[Induced hematopoietic foci in mice. I. Induction of extraskeletal hematopoietic areas using demineralized tooth matrix].

Conditions of formation of induced osteo-hemopoietic foci in mice, in response to ectopic implantation to them of homologous demineralized tooth matrix, have been studied. It is shown that the hemopoietic tissue of the induced foci is similar to the bone marrow of the skeletal bones by the morphological parameters and cellular composition; differentiation of precursor cells takes place in all three directions of myelopoiesis. Induced foci formation under the capsule of the kidney occurs much later (after 5-8 months) than under the skin of muscular fascia (1-1.5 months). The induced hemopoietic foci functioning continues at least for one year.

[Renewal of hematopoietic stem cell clones in long-term bone marrow cultures].

In long-term cultures of murine bone marrow, clonal succession of hemopoietic cells was observed as measured by karyologic analysis. There were high oscillations in self-renewal of CFUs in the cultures. A close correlation between the CFUmix karyotype and mitotic non-adherent cells in culture (but not between these cell types and CFUs) was revealed.

[Hybrid resistance in a long-term bone marrow culture and in foci of ectopic hematopoiesis formed in the culture].

The lack of hybrid resistance to the bone marrow graft has been demonstrated in a long-term bone marrow culture. After adherent cell layer transplantation into the body the hybrid resistance was demonstrated in de novo formed ectopic hemopoietic foci. The resistance was of the recipient type, because of which the existence of the migrating component of the hemopoietic microenvironment is suggested.

[Possibility of using hematopoietic stem cells in relation to their generation age].

A hypothesis of the use of hemopoietic stem cells with a view of satisfying the demand for mature cells depending on their generation age predicts that CFUs that survived after repeated treatment with hydroxyurea should have a greater capacity for self-renewal. It has been demonstrated that after repeated administrations of hydroxyurea according to the scheme devised by the authors of the hypothesis (3-4 times) and as a result of a more prolonged treatment (6 times, every other 12 or 15 h), the capacity of the survived hemopoietic stem cells for self maintenance was not only lower than normal but commonly significantly decreased. The generation-age hypothesis of the use of hemopoietic stem cells thus remains badly needing experimental support.

[Separation of mouse bone marrow cells by preparative electrophoresis].

Free-flow electrophoresis was used to separate mouse bone marrow cells. From five to eight fractions with an amount of cells sufficient for morphofunctional analysis could be obtained in different experiments. Granulocyte precursors were found in fractions with low electrophoretic mobility (EPM), whereas erythrocyte precursors in fractions with high EPM. CFUs were detected both in low and in high EPM fractions. In individual fractions the concentration of these cells was 4 times greater than in the initial cell suspension.

[Importance of cell contacts for the differentiation of the precursor cells of hematopoietic stroma in long-term bone marrow cultures].

The composite adherent cell layer is produced in the culture of bone marrow fragments. On the contrary, in the culture of single cell suspension of bone marrow, only of monolayer of fibroblast-like cells and macrophages is created. After implantation of the adherent cell layer from the Dexter type culture but not from "fibroblast" culture beneath the renal capsule of syngeneic recipients the ectopic hemopoietic sites are formed. Regeneration of the adherent cell layer may occur only in the course of multilayer formation, i.e. one week after bone marrow transplantation but not three weeks after the formation of the adherent cell layer.
