[Inhibition of human granulocyte elastase by antibodies to leukocyte thermostable alpha-glycoprotein].

The activity of granulocyte elastase (GE) was discovered in the preparations of leukocyte thermostable alpha-glycoprotein (LTG) isolated from pus by means of ion-exchange chromatography. The activity of GE was determined according to MeoSuc(Ala)2ProValpNa hydrolysis. The antibodies against LTG were isolated from monospecific antisera. Sepharose with immobilized fraction of pus proteins was utilized as immunosorbent. Isolated antibodies to LTG inhibited the GE activity. An inhibitory effect of antibodies increased with the increase in their concentration. The identity or binding of LTG and GE was suggested. The binding of LTG with pus protein component was discovered, the biological meaning of this phenomenon being unknown.

[Correction (in vitro) of proteinase inhibitor balance in the blood of patients after cardiosurgical interventions].

Proteinase-inhibitor balance in human plasma before and after artificial blood circulation was examined. It was shown that trypsinlike and chemotrypsinlike proteinases activities after this procedure were 1.6 and 2.8 times higher and antitryptic activity 1.2 times lower than in the same patients before the procedure. The 40-50% decrease of proteinase activity in human plasma after artificial blood circulation was observed as a result of in vitro plasmasorption on immobilized human urinary trypsin inhibitor.

[Thermostable leukocyte alpha-glycoprotein: immunochemical study and enzyme activity research].

Using immunochemical analysis with standard antisera, leukocyte thermostable alpha-glycoprotein (LT alpha G) was shown to be distinct from lactoferrin, lysozyme, and fibronectin. The determination of peroxidase and nonspecific elastase in immune precipitates of LT alpha G gave negative results. Affinity sorption of LT alpha G onto the pus protein component was revealed. Purified LT alpha G had amidolytic activity in response to a substrate for elastase (p-nitroanilide succinyl-trialanyl). The ability of LT alpha G to cause the hydrolysis of substrates for thrombin, kallikrein, plasmin was investigated. The identity of LT alpha G and granulocyte elastase is suggested.

[Immunochemical determination of the heterogeneity of leukocyte thermostable alpha-glycoprotein].

A comparative immunochemical and physico-chemical study of leukocyte thermostable alpha-glycoprotein (LTG) has been performed in pus extract, hemolysate, leukolysate and blood plasma. Immunochemically determined LTG is heterogeneous in respect of molecular charge, sialic acid content and temperature stability. LTG in the blood plasma has a form of alpha-globulin, while in hemolysate it is a beta-globulin, with both forms present in pus. In leukolysate LTG possessing incomplete immunochemical identity with pus and plasma LTG was detected. It is suggested that LTG forms detected differ in their biological activity as well.

[Determination of the elastase activity of the leukocyte thermostable alpha-glycoprotein from human blood plasma].

The possibility of isolation of specific human granulocyte antigen--leukocyte thermostable alpha-glycoprotein (LTG) from plasma was shown. The isolation of preparations containing LTG was carried out on CNBr-activated sepharose with immobilized soluble fraction of pus. Using immunochemical analysis with standard antisera, the absence of plasma and leukocyte proteins and the presence of LTG in the preparations obtained were demonstrated. Pus may comprise a protein component capable of binding LTG. Elastase activity of the preparations obtained was established. The identity of LTG and granulocyte elastase is suggested.

[Comparative immunochemical and immunocytochemical study of 3 soluble granulocytic proteins in blood cells and serum].

Using immunofluorescence, it has been shown that leukocyte thermostable alpha-glycoprotein (LTAG) and leukocyte beta-globulin (LBG) are, like lactoferrin, components of polynuclear leukocytes. These proteins were found in the perinuclear zone, cytoplasm membrane and extracellularly. Serum LTAG concentration increases in immune inflammatory diseases. In severe forms of diabetes and glomerulonephritis there is a rise in LBG concentration. The biological role of LTAG and LBG is unknown.

[Use of the hyperventilation test in patients with various forms of angina].

The pulmonary hyperventilation (HV) test was carried out in 31 coronary patients with stable angina and 11 patients with spontaneous (angiospastic) angina. The test results were compared to those obtained with bicycle ergometry (BE), 24-hour ECG monitoring and selective angiography. The HV test was positive in 7 of 24 patients with positive BE results. The BE and 24-hour ECG monitoring were superior to the HV test in terms of sensitivity. Positive HV tests were more common in spontaneous angina. It is suggested that the HV test can be used for diagnosis and medication efficiency control in coronary patients.

[Dynamics of coronary artery lumina during tests with cold and with intracoronary and sublingual administration of nitroglycerin in patients with different clinical forms of stenocardia].

Cold tests and those using intracoronary or sublingual nitroglycerin administration were applied during coronarography to 50 coronary patients with varying clinical manifestations of angina pectoris. The quantitative assessment of angiographic patterns showed the cold and nitroglycerin tests to change significantly the extent of stenosis in patients with spontaneous angina (by +13.3 and -30.8%, respectively) and those with angina of effort and at rest (by +5.6 and -12.0%, respectively) and have no such effect in patients with angina of effort. The change in the stenosed coronary arterial lumen was more marked in cases of excentric localization of the atherosclerotic patch inside the coronary artery, as compared to the concentric arterial narrowing. The data obtained demonstrate the variability of coronary arterial lumen at the site of atherosclerotic stenosis in a certain population of patients.

[Complex immunochemical study of plasma proteins and soluble antigens of leukocytes in patients with ischemic heart disease during hemoperfusion therapy].

An immunochemical analysis of 16 individual plasma proteins and 8 leukocyte soluble antigens (LSA) was performed over time in 11 coronary patients after hemosorption. The concentration of plasma lipoproteins gradually returned to normal within 2-3 weeks. Leukocyte dysfunction in the patients manifested itself in change of the specific LSA concentration in hemolysates as compared to that in donors. Shortly after hemosorption a selective 2-8-fold increase in the LSA concentration in the hemolysates was noted. Response of the leukocyte system was observed in all the cases and recorded in repetition of the procedures. Individual LSA function is under study.

[Comparative study of the effectiveness of korinfar, izoptin and obzidan in patients with variant stenocardia combined with effort stenocardia].

Twelve patients with variant angina combined with angina of effort were examined. Repeated exercise tests prior to the administration of antianginal drugs revealed the spontaneous variability of the patients' tolerance to the exercise and of the pulse-pressure index at the exercise peak which may serve as an indirect sign of the angiospastic factor involvement in clinical manifestations of angina of effort. The treatment of such patients with corinfar and isoptin was associated with the elimination and significant reduction of spontaneous attacks of angina attended by a considerable increase in the exercise tolerance and in the pulse-pressure index at the exercise peak. The treatment with obsidan in these cases was less effective: the incidence of spontaneous anginal attacks changed insignificantly, the tolerance to exercise increased with the reduction of the maximum value of the pulse-pressure index.

[Treatment of patients with exercise-induced angina pectoris with korinfar and isoptin].

Forty-eight patients with stable angina of effort of II and III functional classes were investigated to compare the efficacy of the calcium antagonists corinfar and isoptin, using placebo and a simple cross-over design of the study. The two drugs were found to have a high antianginal effect expressed in a statistically significant reduction of the incidence of anginal attacks and an increased tolerance to load as compared to placebo. The degree of the antianginal action of corinfar in a daily dose of 40 mg and isoptin in a dose of 240 mg was approximately the same. In some patients the antianginal effect of corinfar and isoptin was associated with an elevation in the pulse-pressure index at the peak of the load, which may be accounted for by the direct coronarodilatory action of these drugs.
