HID-AB2.5-PAS and HID-PAS methods for the histochemical analyses of a variety of carbohydrates.

The high iron diamine (HID) staining procedure was combined with alcian blue pH 2.5 (AB2.5) and periodic acid-Schiff (PAS) methods for the simultaneous demonstration of carbohydrates containing sulfate esters, carboxyl groups and oxidizable vicinal diols, whereas it was sequentially employed with PAS alone to differentiate sulfate esters from oxidizable vicinal diols. A variety of rat tissues and the epidermis of four fish species were utilized to test the specificity or selectivity of these methods. The HID-AB2.5-PAS sequence largely coloured carbohydrates containing sulfate esters, carboxyl groups and oxidizable vicinal diols in brownish black, turquoise and magenta shades respectively. The HID-PAS sequence coloured sulfate esters brownish black and oxidizable vicinal diols magenta. On the basis of the experimental and control studies on the both staining techniques, these staining methods could be postulated to represent efficient and useful techniques for precise histochemical analyses and simultaneous differentiations of a variety of carbohydrates in light microscopy.

Effects of iodine pretreatment of sections upon immunogold-silver staining in light microscopic immunohistochemistry.

Effects of Lugol's iodine pretreatment upon immunogold-silver staining were examined in terms of the intensification of the reaction products obtained by physical development. Iodine pretreatment has diminished the duration necessary for the development, whereas the staining reaction of an appropriate intensity, could hardly be obtained by the pretreatment. In addition, essential staining reactions tended to be accompanied by marked nonspecific silver precipitations. It is concluded that iodine pretreatment should either be omitted or used with cautions in immunogold-silver staining procedures for the precise detection of antigenic sites.

The use of periodic acid-thiocarbohydrazide-silver protein-physical development (PA-TCH-SP-PD) procedure for the histochemical detection of neutral carbohydrates in the circumventricular organs of the rat.

A periodic acid-thiocarbohydrazide-silver protein-physical development (PA-TCH-SP-PD) procedure was applied to the rat circumventricular organs (CVOs), which are known to be specialized structures in the brain. In the rat CVOs, vivid PA-TCH-SP-PD reactions were obtained in the apical surface and cytoplasm of the ependymal cells of the subcommissural organ (SCO) and the epithelial cells in the choroid plexuses (CPs) examined, and similar positive reactions were detected in the vascular walls and perivascular connective tissues in all the CVOs tested. Further, varying intensities of PA-TCH-SP-PD reactions were noted in the neuronal and glial networks of the organum vasculosum laminae terminalis (OVLT), subfornical organ (SFO) and area postrema (AP). The results obtained in the present study indicate that in the rat CVOs the histologic structures mentioned contain varying amounts of neutral carbohydrates and possible histophysiological significances of these carbohydrates in these organs have been discussed with references to their functions.

A dual staining method for neutral carbohydrates and DNA (deoxyribonucleic acid) using periodic acid-thiocarbohydrazide-silver protein-physical development (PA-TCH-SP-PD) and Feulgen (Hot HCl-Schiff) techniques.

A dual staining method was established for the histochemical detection of neutral carbohydrates and deoxyribonucleic acid (DNA) in light microscopy. The method consisted of combined periodic acid-thiocarbohydrazide-silver protein-physical development (PA-TCH-SP-PD) and hot hydrochloric acid (H-HCl)-Schiff (Feulgen) procedures, which produced reaction products of blackish and magenta shades respectively. The present dual staining method is believed to be unusually useful for the light microscopic histochemical studies on neutral carbohydrate-containing cells, since it exhibits a high contrast between these two shades and reveals distinctly the localization of cytoplasmic neutral carbohydrates and nuclear DNA in one and the same tissue section.

Lectin-binding glycoconjugates in the tongues of the rat and guinea pig as revealed by lectin-gold-silver methods.

Lectin-gold-silver (LT-G-S) procedures using two lectins (RCA-I and Con A) were applied to appropriately prepared paraffin sections of tongues in the rat and guinea pig. In the lingual mucous membranes of the rat and guinea pig, positive RCA-I-G-S and Con A-G-S reactions were obtained in the intercellular spaces and the cytoplasm of epithelial cells of the basal and intermediate layers respectively. Likewise, the LT (RCA-I and Con A)-G-S techniques gave rise to varying intensities of positive reactions in the serous and mucous gland acini, nerve and muscle fiber bundles and connective tissue elements. The results obtained in the present study indicate that in the rat and guinea pig tongues the histologic structures mentioned contain varying amounts of beta-D-galactose and alpha-D-mannose or alpha-D-glucose residues of glycoconjugates, and that such histochemical properties of some of the lingual structures can be correlated with their possible histophysiological functions.

