[PMN elastase determination in tear fluid in corneal ulcer].

The PMN-Elastase concentration in tear fluid was determined in 87 persons without eye disease and compared with the PMN-elastase values of 28 patients with ulcus corneae. A significant increase in the PMN-elastase concentration was found in patients with ulcus corneae (means = 1196 micrograms/l) in comparison with the control group (means = 73 micrograms/l). The lapse control shows that the PMN-elastase values in the tear fluid of patients with ulcus corneae are elevated for a number of weeks, even though there are clinical signs of healing of the ulcer. The determination of PMN-elastase in tear fluid represents a new parameter indicating the real end of the inflammatory processes in the eye.

Investigation of the hyaluronic acid-copper complex by N.M.R. spectroscopy.

Analysis of the 13C and 1H relaxation data of the hyaluronic acid-copper complex indicates a binding site involving the carboxyl group and O-1 of the D-glucuronic acid moiety. The paramagnetic relaxation of Cu2+ is discussed within the framework of the Solomon-Bloembergen formalism and it is shown that various atoms experience, in addition to the dipolar paramagnetic relaxation, a strong scalar relaxation contribution. E.s.r. spectra have also been obtained in order to determine the binding constants, and measurements at 69 K gave the g-values of the complex.

[Blood sugar and lipid metabolism parameters in diabetic retinopathy].

In Type I and Type II diabetics hemoglobin A1c, triglycerides, total cholesterol, HDL cholesterol, LDL cholesterol, and the apolipoproteins A I and B were determined and compared with a control group. In diabetics the HbA1c values were significantly higher, but were found to fluctuate considerably during several years of observation. Some parameters of lipid metabolism also differed significantly from those in the control group and in most of the diabetics one or more parameters of lipid metabolisms were in the pathologic range. However, no correlation was found between the parameters of blood glucose and lipid metabolism on the one hand and diabetic retinopathy on the other.

In vitro fibrillogenesis of interstitial collagens: electron microscopical studies of long-spacing forms induced by chondroitin sulfate.

Reaggregation behaviour of the three interstitial types of collagen was studied. Pure solutions were prepared and reprecipitation performed by the addition of chondroitin sulfate. The sediments were analyzed in the electron microscope. Classical types of fibrous long spacing collagen could be prepared only from solutions of type I collagen. Segments with an average length of 460 nm and a central overlapping zone of about 120 nm were prepared from solutions of type II collagen. They were termed 'double segment long-spacing collagen' (DSLS). No cross striated fibrils or segments could be obtained from solutions of type III collagen. These results show that there are not only chemical differences between the three interstitial types of collagen but also differences in reaggregation behaviour and in the way of interaction with other macromolecules such as glycosaminoglycans.

Studies on a distinct fraction of bovine vitreous body collagen.

We were able to separate pepsin-solubilized collagen of the bovine vitreous body into three distinct fractions by differential salt precipitation with 0.5 M acetic acid containing 0.7 M, 1.2 M, and 2.0 M NaCl, respectively. The 0.7 M NaCl fraction showed an electrophoretic mobility in polyacrylamide gel identical to that of type II collagen, CNBr-derived peptides very similar to type II collagen CNBr-peptides, and a reaggregation behaviour closely related to type II collagen as observed by electron microscopy. Therefore, the fibres of the bovine vitreous body appear to contain a mixture of a collagen very similar to type II collagen precipitable in the presence of 0.7 M NaCl, at least two different collagen types that can be precipitated at 1.2 M, and 2.0 M NaCl concentrations that have not been identified previously.

[Lipid metabolism disorders in patients with retinal artery occlusions].

Triglycerides, cholesterol, HDL-cholesterol, LDL-cholesterol, the HDL-cholesterol/LDL-cholesterol and cholesterol/HDL-cholesterol ratio, the apolipoproteins A and B, and the ratio apo A/apo B were determined in patients with occlusions of retinal arteries and compared with a control group. The patients showed higher levels of triglycerides, cholesterol, LDL-cholesterol, apo B and the ratio cholesterol/HDL-cholesterol and a decrease in HDL-cholesterol and the two ratios HDL-cholesterol/LDL-cholesterol and apo A/apo B, indicating that disturbances in lipid metabolism may be a risk factor in the development of occlusions of the retinal arteries.

