Selectin-P-mediated adherence of platelets to neutrophils is regulated by prostanoids and nitric oxide.

The expression of Selectin-P was measured in terms of formation of "rosettes" by human gel-filtrated thrombin (30-50 mU)-stimulated platelets on the surface of isolated homologous neutrophilic leucocytes (PMNs) according to Jungi (1986). The monoclonal anti-Selectin-P antibody completely prevented the formation of "rosettes", proving the specificity of Selectin-P-mediated adhesion. The Selectin-P-mediated adhesion of platelets to PMNs was inhibited by both iloprost (ILO) (IC50 = 5.0 nM) and sodium nitroprusside (NaNP) (IC50 = 0.93 microM); thus ILO is ca. 180 times more potent an inhibitor of "rosette" formation than NaNP. The NOS-inhibitors L-NO2Arg (10-30 microM) and L-MetArg (3-30 microM) each suppressed the adhesion, while at lower and higher concentrations these NOS-inhibitors did not influence rosette formation. L-Arginine (up to 1 mM) was not able to influence significantly the Selectin-P-mediated adhesion of platelets to PMNs. The COX inhibitor aspirin (10-30 microM) promoted the adhesion. We conclude that the Selectin-P-mediated adhesion of platelets to PMNs is inhibited by both ILO and NaNP, whereas endogenous prostanoids and nitric oxide seem to exert an antagonist effect on the adhesion of platelets to PMNs.

[Decrease in the level of free cholesterol fraction HDL as a risk factor for threatened atherosclerosis in idiopathic hyperinsulinemia].

Inversive association between concentration of plasma high density lipoprotein (HDL) components and endogenous insulin response to oral glucose uptake was observed in 89 healthy, normolipidaemic, non obese, non diabetic male subjects. Inversive correlation between sum of insulin released during oral glucose tolerance test and HDL free and esterified cholesterol as well as apolipoprotein A1 concentration were confirmed. Coexistence of increased insulin secretion and low ratio of free cholesterol concentration to esterified cholesterol and to proteins in HDL was also observed. In separate group of subjects with normal glucose tolerance in high insulin response subgroup the cholesterol (free and esterified) and protein concentrations were lower than in subgroup with low insulin response. The free cholesterol/esterified cholesterol and free cholesterol/protein (apoA1) ratios were decreased in high insulin response subgroup. It is concluded that low concentration of free cholesterol is a most significant change of HDL composition in hyperinsulinemic, healthy men.

Affinity chromatographic identification of vitamin B12-binding proteins in egg white.

To identify vitamin B12-binding proteins in egg white, an affinity chromatographic isolation procedure was applied. A fraction tightly absorbed on vitamin B12 immobilized in agarose was eluted with 1 mM cyanocobalamin, and then separated by isoelectric focusing in sucrose density gradient. Two isolated proteins (or perhaps two forms of the same protein) of isoelectric points 6.2 and 7.2 and the same Mr 90,000 were found capable of detectable vitamin B12 binding. They are probably glycoproteins, each composed of a single polypeptide chain.
