Homogenated stratum coreum, callus and psoriatic scales were extracted with (1) phenol water (PW) and (2) the combined use of trypsin digestion and phenol water extraction (TPW). The serological properties of the various preparations obtained were compared, using indirect haemagglutination, absorption and inhibition tests. The PW and TPW water phases contained two different antigens which were common to all three tissues. In addition, the periodate-treated TPW water phase of stratum corneum contained an erythrocyte-sensitizing antigenic determinant. This, however, cross-reacted with the untreated and periodate-treated preparation of psoriatic scales, whereas callus lacked the determinant present after treatment with periodate. Apparently callus and psoriatic scales lacked some components present in stratum corneum, but determinants specific for callus or psoriatic scales were not detected.
Two brothers with severe combined immunodeficiency were treated with repeated transplantations of fetal thymus tissue. The first patient was not treated until he was critically ill, and the intramuscular transplants had no effect. He died at 11 months of age of overwhelming pneumonia. At postmortem examination a transplanted thymus seemed viable. In the second patient an intramuscular transplant had no effect, but three subsequent intraperitoneal transplants led to transient increase in circulating T lymphocytes with a concomitant fall in B lymphocytes. The results suggested an additive effect of each transplant. However, delayed hypersensitivity skin tests and in vitro mitogen responses were not influenced. Initially, transfer factor was given, and fetal liver was administered intraperitoneally together with the last thymic transplant. Neither of these measures had any observed effect, and this patient, similarly, died of pneumonia at nearly 12 months of age.
The influence of serum natibodies and thermolabile serum factors on the intracellular killing of Staphylococcus aureus by neutrophil granulocytes has been examined using a method which facilitates a precise in vitro evaluation of the phagocytic and bactericidal activities of human polymorphonuclear leucocytes. The bactericidal activity of the granulocytes was significantly less pronounced in the presence of serum absorbed with Staph. aureus or inactivated at 56 degrees C for 30 minutes than in the presence of untreated serum. Specific antibodies seemed to stimulate the intracellular killing of bacteria more than thermolabile serum factors.
The phagocytosis and intracellular killing of Staphylococcus aureus by humans granulocytes in the presence of immunoglobulin preparations have been examined. Isolated IgG from pooled human serum induced phagocytosis and intracellular killing of bacteria. F(ab')2 fragments had no significant effect, indicating that the Fc piece of the IgG molecule is of importance not only for the phagocytosis but also for the intracellular killing of bacteria. Isolated IgM stimulated the phagocytosis to a minor extent, with no enhancement of the bactericidal activity.
The level of Fc receptor rosette-forming lymphocytes (Fc-RFL) was examined in spleen and lymph node cell suspension from neonatal DA and BN rats inoculated within 24 hr of birth with either allogeneic L (experimental) or syngeneic (control) lymphoid cells. In addition, these levels were compared to fetal and neonatal animals that received no injection. The indicator cells (EA) were sheep erythrocytes sensitized with one-half concentration of the highest dilution of rabbit anti-sheep erythrocyte IgG(A) which agglutinated an equal amount of 1% suspension of E. Care was taken to exclude scoring macrophages by injecting colloidal carbon at least 1 hr before killing the test animals. The spleen of 19-day DA fetal rats exhibited a level of 19.3% Fc-RFL, similar to that of animals having received adult syngeneic cells at birth (40.0%) by day 7. Thereafter the level of Fc-RFL did not vary appreciably between these two groups. However, as early as 2 days after inoculation there was a significantly greater number of Fc-RFL in the spleen of experimental DA neonates compared to controls. The lymph nodes of experimental animals did not exhibit a significantly greater number of Fc-RFL until day 6 with both tissue compartments peaking at day 10 and remaining significantly higher than controls until death. In neonatal BN animals significantly higher levels of Fc-RFL in experimental animals were not evident as early and peaked later (day 12) in both tissue compartments but again these differences remained until death. Cytotoxic alloantisera demonstrated that on days 6, 10, and 12 most, if not all, of the Fc-RFL were host in origion in both DA and BN GVHD, with a very significant host plasma cell response in such GVHD animals. One-micron tissue section revealed the presence of a great number of plasma cell especially prominent in the medulla of lymph nodes with the cortex of lymph nodes and white pulp of the spleen markedly depleted of lymphocytes indicative of cytotoxicity.
Twenty human malignant solid tumors of various histologic types were tested for the presence of Fc receptor using cryostat sections or single cell suspensions of fresh tissue. Sheep erythrocytes sensitized by various amounts of rabbit IgG antibodies served as indicator cells (EA). All tumors possessed Fc receptor, but to varying degrees; eight reacted more strongly than normal spleen without any relation to histologic type. The tumors which gave the strongest reactions in sections also formed the highest percent of EA rosettes in suspensions, thus indicating surface localization of receptors. The reactions with spleen sections localized to the B cell and monocytic areas; the latter also showed high avidity in reactions with uncomplexed IgG. Rabbit antisera to tumors, spleen, and peripheral lymphocytes (polyvalent ALS) in inhibited the reactions, while a T-cell-specific ALS did not. Absorptions of the antisera with lymphocytes or tissue sediments of spleen and tumors removed the inhibiting activity, tissue sediments of muscle and kidney only reduced the titers. Again, results with spleen sections paralleled those obtained with tumor sections. Apparently, the tumor Fc receptor is very similar to the Fc receptors present in normal lymphoreticular tissues.