[HbA1 and HbA1c determination in ophthalmology. New parameters for monitoring metabolism in diabetics].

Determination of glycosylated hemoglobins HbA1 or HbA1c by column chromatography is used for long-term glucose control. In the study reported here the authors compared the advantages and disadvantages of this new method and tried to show the importance of these determinations for patients with eye diseases. In a control group (blood glucose less than 120 mg/dl) HbA1-values of 6.5 +/- 1.2% and HbA1c-values of 4.4 +/- 0.5% were found. Well-controlled diabetics (blood glucose 80-180 mg/dl) had an HbA1 of 8.1 +/- 1.6% and an HbA1c of 5.8 +/- 1.1%, while diabetics with poor control (blood glucose 160-420 mg/dl) had an HbA1 of 10.6 +/- 2.1% and an HbA1c of 7.7 +/- 1.7%. HbA1 and HbA1c, respectively, should be measured in addition to blood glucose determination and oral glucose tolerance test.

Preparation of gels from hyaluronate solutions.

Stable, transparent gels can be prepared from hyaluronate solutions by adding CuSO4. The best gelation was found using solutions of 2 mg/ml hyaluronate in glass-distilled water at a pH of 6.2 after the addition of 1 mg/ml CuSO4. Methylation of the carboxyl groups of the hyaluronate completely abolished the gelation, indicating the importance of the carboxyl groups for the gel formation with Cu2+ ions. Gelation also depends on the molecular size of the hyaluronate, since hyaluronate was not able to form a gel after depolymerization with hyaluronidase.

[HbA1 as a parameter of the progression of diabetic retinopathy].

HbA1 was determined in 12 juvenile diabetics with diabetic retinopathy and in ten adult diabetics at four-weekly intervals. During this time the course of the retinopathy was observed. Retinopathy was constant in juvenile diabetics with HbA1 values below 8%. A moderate progression, which it was possible to slow down by laser treatment, was observed in patients with HbA1 values of between 8% and 10%. Patients with HbA1 values exceeding 10% showed a marked progression, which could not be stopped by intensive laser therapy. The behavior of the retinopathy in adult diabetics corresponded to that of the juveniles when the onset of the disease was between the ages of 30 and 50. In elderly diabetics no significant tendency was observed.

A method for the purification of bovine vitreous body hyaluronic acid.

Bovine vitreous body hyaluronic acid was treated with activated CH-Sepharose 4B after purification with cold 5% trichloroacetic acid. By this method a highly purified hyaluronic acid can be prepared. Contaminating serum proteins were completely removed. The remaining small amounts (0.3%) of the protein contaminants were found, by immunological methods, to be non-plasma proteins.

The inability of superoxide dismutase to inhibit the depolymerization of hyaluronic acid by ferrous ions and ascorbate.

Superoxide radicals were investigated as to their capability of depolymerizing the hyaluronic acid of the bovine vitreous body. Using viscometry it was found that O2 radicals, generated by the hypoxanthine/xanthine oxidase method or the combination of NADH and phenazine methosulphate, degraded hyaluronic acid. This reaction was suppressed by superoxide dismutase, catalase, and peroxidase. In contrast, the depolymerization of hyaluronic acid by oxidation-reduction systems like ascorbic acid or ferrous ions was abolished by catalase and peroxidase while superoxide dismutase showed no effect.

[Light and electron microscopy studies of actinomycosis of the lacrimal duct].

For the first time we present electron-microscopic results of an actinomycosis of the lacrimal canaliculus. The interior of the actinomycotic conglomerate showed no evidence of a cellular defence reaction but in the loosly woven outer network of hyphae we observed a massive granulocytic reaction. Also, after phagocytosis the structure of the actinomycotic micro-organisms within the granulocytes is not significantly damaged. Within the tissue of the lacrimal canaliculus adjacent to the actinomycotic conglomerate we observed an increased number of plasmacells but no pathogenic organisms.
