Thyroid hormone inhibits fibronectin synthesis by cultured human skin fibroblasts.

Fibronectin (Fn) is a glycoprotein composed of two different subunits, each with a mol wt of approximately 230K. Fn is important for cell adhesion and for maintaining normal cell morphology. Recent in vivo studies suggested that thyroid hormone affects the plasma Fn level in man. The aim of this study was to determine whether T3 regulates the synthesis of Fn in cultured human skin fibroblasts. Skin fibroblasts obtained from seven normal subjects were grown in Minimum Essential Medium supplemented with 10% fetal calf serum. After reaching confluence, the cells were exposed for 3 days to the same medium in which fetal calf serum was replaced by 10% thyroidectomized bovine serum without or with added T3. At the end of this period, the cultures were incubated with [35S]methionine (95 microCi/dish) for 4 h before harvesting. Cell lysates and corresponding media were combined, and 35S incorporation into total protein and Fn was determined by trichloroacetic acid precipitation and immunoprecipitation with antihuman Fn immunoglobulin, respectively. Analysis of the immunoprecipitated material by sodium dodecyl sulfate-polyacrylamide gel electrophoresis revealed that most of the 35S activity migrated as a 230K band which was displaced by excess unlabeled Fn. Addition of T3 did not affect trichloroacetic acid-precipitable 35S activity but decreased the 35S activity precipitated with anti-Fn in a dose-dependent manner. While addition of 10(-10) M T3 to the medium had no effect, 35S incorporation into Fn was inhibited by 22.5 +/- 7.4% (mean +/- SD) with 10(-9) M T3 (free T3, 6 X 10(-12) M) and by 31.3 +/- 5.8% with 10(-7) M T3. To assess whether the inhibition of Fn accumulation by T3 was due to suppression of Fn synthesis or enhanced Fn degradation, cells were labeled for 4 h with [35S]methionine and chased for another 4 h with excess unlabeled methionine. T3 had no effect on the rate of decline of [35S]Fn during the chase. We conclude that physiologic T3 concentrations inhibit the synthesis of Fn in normal human fibroblasts. This effect provides a new method to study the action of thyroid hormone which may prove useful in the tissue diagnosis of resistance to thyroid hormone.

Resistance to thyroid hormone diagnosed by the reduced response of fibroblasts to the triiodothyronine-induced suppression of fibronectin synthesis.

The syndrome of generalized resistance to thyroid hormone (GRTH) is due to a defect at the level of target tissues, but its diagnosis is currently based on indirect tests and cumbersome clinical observations. We recently reported that physiological amounts of T3 inhibit fibronectin (Fn) synthesis by human fibroblasts derived from normal donors. The aim of this study was to assess if this observation could be used for the direct tissue diagnosis of GRTH. Skin fibroblasts from seven normal subjects and seven patients with GRTH were grown to confluence and then exposed for 3 days to medium supplemented with 10% thyroidectomized bovine serum with or without added T3. Cells were then labeled with [35S]methionine for 4 h, and the combined medium and cell lysate was examined for the incorporation of [35S]methionine into trichloroacetic acid and anti-Fn-precipitable material, followed by analysis on sodium dodecyl sulfate-polyacrylamide gel electrophoresis. 35S activity in the 230K protein was corrected for trichloroacetic acid-precipitable 35S activity. To assess the selectivity of the T3 effect on Fn in fibroblasts from normal subjects compared to GRTH patients, the responses of Fn to dexamethasone and Na butyrate were also determined. Addition of 10(-9) and 10(-7) M T3 to the medium (free T3, 6 X 10(-12) and 2 X 10(-9) M, respectively) reduced Fn synthesis by 13.2 +/- 3.2% (+/- SD) and 14.3 +/- 8.2%, respectively, in fibroblasts from patients with GRTH, an effect significantly less (P less than 0.02 and P less than 0.005) than the 22.2 +/- 7.4% and 30.2 +/- 5.0% reduction that occurred in fibroblasts from normal subjects. With the exception of one patient with GRTH, individual values from each of the two groups did not overlap; the clinical studies suggested that this patient had a milder form of GRTH. In contrast, dexamethasone and sodium butyrate stimulated Fn synthesis by 25.6 +/- 8.7% and 268 +/- 125%, respectively, in fibroblasts from patients with GRTH compared to 27.6 +/- 7.2% and 304 +/- 125% in fibroblasts from normal subjects. The difference of the responses between the two groups was not significant. The results indicate that measurement of the response of Fn to T3 in cultured fibroblasts can be used for the tissue diagnosis of GRTH.