A double protein A-gold-silver staining method for tissue antigens in light microscopy.

A double staining method has been developed for light microscopic immunohistochemistry. The technique consists of a sequence of protein A-gold-silver procedures, in which the reaction products are visualized in black and brown colours. The results obtained in the rat pancreatic islets and adenohyophysis, in combination with a variety of histochemical controls, substantiate both the specificity and reliability of this method. The double staining method is simple and economical, since the two differently coloured reaction products are obtained by a common mechanism of colouration (physical development) using a single probe, protein A-gold.

A histochemical method for the demonstration of unsaturated lipids.

A method has been developed for the histochemical demonstration of unsaturated lipids in light microscopy. It is a peracetic acid-thiocarbohydrazide-silver protein sequence followed by a physical development procedure. In the present study on paraffin and cryostat sections of liver, brain and ovary, unsaturated lipids were visualized as distinct reaction products coloured various shades of brown and black. The reaction products are easier to see and the method is more efficient than the peracetic acid-Schiff method.

Human scalp hair as evidence of individual dosage history of haloperidol: a possible linkage of haloperidol excretion into hair with hair pigment.

We report a method for determining haloperidol concentration in human scalp hair and discuss a possible linkage of haloperidol excretion into hair with the hair pigment melanin. First, an animal study was conducted to support the idea that hair contains amounts of haloperidol corresponding to the doses given and pigmented hair contains much more drug than does unpigmented hair. The haloperidol concentration was measured using a radioimmunoassay technique after hairs were dissolved in 2.5 N NaOH solution and the drug extracted. Pigmented and albino rats, whose hair from an area on the back had been removed beforehand by plucking, were administered either 1, 3, or 10 mg of haloperidol (i.p.) per kg body weight every day for 3 weeks. At the end of the administration period hair which had newly grown on the denuded area was plucked and collected. In each of the two groups classified by hair color the drug levels in the hair correlated with the doses given; however, the concentrations in the hair from the albino rats were much lower than those in the hair from the pigmented rats (which was less than 8.5%). Second, black and white hair was collected from each of seven human subjects with grizzled hair, who were receiving or had been administered haloperidol at fixed daily doses for more than 1 month, and the concentration of haloperidol in each type of hair was measured. In the same subject the concentration in the white hair was found to be much lower than that in the black (less than 10%).(ABSTRACT TRUNCATED AT 250 WORDS)

An efficient histochemical method for deoxyribonucleic acids using a silver enhancement procedure.

An efficient histochemical method has been worked out for the demonstration of deoxyribonucleic acids (DNA) with light microscopy. This involves a hot hydrochloric acid-thiocarbohydrazide-silver proteinate sequence followed by a physical development procedure. When applied to tissue sections from rabbit organs such as the spleen, cerebellum and testis, the new method has given excellent discrimination of nuclear DNA in all cells examined. Thus, the method is higher in efficiency and visibility of reaction products than the traditional hot HCl-Schiff (Feulgen) method used hitherto. Its specificity is also high.

A method for histochemical demonstration of protein-bound amino groups by light microscopy.

We have established an efficient method for the histochemical demonstration of protein-bound amino groups by light microscopy, using a ninhydrin or alloxan-thiocarbohydrazide-silver proteinate (NHY or ALX-TCH-SP) sequence followed by a physical development (PD) procedure. As a result of the present experimental studies on Carnoy's solution-fixed paraffin sections of a series of rat tissues, including three types of major salivary glands, liver, pancreas, stomach, duodenum, jejunum, colon, kidney, prostate, and spleen, the sensitivity and specificity of the new method were found to be sufficient. In the tissues tested, protein-bound amino groups were visualized by distinct brownish or blackish reaction products. Comparisons of the particular method with the NHY or ALX-Schiff methods employed hitherto have substantiated the fact that the former method leads to apparently higher visibility of reaction products than the latter.

Histochemical demonstration of zinc in rat epididymis using a sulphide-silver method.

We studied the histochemical distribution of zinc in rat epididymis using a sulphide-silver method. In the supranuclear cytoplasm of the principal cells that line the epididymis of rats, varying amounts of sulphide-silver-reactive zinc were visualized. In adult mating rats, significant amounts of zinc were found in the proximal portion of the epididymis, whereas in non-mating, mature and immature young rats, this heavy metal was most prominent in the distal portion of this organ. In all of the rats studied, zinc was sparsely distributed in the intermediate portion of the epididymis. From these results, it can be assumed that the zinc present in the epithelial lining of rat epididymis plays an important role in the maturation of spermatozoa. The present results represent a useful contribution to our understanding of the functional morphology of rat epididymis.

Lectin binding sites of glycoproteins as revealed by lectin-gold-silver and lectin-peroxidase-diaminobenzidine methods.

