Dithranol-induced down-regulation of 12(S)-hydroxyeicosatetraenoic acid [12(S)-HETE] receptors in a human epidermal cell line.

The effects of dithranol and its therapeutically inactive oxidation product, danthrone, on 12(S)-HETE binding to the human epidermal cell line SCL-II were studied. Dithranol (0.25-1 microgram/ml), in contrast to danthrone, induced a substantial decrease in 12(S)-HETE binding in a dose-dependent manner. The inhibition occurred after a latency period of 6 h, reached its maximum at 18-24 h and slowly declined thereafter. At a concentration of 1 microgram/ml, the drug led to an approximately 50% decrease in the number of specific high-affinity 12(S)-HEFE receptors (Bmax), whereas receptor affinity (Kd) showed no change. The down-regulation of 12(S)-HETE receptors on epidermal cells by dithranol may contribute to its antipsoriatic action.

Inhibition of 12(S)-hydroxyeicosatetraenoic acid [12(S)-HETE] binding to epidermal cells by ultraviolet-B.

12-hydroxyeicosatetraenoic acid (12-HETE), the main eicosanoid in skin, is assumed to have both pathophysiologic effects in inflammatory skin diseases such as psoriasis and atopic eczema and a physiologic role in the biology of cutaneous reparative processes. Because 12-HETE exerts its effects via specific high-affinity epidermal receptors, and ultraviolet-B (UV-B) is capable of modulating various cell-surface molecules, the effects of single and repeated UV-B irradiations on the 12(S)-HETE binding sites in a human epidermal cell line, SCL-II, were studied. UV-B (100-300 J/m2) induced a large decrease in 12(S)-HETE binding in a dose-dependent manner. The inhibition occurred after a latency period of 6 h, reached its maximum at 18 h and slowly declined thereafter. A single UV-B dose of 300 J/m2 or repeated irradiation with 50 J/m2 of UV-B resulted in a 70% decrease in the number of binding sites (Bmax), whereas receptor affinity remained unaffected. The modulation of epidermal 12-HETE receptors by UV-B may partly explain the therapeutic effects of UV-B, but possibly also contribute to photodamage to skin.

Effect of ciclosporin on epidermal 12(S)-hydroxyeicosatetraenoic acid binding sites.

12-Hydroxyeicosatetraenoic acid (12-HETE) is assumed to play an important role in the pathogenesis of inflammatory skin diseases. Recently, high-affinity binding sites for this eicosanoid have been identified on human keratinocytes by our group. Since ciclosporin exerts therapeutic effects in chronic inflammatory dermatoses such as psoriasis or atopic eczema, the influence of the drug on 12(S)-HETE binding to human keratinocytes was studied. No competitive inhibition of 12(S)-HETE binding was observed in ciclosporin concentrations between 10(-10) and 10(-6) M. In contrast, pretreatment of epidermal cells for 24 h resulted in a dose-dependent decrease of specific 12(S)-HETE binding. The analysis of saturation curves showed that the inhibition of 12(S)-HETE binding by ciclosporin was due to the decrease of 12-HETE binding sites, while receptor affinity remained unchanged. In addition, ciclosporin blocked the interferon-gamma-induced increase in epidermal 12(S)-HETE binding. These findings suggest that the effects of ciclosporin in cutaneous disorders could be partly mediated via an influence on epidermal 12(S)-HETE binding.

Cultivation of psoriatic keratinocytes and an attempt to isolate and detect a specific psoriatic antigen.

Keratinocytes from psoriatic lesion and healthy skin region obtained from the skin of a psoriatic patient were cultivated on lethally irradiated 3T3 cells. They multiplied twice as quickly as normal keratinocytes from healthy skin. Ten antigen fractions were prepared from cell suspensions of psoriatic keratinocytes and from the medium left after their cultivation. Later, they were evaluated in ELISA tests performed with the sera of psoriatic patients and healthy persons. The presence of specific antigens was not demonstrated.

Regulation of 12(S)-hydroxyeicosatetraenoic acid (12(S)-HETE) binding sites on human epidermal cells by interferon-gamma.

We recently detected specific high-affinity binding sites for 12(S)-HETE, the main arachidonic acid metabolite in skin, on epidermal cells. The putative receptor is involved in keratinocyte chemotaxis toward 12(S)-HETE, which points to its participation in wound healing. In an effort to further characterize the 12(S)-HETE receptor, we investigated its regulation by various cytokines. Of the tested cytokines, only interferon (IFN)-gamma led to a massive induction of the 12(S)-HETE receptors. The effect was dose and time dependent and blocked by cycloheximide. The up-regulation of 12(S)-HETE receptors by IFN-gamma may represent an amplification mechanism of the assumed role of 12(S)-HETE in skin wound repair.

[Lower leg ulcers and 2,3-diphosphoglycerate blood level].

In a study on 30 patients suffering from arterio-venous leg ulcers, we found an increased blood level of 2,3biphosphoglycerate (10.33 mumol/ml; control group: 5.16 mumol/ml). This difference, which is statistically significant (p less than 0.001), shows that the blood oxygen pressure is lower both in the tissue of the leg ulcer (literary communication) and in the whole body.

[Effect of tar and dithranol on fibrinolysis in treatment of psoriasis].

There are well known side effects of drugs for topical use due to systemic resorption. In the present study, however, 36 psoriasis blood sera were examined before and fourteen days after the coal tar and dithranol (anthralin, cignolin) therapy onset to determine D and E fibrinogen fragments. These serum levels were compared with those in 36 blood donors. The higher levels in psoriatics and their causes are being discussed.

[Evaluation of automatic sphygmomanometers].

In clinical evaluation of a new type of automatic sphygmomanometer, a strict comparability of results with the standard indirect auscultatory method is required. We have compared the oscillometric apparatus BP 103 N Nippon Colin (B) with the Hawksley random zero mercury sphygmomanometer (A) in a stratified sample of 26 subjects using a cross-over design of four simultaneous pairs of measurements in both arms by two observers. Mean values obtained by machine B were higher than values obtained by machine A, by 0.56 kPa systolic and by 0.62 kPa diastolic. Analysis of variance showed that the differences between both machines were within the random variability of blood pressure values obtained, and therefore irrelevant at single measurements. In case of blood pressure measurements in an epidemiological study, a correction of values should be done using the regression model B = K. Ac.

[Symptoms of alcoholism from the viewpoint of the dermatologist].

There is a wide range of dermatological symptoms that are indicative of alcoholism. In the literature little attention is paid to these manifestations. In the present study 201 alcoholics were examined before beginning treatment and were compared with 42 teetotallers. The symptoms observed and their causes are discussed.

Clinical evaluation of the Nippon Colin BP-103N blood pressure monitor.

The automatic oscillometric Nippon Colin BP-103N blood pressure-monitor was evaluated in comparison with the Hawksley random zero mercury sphygmomanometer using standard methods. The monitor under test was found significantly to over-read mean systolic pressures by 4.2 mmHg and mean diastolic pressures by 4.6 mmHg. The coefficient of variation was 5.4% for systolic and 6.4% for diastolic pressures. Analysis of variance showed that the difference between the two manometers was within the random variability of readings obtained.
