Vanadate interaction with the Na, K-ATPase. An assay of serum vanadate based on the displacement of (48V) vanadate from Na, K-ATPase.

Under optimum conditions for vanadate binding to Na, K-ATPase equilibrium binding date for a range of vanadate concentrations were compiled. Scatchard plots indicated an identical binding capacity for vanadate and ouabain with conventionally prepared Na, K-ATPase batches. A method for determination of vanadate in serum based upon the high affinity of vanadate for Na, K-ATPase is described. The method takes advantage of the shift in equilibrium binding of (48V) vanadate upon addition of an aliquot of vanadate-containing serum. It is shown that the interplay between vanadate and ouabain in vanadate-facilitated ouabain binding to Na, K-ATPase leads to an enzyme-vanadate-ouabain complex to which vanadate is rather firmly bound.

Atomic force microscope characterization of a resonating nanocantilever.

An atomic force microscope (AFM) is used as a nanometer-scale resolution tool for the characterization of the electromechanical behaviour of a resonant cantilever-based mass sensor. The cantilever is actuated electrostatically by applying DC and AC voltages from a driver electrode placed closely parallel to the cantilever. In order to minimize the interaction between AFM probe and the resonating transducer cantilever, the AFM is operated in a dynamic non-contact mode, using oscillation amplitudes corresponding to a low force regime. The dependence of the static cantilever deflection on DC voltage and of the oscillation amplitude on the frequency of the AC voltage is measured by this technique and the results are fitted by a simple non-linear electromechanical model.

Optimised cantilever biosensor with piezoresistive read-out.

We present a cantilever-based biochemical sensor with piezoresistive read-out which has been optimised for measuring surface stress. The resistors and the electrical wiring on the chip are encapsulated in low-pressure chemical vapor deposition (LPCVD) silicon nitride, so that the chip is well suited for operation in liquids. The wiring is titanium silicide which-in contrast to conventional metal wiring-is compatible with the high-temperature LPCVD coating process.

AFM lithography of aluminum for fabrication of nanomechanical systems.

Nanolithography by local anodic oxidation of surfaces using atomic force microscopy (AFM) has proven to be more reproducible when using dynamic, non-contact mode. Hereby, the tip/sample interaction forces are reduced dramatically compared to contact mode, and thus tip wear is greatly reduced. Anodic oxidation of Al can be used for fabricating nanomechanical systems, by using the Al oxide as a highly selective dry etching mask. In our experiments, areas as large as 2 micro m x 3 micro m have been oxidized repeatedly without any sign of tip-wear. Furthermore, line widths down to 10nm have been routinely obtained, by optimization of AFM parameters, such as tip/sample distance, voltage and scan speed. Finally, AFM oxidation experiments have been performed on CMOS processed chips, demonstrating the first steps of fabricating fully functional nanomechanical devices.

[My fingers itch, but my hands are bound. An exploratory psychosomatic study of patients with dyshidrosis of the hands (cheiropompholyx)].

The purpose of a pilot study with 20 patients, suffering from dyshidrotic exzema of the hands, was to investigate the relevance of a psychosomatic approach for its etiology and therapy. A set of psychological and allergological tests revealed the following results. - 10 patients suffered from a contact allergy (epicutaneous test), 6 of them were allergic to nickel - the patients were less aggressive and more permissive (Freiburger Personality Inventory; FPI) as compared to a standard population. They reported more somatic complaints (HHM) as healthy subjects. - the results of a semistructured interview support the hypothesis, that the dyshidrotic exzema often has an expressive function: the patients present their dependency conflict in a symbolic manner by means of their hands. They try to take their life in their own hands; autonomous actions however are hampered by their own dependency needs. The development of the dyshidrotic symptoms corresponds to the autonomy, which has been achieved with regard to the individual conflict of ambivalence. According to our findings the contact with nickel is not predominant, neither causative nor triggering, factor. Following the psychosomatic concept of Engel and Schmale, the authors discuss the pathogenesis of dyshidrosis with the meaning of a complication of (pregenital) conversion, with the hands as the part affected.

[A psychosomatic theory of allergic sensitization; allergy as a quasi conditioned reaction].

A psychosomatic theory concerning the sensitization process of immediate type allergy is presented. According to this theory the allergy systems from a deficient local defense function of the respiratory tract's mucous membranes. It is assumed that persons, with a specific disposition and learning experiences, in certain conflict situations where emotional impulses cannot be expressed, tend so suffer an obstructive hyperfunction of the nasal and/or bronchial mucosa including hyperemia, swelling, hypersecretion (and spasms of the bronchial muscular system). These modifications of mocous function result from suppressed aggressive expressions (rage, anger, attack) in connection with avoidance tendencies. Thus the immunological reaction may also be the result of bronchial asthma and of chronic rhinitis. This theory allows to regard the process of sensitization as a learning process and to consider the allergy as a classic conditioned response to the atopic allergen as a conditional stimulus. The obstructive hyperfunction (which, of course, can also occur by stimuli other emotional) creates "critical phases". During these the immune system can be activated against atopic allergens in sufficient quantity. However, anyone is probably more sensitized against allergens found in locations where his emotional impulses are not expressed. In this context a house dust allergy, for example, refers to conflicts within the confines of the house, but does not necessarily attribute symbolic qualities to house dust itself.
