Effect of complex phytoadaptogen on MPTP-induced Parkinson's syndrome in mice.

Oral administration of 10% solution of Phytomix-40 (multicomponent plant phytoadaptogen) to C57Bl/6 mice with MPTP-induced Parkinson's syndrome alleviated symptoms (oligokinesia and muscle rigidity), compensated for the deficiency of dopamine and its metabolites (DOPAC and homovanillic acid), and reduced the level of lipid peroxides in the striatum. In vitro Phytomix-40 in a concentration of 3.3 x 10(-2) g/liter exhibited a pronounced antioxidant effect (5-fold decreased MDA level in mouse brain homogenate in Fe(2+)-ascorbate-dependent LPO).

Leukoplakia of oral mucosa: pathogenesis and possible correction with phytoadaptogen.

Leukoplakia of the oral mucosa is characterized by impairment of the epithelial differentiation program. The use of complex phytoadaptogen in the treatment of patients with leukoplakia normalized expression of Fas-APO-1 antigen and keratin 17; increased expression of CD54 attested to activation of immune effectors. The clinical effect manifested in involution or shrinkage and loosening of the pathological focus and complete epithelialization of erosive surfaces. The phytopreparation exhibited adaptogenic effects normalizing the immune and IFN status and improving the state of adaptation systems in the body.

[Improving the efficiency of combined therapy of patients with leukoplakia of the buccal mucosa using Phytomix-40 combined plant preparation].

Today leukoplakia of the buccal mucosa is regarded as a typical pre-tumorous condition, both in medicine in general and in dentistry, in particular. Leukoplakia is in fact resistant to conservative treatment, and therefore search for new approaches to therapy of this disease is a pressing problem. Phytomix-40 (PM-40) is a mixture of natural extracts of 40 medicinal plants developed for practical oncology. PM was used in combined therapy of patients with leukoplakia with good results.

Phytoadaptogen for preventive oncology: immunobiological criteria of composition.

A system of in vitro immunobiological tests is developed for screening of phytopreparations intended for the use as immunomodulators in oncology. Proliferative activity of human tumor cells decreased after treatment with complex phytoadaptogen. The immunomodulatory effect of this phytoadaptogen on immunocompetent cells of cancer patients and its nonspecific interferonogenic effect were detected. The composition of adaptogenic complex for preventive oncology is determined.

A method for biological control of a complex phytoadaptogen.

We propose a method for standardization of complex adaptogen-containing preparations. The method is based on acceleration of baking yeast strain growth on energy-depleted medium in the presence of the test agent. This method allows simple quantitative biological control of phytoadaptogens and comparison of adaptogenic activity of mono- and complex preparations.

[Adhesive cell interactions in the biology of cancer].

The present review describes a hypothesis for a critical role of cell adhesive interactions in tumorigenesis. Dysregulation of tissue cell-cell interactions initiates first of all local (in the tissue) and then general (in whole body) conditions for tumor growth. Otherwise imbalance of tissue-specific adhesion factor at the very beginning of carcinogenesis is considered to trigger a cascade of pathological reactions responsible for more severe adhesive disorders that are in turn critical for the "totalitarian" behavior of a tumor and its "colonization" of other tissues and organs. Impaired disturbance is likely to be the key mechanism of carcinogenesis since it is significantly associated with the main features of a tumor: tissue proliferation control loss, anaplasia, invasion, metastasis, and immune surveillance deficit. The hypothesis is supported by evolutionary, biological, histological, immunological, and clinical arguments whose combination does not characterize any other known mechanisms of oncogenesis. The concept of adhesiveness opens new possibilities for the diagnosis, prevention, and treatment of tumors and also improves a strategy for designing new drugs.

[Adaptogens as agents for prophylactic oncology].

The paper considers whether it is advisable to use the new acting preparations adaptogens, including phytoadaptogens, in oncological care for prevention of cancer. The adaptogens are shown to have a regulating action, to be able to activate the protective properties of the body, to protect it from extreme exposures and to stimulate regenerative processes. The author associates the antitumor effect of adaptogens with the immunomodulating (they can activate macrophages, natural killer cells, antigen-dependent T lymphocytes) and interferonogenic actions and with the their ability to suppress experimental tumor growth, to enhance tissue differentiation, to improve intercellular adhesion, to reduce the likelihood of metastasis spreading. Furthermore, the use of adaptogens, and phytoadaptogens in particular, decreases the toxic effects of chemotherapy and improves drug tolerance.

[Testing of plant extracts preparations for prevention and non-toxic therapy of oncologic diseases in experimental models].

