Development of a model system for evaluation of human endothelial cell proliferation in vitro: possible clinical applications.

A system for evaluation of the ability of human blood serum to affect endothelial cell proliferation was developed and tested. The system based on incorporation of (3)H-thymidine into DNA was used to analyze the effects of hormone replacement therapy on endothelial repair and angiogenesis. Blood serum from 12 menopausal women less effectively activated endothelial proliferation compared to control donor serum. After 6-month hormone replacement therapy with Divina (a combination of estradiol and medroxyprogesterone), this index increased in seven female subjects (58.3%), but remained below the control level. The model proposed by us can be used in clinical practice and drug testing for evaluation of the influence of blood serum on vascular endothelium.

[Prognostic value of blood indices in children with acute appendicitis].

The results of comparative analysis of blood formula, volume and cellular content of peritoneal effusion in children operated on for acute appendicitis are presented. In unfavourable course of the postoperative period, a significant decrease in content of neutrophil granulocytes in the peripheric blood and its increase in the peritoneal effusion as compared with the indices in the control group was noted.

[Dynamic of lectin activity during germination of bean seeds (Phaseolus vulgaris L.)].

PHA quantity and activity dynamics during early germination of bean seed were investigated. Electrophoretic characteristics, subunits composition and carbohydrate-binding specificity of lectin extracted from white kidney bean cv. Bilozerna were studied. It was shown that investigated lectin consisted of 2 subunits E and L with molecular weight 34 and 36 kDa, respectively, analogously to purified PHA ("Serva", Germany), and specifically bound N-acetyl-D-galactosamin and galactose. During germination both quantity and activity of PHA were dramatically decreasing in embryonic axes and in cotyledons, possibly, as a result of the lectin release from seeds to the environment. It is very likely that one of the defence mechanisms of germinating seeds is related with the releasing of lectins that are able to bind components of the bacterial cell wall and to inhibit their growth.

[Improving of methods of production and screening of monoclonal antibodies to Salmonella antigens].

Methods for the production of and screening for monoclonal antibodies (MAB) to Salmonella antigens have been optimized. Schemes for immunization of animals have been developed for various Salmonella antigens. Prolonged (up to 6 months) immunization is needed to obtain antibodies in high titers to 0-antigen, short (up to 1 month) to obtain antigens to K-antigen. Affinity purification is desirable if MAB to bacterial antigens from ascitic fluid are used. Study of the kinetics of MAB binding to antigens helps appreciably accelerate enzyme immunoassay. Microplates manufactured by Medpolimer National Research Institute (St. Petersburg) may be recommended as the solid phase. Glutaraldehyde treatment of the plates to improve their adsorption capacity is not recommended.
