Malignant lymphoma of the testis: histological and immunohistological study of 28 cases.

Histological and immunohistological studies were made in 28 patients with testicular lymphoma who presented with a right (15), left (11) or bilateral (2) scrotal swelling. Patient age at hospitalization ranged from 39 to 87 years, with a median age of 64 years. The disease was stage 1 in 12 patients, stage 2 in 11 and stage 3 in 5. Histologically, most of the neoplasms were of the diffuse large cell type, with an immunoblastic type being the most common. Immunohistochemistry revealed that all but 2 cases were of the B cell type. None of the tumors reacted with monoclonal antibody for T cell. These findings suggested that testicular lymphoma exclusively consisted of the B cell type, identical to previous reports of extranodal lymphomas other than those in the skin. The pertinent literature is reviewed and the histogenesis of testicular lymphoma is discussed.

Distribution of vitamin B12 R-binder in lung tumors. Implications for cell differentiation.

Expression of vitamin B12 R-binder, a specific binding protein for vitamin B12, was studied immunohistochemically in normal lung tissues and 107 lung tumors of various types. In normal tissues, vitamin B12 R-binder (R-binder) expression was restricted to the mucous cells of bronchial or bronchiolar epithelium and submucosal glands as well as to nonciliated bronchiolar (Clara) cells. Among lung carcinomas, 38% of squamous cell carcinomas, 42% of adenocarcinomas and 23% of large cell carcinomas showed positive staining for R-binder whereas small cell carcinomas did not. These findings offer the possibility that a majority of the histologic types of lung carcinoma have common histogenetical characteristics with mucous or Clara cells. Of the bronchial gland tumors, R-binder could be detected in a mucoepidermoid carcinoma but not in adenoid cystic carcinomas. Epithelial components in both pulmonary blastomas and hamartomas showed a reactivity for R-binder, suggesting that these tumors contained components composed of cells with bronchiolar cell differentiation. The immunohistochemical examination of lung tumors, using anti-R-binder antibody, may have some implications in the cell differentiation of lung tumors.

[Pharmacological actions of S-145, a novel thromboxane A2 antagonist, in various smooth muscles].

Antagonistic actions of S-145 ((+-)-5(Z)-7-[3-endo[(phenylsulfonyl)amino]bicyclo[2.2.1] hept-2-exo-yl]heptenoic acid) against U-46619, a thromboxane A2 mimic, were studied using isolated thoracic aorta of the rat and the trachea, lung parenchyma and ileum of the guinea pig. S-145 as well as SQ-29548 and ONO-3708 inhibited the contraction of aorta induced by U-46619 in a concentration-dependent manner. The IC50 value of each compound was 1.4, 14.5 and 52.6 nM. S-145 also inhibited contractions of the aorta induced by high concentrations of PGE1, PGE2 and PGF2 alpha, but failed to affect the responses to K+, Ca2+, NE, 5-HT, and angiotensin II. Contractions of trachea and lung parenchyma of the guinea pig induced by U-46619 were concentration-dependently inhibited by S-145, but those induced by histamine and leukotriene D4 were not affected. Ileac contractions by PGE2 and PGF2 alpha were not inhibited by S-145. The (+)-isomer of S-145 was more potent and the (-)-isomer was less potent than S-145 for antagonistic action against U-46619. These results suggest that S-145 is a potent and specific antagonist to the thromboxane A2 receptor; and in the aorta, the thromboxane A2 receptor may respond to high concentrations of PGs.

Electronic structure of DNA by DV-X alpha cluster calculations: II. d(GG).d(CC), d(CG)2, d(GC)2 A and B conformations. (Part 2) Sugars and bases.

The electronic structure of d(GG).d(CC), d(CG)2, d(GC)2 which are stacked base pairs in the DNA double helix, are elucidated for both A and B conformations in detail by DV-X alpha cluster calculations. These three DNA double helix fragments are contracted from the same bases, G and C, but the electronic structures of the fragments for both A and B conformations are different from each other characteristically. There are some delicate differences in the admixture of the orbital components and the overlap populations of intra- and inter- strand stacked bases among the stacking isomers. On the other hand, the electronic states of sugars differ in the 5'-3' direction, but are not almost dependent on stacked base pairs.

[Malignant lymphoma of testis: a clinicopathologic study of twenty-six cases].

A clinicopathologic study was made on 26 patients with testicular lymphoma. Because they all presented scrotal swelling as an initial manifestation, they were thought to be affected with primary testicular lymphoma. The age at admission ranged from 39 to 87 years, with a median of 63 years. Stage of disease was I E in 8 patients, II E in 7, and III E in 8. Two- and five-year survival rates were 38% and 26%, respectively. The most common histological type was the diffuse large cell type, especially immunoblastic type. Immunohistochemistry revealed that almost all cases were B cell derived tumors. In our study, an obvious correlation between the histological type and the prognosis for patients could not be observed. However, the initial stage of the disease appeared to affect the outcome in patients with testicular lymphoma.

[Single-agent trial with epirubicin in hematological malignancies].

Twelve patients with hematological malignancies were treated with epirubicin and ten patients were evaluable. One out of our four patients with ALL, who had a previous therapy of anthracycline, achieved a partial remission (PR: 25%). In two patients with AML, remission was not obtained. Of four patients with NHL, one with B-cell lymphoma achieved complete remission (CR) and one with ATLL partial remission (CR + PR: 50%). Stomatitis was observed as a major side effect in three patients with acute leukemia and in one with NHL. In conclusion, our trial seems to show the efficacy of epirubicin in lymphoid malignancies.
