[Plasma histamine and exercise-induced bronchospasm].

The pathogenesis of bronchospasm of effort is not yet elucidated and it is probable that is determined by multiple factors. In this study we have tested the role of mastocytes in taking as an index of their activation the fluctuation of plasma histamine levels during repeated exercise tests. We will show that asthmatic subjects who develop bronchospasm on effort have a significant rise (times 3) in their level of serum histamine whilst normal subjects don't show much change. Furthermore in a repeated effort test we have observed a strict parallelism between the occurrence of bronchospasm of effort and a rise in plasma histamine levels. These different observations enable us to consider that the activation of mastocytes and the liberation of mediators which follow play an important role in the pathogenesis of bronchospasm of effort.

Some remarks on the axial skeleton in Protopterus (Pisces, Dipnoi).

Based on a comparative morphological study of the axial skeleton in a broader range of "primitive bony fishes" a reevaluation of some characteristic traits of the vertebral column in the African lungfish has been done. The axial skeleton of living dipnoans has to be regarded as reduced from a more complex phylogenetic state and may be neotenic in some respect. The formation of perichordal bony autocentra could be confirmed, at least in one specimen. The well-known invasion of mesenchymal cells into the fibrous sheet of the notochord is interpreted as a rudimentary chordacentric "vertebralization". Also included is an analysis of the peculiar urostyle of lungfishes, its regeneration capacity and its bearing upon the principles of vertebra-formation.

Animals

[Visual evoked potentials and water deprivation of rats during intrahypothalamic application of substance P].

Visually evoked potentials (VEP) were recorded in the visual cortex, the dorsal geniculate nucleus and the lateral hypothalamus of satiated and water-deprived rats with and without additional drinking sounds to strengthen motivation and were compared with those during substance-P-injections. Differences in VEP and corresponding spatial and temporal interrelationships of these areas were found by using the method of coincidence of single VEP. Variations of drinking behaviour, weight loss and motor activity were recorded. The findings are discussed with regard to SP and its function as a "regulid peptide".

Quality of life of inoperable non-small cell lung carcinoma. A randomized phase II clinical study comparing radiotherapy alone and combined radio-chemotherapy.

Eighty one patients with inoperable non-small cell lung carcinoma (NSCLC) were entered in a randomized phase II trial comparing split-dose irradiation alone to combined treatment radiotherapy and polychemotherapy (C.A.P. + V.D.S.). The quality of life and the survival of the patients were studied. We have defined three classes of quality of life responses based on the time elapsed before the performance status index drops. A higher quality of life failure rate was observed in the combined treatment group (p non-significant) but the time elapsed before the Karnofsky index drops is longer in the combined treatment group for the quality of life "no change" subgroup (p = 0.15). Survival and quality adjusted survival are similar in both treatment groups. The same conclusion holds for retrospective stratified treatment groups. The results of the study are presented according to the decision tree theory. We conclude that as far as the quality of life is concerned, polychemotherapy combined with the particular split-dose irradiation schedule used is an effective treatment of inoperable NSCLC. Its efficiency is comparable to, but not better than, the same radiotherapy schedule alone taken as a reference baseline.

[A radioisotope method of measuring pulmonary vascularization].

The study of regional perfusion of the lungs by a radioisotopic method using a multiple detectors system and 133 Xenon as radioactive tracer, seems to us, an interesting tool in the investigation of valvular or congenital cardiopathy. This method can be repeated as much as necessary and is not traumatic. The reproductibility is good. The radiation exposure is minimal as well for the patient as for the physician.

[Effect of repeated serotonin injections on temperature regulation in the rat].

The effect of repeated serotonine injections was investigated in thirty Wistar rats tested under normo-, hypo- und hyperthermic conditions. 7 mul of a 0.5% serotonine solution (5HT) were injected into the third ventricle at 1-hr intervals. Temperature responses such as oxygen consumption and rectal temperature were measured and the heat loss was calculated. Intraventricular serotonine produced cooling in all animals. The central mechanisms of heat production were inhibited and those of heat loss were activated. The serotonine responses were most pronounced in hypothermic rats and least effective in hyperthermic animals.

[Influence of mediopreoptic stimulation on temperature regulation in rats before and after serotonin injection].

The effects of electrical stimulation of the Area praceoptica medialis (APM) and intraventricular serotonine injection were investigated in thirty Wistar rats tested under normo-, hypo- and hyperthermic conditions. Temperature responses such as oxygen consumption and rectal temperature were measured and heat loss was calculated. The effects of the first stimulation before serotonine injection were compared with those of the second one after serotonine application in order to prove the influence of serotonine on temperature responses due to APM stimulation. On the other hand, the influence of stimulation on changes produced by serotonine injection should be shown by the recordings of the second serotonine responses with stimulation effects and by comparison of these with those of the first serotonine responses without stimulation effects. The first APM stimulation depending on the rectal temperature before stimulation resulted in decreasing oxygen consumption in hypothermic rats while increasing that of hyperthermic animals. There was a little decrease of rectal temperature under hyper- and normothermic conditions. while under hypothermic conditions animals showed marked depression. The increase in heat loss, however, was least in hyperthermic rats and most pronounced in hypothermic animals. The second APM stimulation did almost not affect the temperature responses after serotonine application in normothermic rats, while causing supercooling agin under hypothermic conditions and insignificant cooling at hyperthermic environment temperature. Comparison of the first and second stimulation effects revealed statistical significant differences at normo-, hypo- and hyperthermic temperatures. The results are discussed under consideration of the different starting conditions before the first and second stimulation. These findings do not indicate that serotonine would shift the sensibility of mediopraeoptic structures and thus unequivocally modifies stimulation effects.

Live influenza vaccine in patients with chronic bronchopulmonary diseases. A multicenter study with two consecutive vaccinal strains.

Live influenza vaccines RIT 4025 and RIT 4050, containing the recombinants of A/Scotland/840/74 and A/Victoria/3/75 with A/PR/8/34 virus respectively, were administered during the vaccination campaigns 1975-76 and 1976-77 to patients with established chronic bronchopulmonary diseases. The study was designed to assess the clinical acceptability of these vaccinal strains and to measure the effects of vaccination on pulmonary function in this population. Clinical and respiratory function findings were compared in a control group of healthy volunteers. Good immunogenicity and good clinical tolerance were found in both populations. Pulmonary function tests had been performed before administration of the vaccine and repeated several times during the post-vaccinal period; the longest post-vaccinal follow-up of respiratory function in patients was four weeks. Apart from transient fluctuations, no significant changes in pulmonary function were found; results obtained in the group of patients were not different from those o0served in healthy subjects. These results correlate very well with observations reported with previous vaccinal strains (Ann and Alice) in pulmonary "high risk" patients.