Architecture of sheep haemal nodes.

Sheep haemal nodes are reported to be lymphatic organs within the blood circulation. In this study they were seen to be covered by a capsule of connective tissue, with a hilus in which some blood vessels were detectable. Below the capsule a subcapsular blood sinus and lymphatic nodules were present. A reticular meshwork extended through the interior; the interstices of the reticular meshwork contained a large number of free blood cells, many macrophages, lymphocytes and plasma cells. Blood collected in the subcapsular sinus and flowed through the reticular meshwork aided by the particular alignment of some of the reticular cells in channel-like passageways. Venules originated in the parenchyma and drained into efferent veins that left the organ at various points in the capsule. A large efferent blood vessel, apparently connected with the subcapsular sinus, was present in the hilus.

Role of autoimmune and familial factors in goiter prevalence. Studies performed in a moderately endemic area.

The goitrogenic role of autoimmune phenomena in endemic goiter is still uncertain. Scanty and discrepant results have been reported in different areas of the world. This prompted us to evaluate the prevalence of circulating thyroid antibodies in an area of North-Western Tuscany during a survey for endemic goiter. The survey was carried out according to the P.A.H.O. criteria in a stable community. In all schoolchildren (n = 142, age range 7-15 yr) and in most of their parents (n = 159), thyroid size was evaluated and urine was collected for iodine determination. Blood was drawn for determination of circulating thyroid microsomal (MAb) and thyroglobulin antibodies (TgAb), TT3, TT4 and TSH. Prevalence of goiter in schoolchildren was 77.9% and 94.8% in their parents. Mean (+/- SD) urinary iodine excretion was 55.0 +/- 2.1 micrograms/24 h. The overall frequency of TgAb and MAb in the adult population was 14.4%, statistically higher than that of control subjects matched for sex and age. The frequency in schoolchildren was 4.3%. The presence of goiter in children was unrelated to the presence of thyroid antibodies in parents, whether goitrous or nongoitrous. A higher prevalence of goiter was found in children with goitrous parents as compared to children with nongoitrous parents (p less than 0.005). In conclusion, the frequency of thyroid autoantibodies in the adult population of the endemic area studied was increased, but showed no relation with the presence of goiter. The prevalence of goiter in children was associated with the presence of goiter but not of thyroid autoantibodies in parents. These data suggest that autoimmune phenomena are of limited importance in the development of endemic goiter.

Cytochemical identification of lymphocytes and other mononuclear cells in ovine and bovine hemal nodes.

Cytochemical and immunocytochemical studies were carried out with specific markers for B lymphocytes, dendritic reticulum cells (ATPase, 5'Nase, SIg) and T lymphocytes (ANAE, A.P.) in an attempt to identify the mononuclear cells present in bovine and ovine hemal nodes. The results show that primary nodes and mantle of secondary nodes are composed of B cells, whereas T cells are mainly localized in the interfollicular cords. Since such an arrangement resembles the picture in normal lymph nodes, but the direct contact between lymphatic tissue and blood is more reminiscent of the spleen, hemal nodes probably perform immunological functions similar to those of both normal lymph nodes and spleen.

Thyroid-stimulating antibody mimics thyrotropin in its ability to desensitize the adenosine 3',5'-monophosphate response to acute stimulation in continuously cultured rat thyroid cells (FRT-L5).

TSH preincubation induces refractoriness to further stimulation of cAMP accumulation by the hormone in cultured thyroid cells. The question of whether the thyroid-stimulating antibodies (TSAb) present in sera of patients with Graves' disease have the same desensitizing effect as TSH was evaluated in this study using a differentiated strain of rat thyroid cells (FRT-L5) which requires TSH for growth and develops refractoriness to acute TSH stimulation of cAMP accumulation. Cells cultured for 4-5 days in medium deprived of TSH recovered responsiveness to TSH and TSAb. Refractoriness to TSH was reinduced in these cells by a 24-h preincubation with 250 microU/ml TSH. The addition of 100 microM KI or 100 microM methimazole to the medium together with TSH did not modify TSH-induced refractoriness. Four different TSAb preparations all induced refractoriness to TSH-stimulated cAMP accumulation. TSAb preincubation induced refractoriness to acute TSAb as well as TSH-stimulated cAMP accumulation. TSAb-induced desensitization was dose dependent and required prolonged (8-24 h) exposure to TSAb. Furthermore, the desensitization induced by TSAb as well as that caused by TSH were significantly inhibited by cycloheximide, indicating the need for de novo protein synthesis. In conclusion, TSAb mimics TSH in the induction of refractoriness in FRT-L5 cells.

