensitive to clazamycin as their DNA-repair proficient counterparts.

[Effects of PS-K as adjuvant therapy for primary lung cancer--an autopsy case].

A 57-year-old woman underwent left lower lobectomy for primary lung cancer (papillary adenocarcinoma, T2NOMO). Despite postoperative adjuvant immunochemotherapy with 5-Fu, Carboquone and PS-K, bone metastases were recognized at 9 months after the surgery. She received radiation therapy and was administered PS-K alone, 3.0 g daily. As a result, long-term good condition was maintained with a positive PPD skin test. The bone metastases developed slowly, and the patient survived 5 years and 8 months after the operation. At autopsy, no obvious metastatic lesions were identified except for the bone metastases. The cause of her death was thought to be acute renal failure due to severe hemorrhage from an esophageal ulcer. Moreover, she was doomed because of advanced polycystic disease.

Scanning and transmission electron microscopic study on the cerebral arterial endothelium of experimentally hypertensive rats fed an atherogenic diet.

In order to clarify the roles of hypertension and hypercholesterolemia in atherogenesis, the cerebral arterial endothelium of experimentally hypertensive female rats fed an atherogenic diet were investigated by scanning and transmission electron microscopy. The hypertension was induced by constriction of the bilateral renal arteries. In both hypertensive groups, group I (hypertension + hypercholesterolemia) and group III (hypertension), many endothelial cells covered with numerous microvillous projections were observed. Pinocytotic vesicles and caveolae were also increased in these endothelial cells. On the other hand, in group II loaded with only hypercholesterolemia, microvillous projections on the endothelial cells tended to be increased, but its grade seemed to be lower than those of the hypertensive groups. These findings suggest that hypertension appears to be more effective than hypercholesterolemia in the cause of the increase of microvilli on the endothelial cell surfaces and the increase may be related to increased permeability in the cerebral arteries.

Aminopyrazine analogues as chemiluminescence derivatization reagents for pyruvic acid.

Aminopyrazine analogues were studied as sensitive and selective chemiluminescence derivatization reagents for pyruvic acid. These analogues reacted with pyruvic acid under acidic conditions at 100 degrees C to produce Cypridina luciferin derivatives, which exhibit chemiluminescence by reaction with hydrogen peroxide in the presence of potassium t-butoxide in dimethylformamide. Of the four aminopyrazine analogues (2-amino-5-phenylpyrazine, 2-amino-5-(4-hydroxyphenyl)pyrazine, 2-amino-5-(3,4, 5-trimethoxyphenyl)pyrazine, and 2-aminoquinoxaline), in the present test 2-amino-5-(3,4,5-trimethoxyphenyl) pyrazine was the most sensitive for pyruvic acid, and the chemiluminescence intensity was about four times higher than that obtained with aminopyrazine.

Pharmacodynamic study of procaterol hydrochloride dry powder inhaler: evaluation of pharmacodynamic equivalence between procaterol hydrochloride dry powder inhaler and procaterol hydrochloride metered-dose inhaler in asthma patients in a randomized, double-dummy, double-blind crossover manner.

Therapeutic equivalence between procaterol hydrochloride dry powder inhaler (Meptin DPI) and procaterol hydrochloride metered-dose inhaler (Meptin MDI), the currently marketed formulation, was assessed in 16 patients with bronchial asthma. The study was conducted in a randomized, double-dummy, double-blind crossover manner, using forced expiratory volume in the first second (FEV1) as an index of bronchodilatory effect. In Period I, the patients received 20 mcg of either Meptin DPI or Meptin MDI, and then crossed over in Period II after a washout interval of 3--28 days. Pharmacodynamic equivalence was accessed using AUC (FEV1)/h and peak FEV1 as indices, and the data were analyzed by analysis of variance (ANOVA). Factors used for the analysis were the treatment group and/or carryover effect, patients within each group, period, and treatment. The 90% confidence intervals for the differences between the two treatments were --0.0995 to --0.0204 (L) for mean AUC (FEV1)/h and --0.102 to --0.022 (L) for mean peak FEV1, both within the acceptance criteria of --0.15 to 0.15 (L). Meptin DPI was therefore assessed as being equivalent to the current Meptin MDI.

Origin of an 84-kDa protein with ABH blood-typing antigen activity in human seminal plasma.

In order to investigate the origin of an 84-kDa protein with ABH blood-typing antigen activity (p 84) and its concentration in human seminal plasma, a monoclonal antibody (mAb p 84) was produced. The protein was recognized in breast milk as well as in seminal plasma by an indirect, enzyme-linked immunosorbent assay (ELISA) using this mAb. mAb p 84 identified 84-kDa and 83-kDa forms of the protein in seminal plasma and breast milk, respectively, on immunoblotting. The mean concentration of p 84 in seminal plasma was 949 microg/ml (n = 54 subjects). There was no significant difference in the concentration of p 84 between individuals who secreted (Se) or did not secrete (non-se) the ABH antigen into their seminal plasma, nor were there any significant correlations between the concentration of p 84 and the total seminal protein concentration. An immunohistochemical study using mAb p 84 with light microscopic detection showed that p 84 was located in the cytoplasm of the inner layer of pseudostratified cuboidal epithelial cells of the seminal vesicles, but no immunoreactivity was found in the prostate. These data indicate that p 84 originates from a single tissue, the seminal vesicles, and suggest that p 84 is an ABH epitope-bearing protein that has not previously been identified but possesses some immunological properties similar to those of lactotransferrin.
