Reduced levels of erythrocyte sorbitol dehydrogenase activity in human diabetics.

Erythrocyte sorbitol dehydrogenase activity was significantly lower for 34 diabetic patients (mean +/- SD: 0.219 +/- 0.085 U min-1 g Hb-1) than for 27 normal blood donors (0.34 +/- 0.094 U min-1 g Hb-1). This finding agrees with the previous report of low sorbitol dehydrogenase activity in the lens of diabetic patients but contrasts with the data of Crabbe and collaborators (M.J.C. Crabbe, A.B. Halder, C.O. Peckar and H. Cheng, Lancet, 2: 1268-1270, 1980) who found a large variation in the levels of the diabetic population and which was not significantly different from the normal control.

Biochemical characteristics of erythrocyte sorbitol dehydrogenase from selected mammals.

The erythrocyte sorbitol dehydrogenase (EC 1.1.1.14) activity, regarding its action on sorbitol oxidation to fructose, was studied in 19 species of mammals, showing a striking variability, with high activity in rodents. Enzyme activity was studied against other polyols, namely xylitol, inositol, manitol and dulcitol. Most animals showed activity against all the polyols studied, but hamster and red deer only presented activity on sorbitol and xylitol. Michaelis-Menten constant determinations for sorbitol were performed, and it was observed that animals which presented high activity had a high Km. pH curves were obtained from 8 animals, with an optimum pH ranging from pH 8.0 to pH 10.0; four of the animals presented an optimum pH at 8.5.

Starch gel electrophoresis of erythrocyte hypoxanthine-guanine phosphoribosyl transferase and adenine phosphoribosyl transferase: a population survey.

Vertical starch gel electrophoresis of hypoxanthine-guanine phosphoribosyl transferase (HGPRT) and adenine phosphoribosyl transferase (APRT) was performed in several population groups including Caucasians, Negroes and Orientals. The red cells of a total of 244 individuals were examined for HGPRT and those of 265 for APRT and no variants were detected.

The sorbitol-oxidizing enzyme of red blood cells.

The sorbitol-oxidizing enzyme of human erythrocytes has been partially purified and characterized. Its kinetic characteristics were found to be similar to the sorbitol dehydrogenase of sheep liver and the DPN-xyluloxe dehydrogenase which have been isolated from sheep liver and guinea pig liver, respectively. Although this enzyme is able to oxidize sorbitol relatively efficiently, it has very little activity toward dulcitol, and therefore cannot account for the high K-m pathway of methemoglobin reduction by galactose.

Methemoglobinemia associated with loxoscelism.

In twenty five patients who presented the cutaneous form of loxoscelism, serum haptoglobin and lactic dehydrogenase, erythrocyte glucose-6-phosphate dehydrogenase, glutathione reductase, glutathione peroxidase, methemoglobin, bilirubin and reticulocytes were investigated after bite. No hemolysis was detected but an increase in methemoglobin was found in 54% of the cases; in 7% it was between 1.1% and 2%, in 27% it ranged from 2.1% to 4%, and in 20% from 4.1% to 8%. Blood samples of a normal, blood group 0 individual and of a patient who exhibited methemoglobinemia after Loxosceles bite were incubated separately with antisera against Loxosceles gaucho, Crotalus terrificus, Bothrops jararaca, with Loxosceles gaucho venom and 0.3% phenol. No methemoglobin was found after 1, 4, 8 and 15 days in both sets of samples. At the 25th day all the samples, including the controls, exhibited similar methemoglobin reductase decrease. The data suggest that the methemoglobinemia which occurs in 50% of the patients probably arises from in vivo venom metabolism, inasmuch as the crude venom does not induce methemoglobinemia.

Search of intravascular hemolysis in patients with the cutaneous form of loxoscelism.

Haptoglobin assay, a highly sensitive method to detect intravascular hemolysis was carried out in the sera of 19 patients referred to Hospital Vital Brazil with the cutaneous form of loxoscelism in order to investigate the occurrence of mild intravascular hemolysis. Data from this series did not show decreased levels haptoglobin, ruling out intravascular hemolysis in these patients with cutaneous form of loxoscelism.

[Helminth and protozoan intestinal infections in pregnant women in their first consultation at Health Centers of the State in the Butantã Subdistrict, São Paulo City].

481 pregnant women attended at first consultation from a São Paulo city suburb-subdistrict of Butantan-São Paulo State, Brazil, from April to October of 1988, were investigated regarding the prevalence of verminosis. The mean age was 24.5 years (range 14-46), the average family income was 0.97 PCMW (per capita minimum wage) and the average number of members of the family was 4 (1 to 15). The verminosis prevalence was 45.1% among the women. The most frequent parasites were: Ascaris lumbricoides (19.0%), ancilostomídeos (16.7%) and Trichuris trichiura (15.9%). The verminosis prevalence was significantly higher among the pregnant women (p < 0.05) who belonged to the families with income below 0.5 PCMW and greater than 4 members.
