Frontal gel chromatographic analysis of the interaction of a protein with self-associating ligands: aberrant saturation in the binding of flavins to bovine serum albumin.

Frontal gel chromatography is an accurate method to obtain the total free ligand concentration of a protein-ligand mixture in which ligands self-associate. The average number of bound ligands per protein molecule is obtained as a function of the total free ligand concentration. The method was applied to the interaction of bovine serum albumin with self-associating flavins. The binding curves for FMN and FAD leveled off at about 0.7 and 0.5, respectively. These data were simulated well by a binding model where flavins undergo isodesmic indefinite self-association and the monomer alone binds to a single binding site of albumin. The isodesmic association constants of FMN and FAD were (1.7 +/- 0.1) x 10(2) and (2.2 +/- 0.3) x 10(2) M(-1), respectively. The binding constants of the monomer of FMN and FAD were (7.6 +/- 0.2) x 10(2) and (3.5 +/- 0.2) x 10(2) M(-1), respectively. FMN competitively inhibited the binding of FAD to albumin. The affinity to flavins was in the following order at pH 5.8: lumiflavin, FMN, riboflavin, and FAD. The SH modification and the binding of palmitate did not affect the FMN binding to bovine albumin at pH 5.8. As pH increased from 5.8 to 9.0, the affinity to FMN of bovine albumin decreased 3-fold, whereas that of human albumin increased about 80-fold. The present study clearly showed how isodesmic self-association of a ligand can cause apparent saturation of the interaction of a protein with the ligand at levels lower than 1.

Anesthesia for insertion of a Dumon stent in a patient with a large tracheo-esophageal fistula.

PURPOSE: To present the anesthetic management for the insertion of a Dumon silicon stent to the trachea of a patient with a large tracheo-esophageal fistula. The aim of the stent insertion was to seal the fistula in order to prevent aspiration of esophageal content and subsequent pneumonitis. CLINICAL FEATURES: A 45-yr-old man with a large tracheo-esophageal fistula was scheduled for the insertion of the Dumon stent. Since placement of the stent necessitates the insertion of a rigid bronchoscope, under general anesthesia, with its tip just proximal to the fistula, controlled ventilation was expected to be difficult to achieve because of the diversion of oxygen through the large fistula to the esophagus. We successfully ventilated the lungs, after the fistula was sealed using a large balloon which was inserted in the esophagus, and the stent insertion was completed uneventfully. CONCLUSION: Anesthesia for procedures involving the central airway is challenging. This report describes a simple and practical method to facilitate ventilation by temporary seal of a tracheo-esophageal fistula using a modified esophageal balloon.

[Solvent power of new denture cleaner for denture calculus and the influence of this cleaner on the denture materials].

In order to chemically remove the calculus adhering to a denture, new denture cleaner was prepared on an experimental basis. The principal ingredient of this solution was a chilating agent which contained the phosphoric acid. This study consisted of the fundamental and the clinical examinations of this solution the purpose of which was to test the solvent power of this solution on the calculus and to evaluate its influence on the denture materials and its effectiveness in clinical practice. 1. the fundamental study 1) method (1) The test for the solvent power of this solution. The powder of tricalcium phosphate (T.C.P. powder: Wakohjunyaku Co.) was substituted for the calculus adhering to a denture. (1) The influence of the solution temperature on the solvent power. The T.C.P. powder was dissolved at 20 degrees C, 30 degrees C, 40 degrees C, 50 degrees C and 60 degrees C temperature in 10 g of the solution within 10 minutes and the amount of dissolved powder was weighted. The time was measured with a stop watch (Seiko Co.) until 0.5 g of T.C.P. powder had been dissolved in 10 g of the solution. (2) The effect of using it with an ultrasonic cleaner. The time to dissolve 0.1 g of T.C.P. powder in 10 g of this solution was compared with that of the 15% EDTA solution. The measurement was repeated 5 times and the arithmetic mean of the 5 sample values was calculated in order to show the experimental results. The room temperature was 19 +/- 2 degrees C, the humidity was 56 +/- 3%. (2) The influence of this solution on the denture materials. (1) resin specimen. In order to test the transverse strength, transverse deflection, Young's modulus and Knoop hardness, resin specimens were processed in the manner specified in I.S.O. 1567. The data were compared the resin specimens steeped in this solution and with the control sample that was steeped in water. The specimens were immersed up to 24 hours. Furthermore the existence of tarnish was observed with a 40 times microscope. (2) metal specimen. In order to test the corrosion, metal specimens were produced according to the manner of a corrosion test. The metal specimens were held vertically and the end immersed to 20 mm in this solution for 24 hours and 120 hours each. After steeping, each specimen was measured for loss in weight. The data were calculated as the loss in weight per hour (mg/cm) and the depth of corrosion per year (mm/yr).(ABSTRACT TRUNCATED AT 400 WORDS)

[99mTc-MAA brain scan and CT in the diagnosis of intracranial diseases (author's transl)].

Brain scan with 99mTc-labelled macroaggregated albumin (MAA brain scan) and computed tomography studies (CT) were performed on 78 patients with intracranial diseases. 99mTc-MAA was injected into right and left carotid artery respectively, at the different time. And then diagnostic evaluation of both techniques were investigated. The MAA brain scan was useful for the detection of ischemic lesion in the early stage after onset of cerebral infarction. It was shown in 29.5% that partial field of anterior cerebral artery was injected through the contralateral artery. In these cases, the MAA brain scan was also useful. In the cases of hemorrhagic lesions, however, the CT was more useful than MAA brain scan. Therefore, the both techniques helped each other in the diagnosis of intracranial diseases.

[Diagnostic evaluation on cerebral hemisphere scintigraphy using 99mTc-MAA (author's transl)].

Using 99mTc-labeled macroaggregated albumin (99mTc-MAA), we made an examination of the cerebral blood flow distribution with various kinds of cerebrovascular diseases in 151 cases. The particle size of MAA used in this study ranged from 10 to 100 micron. Scan was performed after injection of 3 mCi of 99mTc-MAA into common carotid artery. No side effects were observed except for one case with headache. Distribution of 99mTc-MAA was found to be homogeneous in normal cerebral hemisphere but not in the lesion. Most lesion was shown as nonradioactive area. Meningioma with hypervascularity was represented as hot area. By this method cerebral infarct could be easily represented not only in main artery but also in such a very small artery as hardly detected by CAG. This method is therefore useful for detecting cerebral infarct, sequela of cerebral apoplexy, head injury and others. The dose of 99mTc-MAA used in this method is much lower compared with those of common brain scan, its excretion is speedy, and therefore internal radiation exposure is diminished.
