Cells expressing HLA molecules in the B15 family were identified by serologic typing in routine testing of volunteer donors of various ethnic backgrounds for a bone marrow registry. DNA sequencing was used to identify HLA-B15 alleles associated with each serologic type and to examine the diversity within the B15 antigen family. Alleles which appeared predominantly in each B15 serologic cluster included: B15 with no defined serologic subdivision (B*1501), B62 (B*1501), B63 (B*1516, B*1517), B75 (B*1502, B*1521), and B76/77 (B*1513). Other B*15 alleles were also found associated with the serotypes and some of these alleles (e.g., B*1501 and B*1516) were found in two or more serologic clusters illustrating the complexity of this family. The B15 unsplit and B75 groups were the most complex exhibiting 16 and 7 alleles, respectively, within each serotype. Five new B*15 alleles (B*1530, B*1531, B*1533, B*1534, B*1535) and 5 other new HLA-B alleles (B*38022, B*3910, B*4010, B*51012, and B*5108) were also identified.
HBs antigen (HBsAg) incidence and subtype ratio were studied in apparently healthy HBsAg carriers and correlated with the ABO blood group of the subjects investigated. The incidence of subjects belonging to blood group AB was higher among the healthy carriers than in the unselected normal population, and the ay : ad ratio was much lower in this blood group than in the other ones.
A study was conducted on the specific and non specific immunomodulator effects of the inactivated vaccine NIVGRIP. The results revealed the seroconversion of HAI antibodies against influenza and mumps viruses and the reduction of viral and microbial flora of the pharynx. The kinetics of some proteins specific to the acute stage (siderophiline antitrypsin, microglobulin, complement C1q and C2 factors, C reactive protein and fibronectin) was also investigated and significant quantitative modifications were noticed in vaccinated subjects. The mean illness duration (acute respiratory disease) was three times as short in vaccinated subjects as in controls.
A study was performed on 756 serum samples collected from the umbilical cord of newborn infants. HBV markers were found in 4.63% of the tested subjects. Of these samples, 291 were tested for the HIV markers and 0.34% were found positive (versus 0 in the controls, most of them men from the same region). Results confirmed the possibility of vertical transmission of both viruses, HBV and HIV, in Romania.
Investigations pointed out the high level of respiratory diseases and pneumonias in children, especially in the youngest ones (0-1 year-old) during all the surveyed period. An influenza outbreak during the second half of February was registered in the infantile population--school and low-age children--which gave most of the cases. Serology and isolations incriminated an influenza virus related to the A/Beijing 353/89 (H3N2) strain. Isolations worked out after the influenza epidemics pointed out circulation of parainfluenza and adenoviruses. The necessity is emphasized of the anti-influenza vaccination of low-age children.
The presence of some viral and inframicrobial antigens in peripheral leukocytes was investigated by the indirect immunofluorescence technique in 120 patients with different forms of rheumatism and 50 clinically healthy controls. Mycoplasma pneumoniae and type 3 para-influenza virus were detected most frequently. The determination of serum interferon titer revealed a rise of this product in rheumatic patients.
An acute respiratory disease outbreak appeared during February and March 1988 in a half-closed community. Virologic investigations were conducted and an influenza virus type B strain was isolated, antigenically like B/Victoria 2/87. Administration of the anti-influenza vaccine in the fifth day of the outbreak evolution led to considerable reduction of the new cases and to outbreak extinction after 48 hours.
The study of the kinetics of interferon in the nasopharyngeal secretions (NPS) of adults vaccinated by intranasal route with the NIVGRIP inactivated influenza vaccine and of unvaccinated controls led to the following observations: the large variety of the individual reactions to the IFN inducing action of the NIVGRIPR vaccine: the presence of low titers of a "physiological" IFN in the NPS of 26% of the patients before vaccination; a positive conversion to IFN in 60% of the patients after the first and in 33% after the second administration of the vaccine. No changes were noted in the percentage of IFN positive controls during the observation period. A "physiological" IFN was detected in the NPS of 10% of the controls.
The circulation of respiratory viruses was investigated comparatively in a group of 24 subjects vaccinated intranasally with the NIVGRIP inactivated influenza vaccine and in 20 unvaccinated controls. The study relied on the detection by immunofluorescence reactions of viral antigens in exfoliated cells present in the nasopharyngeal secretions collected in January, February and March 1984. In January-February the prevalence of influenza, parainfluenza and adenovirus type 5 antigens was considerably lower in vaccinees than in controls. Simultaneous detection of more than two viral antigens in the same subject was more frequent in controls. The data demonstrate that the influenza vaccine administered not only conferred specific protection, but also stimulated the mechanisms of local nonspecific antiviral defence.
A comparative study was conducted in patients vaccinated with the NIVGRIP trivalent inactivated influenza vaccine and in placebo receiving controls on the kinetics of the serum hemagglutination inhibiting (HAI) antibodies and the neutralizing secretory antibodies in the nasopharyngeal secretions (NPS), of the blastic transformation of lymphocytes index, of the rosette formation index and of the serum immunoglobulins. A significant rise of the H.A.I. and the neutralizing secretory antibodies as well as of the blastic transformation of lymphocytes index was recorded after stimulation with the influenza vaccine. There were no significant changes in controls. No significant variations of the blastic transformation of lymphocytes index after stimulation with P.P.D. and of the rosette formation index were recorded in both investigated groups. Serum immunoglobulin titres showed significant variations in vaccinated as well as in control groups. The results point out the stimulating effect of the NIVGRIP inactivated influenza vaccine on both humoral and cell mediated immune responses.
The anti-HBc response was investigated by immunofluorescence in the sera of 109 asymptomatic HBsAg carriers. The results suggest the presence of at least two antigenic HBcAg determinants, possibly correlated with the replication phase. The subjects with intensive anti-HBc response showed immunological alterations consistent with the presence of active liver lesions and of hepatocellular membrane injury.
The trivalent inactivated NIVGRIP influenza vaccine was administered by nasal route to 26 young adults; a matched control group was represented by 19 subjects who received a placebo. Vaccination was followed by a higher proportion of rises in the level of neutralizing secretory antibodies (70-77%) than in that of serum HAI antibodies (19-42%) to the three influenza strains contained in the vaccine. Simultaneous rises in secretory antibody titers to three and two influenza antigens occurred in 50% and 23% of the vaccinees, respectively. The vaccine conferred a protection rate of 75%, as estimated by a 7-month clinical follow-up.
An outbreak of 111 cases of acute respiratory tract infection was recorded in a community of the town "T" in April-May 1984. The clinical picture was severer than usual; 28% of the cases had to be hospitalized, average absenteeism being as high as 26 days per case. Serological investigations demonstrated the previous circulation of influenza virus B/Singapore/222/79 and the simultaneous circulation during the outbreak of influenza virus A/England/333/80 (H1N1) and of Rickettsia burneti (as also ascertained by isolation in the chick embryo of the former and by visualization by immunofluorescence in exfoliated cells of the latter pathogen). The association of the two etiological agents appears to account for the severe and protracted course of the disease.

The results obtained by subtyping and titration of 872 HBsAg from apparently healthy carriers were grouped according to blood group and sex. The incidence of blood group AB is higher among healthy HBsAg carriers than in the unselected population. There are large variations as regards the ay : ad ratio (overall mean value : 4.74) : minimal ay : ad values occur in the AB blood group, both in men (4.00) and in women (1.33), as compared with the mean values of 5.07 and 2.95 recorded in men and women, respectively. Titer analysis shows a cluster of higher titers in women. The involvement of immunogenetic mechanisms in asymptomatic HBsAg carriage is suggested.