For the precise histochemical detection of lectin binding sites of glycoproteins, the results obtained by lectin-gold-silver (LT-G-S) staining methods have been systematically compared with those revealed by alternative techniques of lectin-peroxidase-diaminobenzidine (LT-PO-DAB) reactions in a series of organs from different mammalian species. Ricinus communis agglutinin-I and concanavalin A were the lectins used in the present study. In the tissues subjected to the LT-G-S procedures, reactive tissue structures exhibited positive reactions of varying intensities of black. The results of control staining for the LT-G-S methods substantiated the view that the reaction products demonstrated the precise lectin binding sites of glycoproteins. The staining images obtained by the LT-PO-DAB techniques were not necessarily correlated precisely with those revealed by the LT-G-S procedures, and unavoidable background staining in pale brownish shades was noted in the majority of LT-G-S negative tissue structures. In view of these results, the LT-G-S staining methods employed in the present study are believed to be a reliable technique for the precise localization of saccharide residues of glycoproteins in light microscopy.

The cytochemistry of glycoconjugates in the planum nasolabial gland of the goat as studied by electron microscopic methods.

In the planum nasolabial glands of the goat, glycoconjugates of glandular and duct cells have been studied by means of a series of electron microscopic cytochemical methods. In the glandular cells glycoconjugates with vicinal diol groupings were present in secretory granules, certain elements of the Golgi complex, lysosome-like dense bodies, the surface coat of the plasma membrane, the majority of intracellular cytomembranes, glycogen particles and the basal lamina. In duct cells, glycoconjugates with the same properties were localized in similar ultrastructures, except for secretory granules, which were not detected in these cells. By lectin cytochemistry, glycoconjugates in glandular cell secretory granules contained a variety of saccharide residues such as alpha-D-mannose, alpha-D-glucose, N-acetyl-D-glucosamine and alpha-L-fucose. The cytochemical properties of the secretory glycoconjugates are discussed in relation to the physiological functions performed by the planum nasolabial glands in the goat.

Histochemistry of glycoconjugates in the goat nasolabial skin with special reference to eccrine glands.

The histochemistry of glycoconjugates in the nasolabial skin of the goat has been studied by means of a series of selected methods of light microscopy. The epidermis of the nasolabial skin was found to contain neutral and acid glycoconjugates with different saccharide residues. The secretory epithelial cells and secretory substances of the sebaceous glands contained primarily neutral glycoconjugates, whereas those of the apocrine glands involved largely strongly acidic and neutral glycoconjugates. In the epithelial cells and secretory substances of the nasolabial eccrine glands, glycoconjugates involved were characteristically strongly neutral but weakly acidic in nature. From the present results, the histophysiological significance of glycoconjugates in the particular histologic structures of the nasolabial skin has been discussed with special reference to the functions of this particular skin type in the goat.

Light and electron microscopic studies on capillaries of the goat cardiac muscle with special reference to the topographical relationship between the vessels and Purkinje fibers.

The capillaries of the cardiac muscle were investigated in the goat by means of light microscopy, transmission electron microscopy and scanning electron microscopy, and the following results were obtained. The capillaries of the working cardiac muscles were numerous, arranged mainly parallel to the long axis of muscle cells and formed dense elongated networks. On the contrary, those of the terminal Purkinje fibers were relatively few in number, oriented in various directions and formed loose and circularly meshed networks surrounding the fibers. Such findings were discussed in correlation with the physiology and functional morphology of various types of the cardiac muscle cells.

Protein A gold-silver staining method for light microscopic immunohistochemistry.

A new sensitive method has been established for light microscopic immunohistochemistry. It involves a protein A gold technique instead of immunoglobulin-gold method, followed by an improved procedure of physical development. In this procedure, two selected reagents were employed: bromohydroquinone, a more potent developing agent than hydroquinone, and specifically purified gum arabic solution. The results obtained by this method in the pancreatic islet tissues of the rat under a variety of histochemical control conditions have substantiated both the high specificity and fidelity of this method.

Light and electron microscopic studies on the cytological changes in the rat pancreatic islets induced by sodium diethyl dithiocarbamate (DEDTC) administration.

The effect of sodium diethyl dithiocarbamate ( DEDTC ) administration upon the structure of the pancreatic islet cells has been examined in the rat. In light microscopy, the aldehyde fuchsin stainability of the B cell granules was found to increase notably at 6 h and thereafter following the medication. In electron microscopy, the B cell granules with a porous core were shown to predominate over those with a homogeneous core containing paracrystalline structures between 30 min and 3 h following the medication. Between 6 and 12 h following medication, occasional B cells were seen to be provided with large secretory granules with a porous core. The results of the present study have been discussed with special reference to the chelating action of DEDTC .