The extracts from Panax ginseng and Rhodiolae rosea from natural roots, from cultural callus tissue and from Ganoderma mushroom have been studied for adaptogene, T-immunogene activities and for stimulation of mechanical integration in tissues predisposed to hereditary tumors. The least activity has been shown by extracts from Ganoderma M., that from Rhodiolae rosea natural root has turned out to the most effective. This study can serve as the basis for further investigations by using the extracts on inbred mice and in the clinical setting for the prevention and intoxic therapy of tumors.

[Preventive effect of Rhodiolae rosea in spontaneous liver carcinogenesis in a mice model of high-tumor strain].

Different schemes of Rhodiolae rosea usage per os for prophylaxis of mouse hereditary hepatomas have been studied. A long-term effect on stimulation of tissue integration, T-immune activity and sufficient decrease of tumour frequency has been shown for R. rosea usage in early ontogenesis. A short-term effect on tissue and immune parameters has been demonstrated for R. rosea usage in middle ontogenesis. High-tumour frequency (as in the control), but tumour size decrease (comparing to the control) has been noticed in this case. A tendency to synchronic alterations of tissue integration and T-immune activity has been established in these experiments. It can be attributed to the assumption of communication between tissue (local) and immune (common) defence from tumour growth in the organism.

[Mechanical properties of the intercellular contacts of hepatocytes of inbred strains of mice and mice predisposed to spontaneous hepatomas].

The adhesiveness of hepatocytes was evaluated in 7 inbred mice and compared to the reported data on the incidence of spontaneous hepatomas. The adhesiveness was shown to be significantly less in mice predisposed to spontaneous hepatomas as compared to more resistant species. Electron microscopy revealed the presence of little and highly adhesive areas (HAA) in nonspecialized regions of hepatocytic contacts, this presence being shown by elevated (20-30 mm Hg) hydrostatic pressure in the liver blood bed. The amount and mechanical strength of HAA was less in CBA mice predisposed to spontaneous hepatomas as compared to resistant C57B1 mice, and correlated with cell adhesiveness.

[A toxicological study of the preparation Phytomix-40].

The results of toxicological evaluation of the new plant preparation Phitomix-40 (PM-40) used in the treatment of cancer are presented. To prevent a patient's individual immunological responsiveness to some phytoadaptogens, the preparation comprises 40 plant extracts (Rhodiola r. L.; Juniperus C. L.; Helichrysum a. L; Viburnum o. L., etc.). Animal experiments demonstrated that PM-40 had a low toxicity; LD50 was more than 20 and 15 ml/kg for rats and mice, respectively.

[Changes in the intercellular contacts of hepatocytes in the ontogeny of inbred mouse strains with a high (CBA) and low (C57BL) frequency of spontaneous hepatomas].

Some parameters of cellular contacts between hepatocytes were studied in ontogenesis of the inbred mouse lines with the high (CBA) and low (C57BL) frequency of spontaneous hepatomas. In both the lines the force of intercellular linkage of the hepatocytes drops down by the end of the animal's life. But in the C57BL mice this characteristics increases sharply between the 5th and 10th days after birth, whereas in the CBA mice no such increase is observed. During the whole postnatal development the force of linkage in the CBA mouse liver is thrice lower than in the C57BL mouse liver. The stability of contacts between the hepatocytes in the C57BL mice is not affected during ontogenesis, as well as in the CBA mice, except in the males at the age of 15-24 months when the stability of contacts decreases. This parameter decreases still more in the tumour. Since the CBA males are characterized by a higher frequency of spontaneous hepatomas, a suggestion is put forward to the effect that the primary disturbance of the stability of contacts reflects the early changes characteristic for the spontaneous liver carcinogenesis.

[The effect of a Rhodiola rosea extract on the incidence of recurrences of a superficial bladder cancer (experimental clinical research)].

Oral administration of Rhodiola rosea extract to a small group of patents (n = 12) with superficial bladder carcinoma (T1G1-2) improved the characteristics of the urothelial tissue integration, parameters of leukocyte integrins and T-cell immunity. The average frequency of relapses for these patients has been found to fall twice, though statistical differences were not significant.

[The prognostic value of flow cytometry in prostatic cancer].

DNA quantitation by flow cytometry was made in 36 patients with morphologically diagnosed prostatic cancer (PC) in stage II, III and IV (4, 14 and 18 patients, respectively). All the patients received adequate treatment. Aneuploid tumours were found in 25 patients (group 1), diploid ones in 11 patients (group 2). A 3-year survival in group 1 made up 65.5%, in group 2 all the patients survived 3 years. The recurrence-free survival was 22.2% and 78.8%, respectively. The above findings make it possible to regard flow cytometry DNA estimation as a reliable prognostic indication in PC.