Innervation of the bovine naso-labial glands.

An investigation was carried out on the innervation of bovine naso-labial glands. Histochemical techniques were used to establish the presence and distribution of cholinergic fibres and peptidergic fibres containing VIP and Substance P were revealed by immunohistochemical methods. Parasympathetic nerve fibres were localized around the secretory units and intralobular excretory ducts. VIPergic nerve fibres, the only peptidergic fibres encountered, were arranged in a similar pattern. Adrenergic fibres were only seen in the periglandular connective tissue around blood vessels of medium size. We are, therefore, of the opinion that the secretory activity is mainly regulated by the parasympathetic nerves and that the VIP acts as a modulator.

Synchronous adenoacanthoma of the ovary and endometrial adenocarcinoma. Report of two cases.

Two cases of synchronous endometrioid adenocarcinoma of the endometrium and ovary are here presented. The clinical aspects and the pathologic findings support an autochthonous origin of these neoplasias. The existence of the extended Müllerian system can explain such a possibility. The recognition of this possible association is really important because it implies a different therapeutic approach.

Immunohistochemical localization of cyclic AMP and ultrastructural demonstration of adenylate cyclase activity in the testis of Esox lucius at time of spermiation.

In the testis of Esox lucius at the time of spermiation, activity of cyclic adenosine 3',5'-monophosphate (cAMP) was immunocytochemically localized at the level of the Sertoli cells. In these cells adenylate cyclase activity was also ultracytochemically demonstrated by using adenylyl imidodiphosphate as a substrate. Reaction products of adenylate cyclase were primarily detectable on the basal and adluminal plasma membranes and on the surface of protrusions of the cell body into the lumen.

Reciprocal changes of serum thyroglobulin and TSH in residents of a moderate endemic goitre area.

Subjects living in iodine deficient areas were reported to have elevated serum thyroglobulin (Tg) concentrations. This finding was interpreted as related to thyroid stimulation. Discrepant results, however, were found when serum Tg concentrations were correlated either with serum TSH or with goitre size. In this study we investigated the relationships between goitre size, serum Tg and serum TSH in 488 unselected adult subjects living in an endemic area of North-Western Tuscany (Garfagnana district). The control group comprised 352 subjects residing in a non-endemic area. In the endemic area a high prevalence of goitre was found (80.1%), thyroid enlargement being slight to moderate in the majority of cases and very large only in six subjects. Serum Tg concentrations increased and serum TSH levels decreased with the size of goitre. Statistical analysis by the chi-square cross correlation test showed that the converse changes of serum Tg and serum TSH in relation to goitre size were highly significant. These findings indicate that the increase of serum Tg occurring in endemic goitrous subjects may be related to factors other than TSH stimulation. Functional autonomy of the thyroid may account for the finding of low serum TSH and elevated serum Tg values in patients with large goitres. The present data do not exclude the possibility that the release of Tg is influenced by TSH stimulation, but indicate that other factors may be responsible for the increased levels of Tg found in endemic goitre.

Histochemical characterization of the naso-labial glands of two species of ruminants (Ovis aries and Bos taurus).

The bovine and ovine naso-labial glands have been examined. In both species these glands are multilobular tubulo-acinar, but the cells of secretory units display differences regarding nuclear shape and location, RER arrangement and secretion pattern. In this paper, histochemical characterization of naso-labial gland secretion was performed using the periodic acid Schiff reaction (PAS), the Alcian blue staining (AB) at pH's ranging between 1.7 and 2.5, the Alcian blue-high iron diamine (HID-AB) method, an immunohistochemical staining for muramidase at the light microscopic level and the periodic acid-thiocarbohydrazide-silver proteinate (PA-TCH-SP) technique at the electron microscopic level. In bovines the secretion is composed of glycoproteins, which are reactive to PAS, but not to Alcian blue staining. In ovines, a large quantity of acidic polysaccharides in association with a small quantity of sulfomucins has been found. Ultrastructural and cytochemical observations show that secretory granules do not have a homogeneous structure. They appear to be formed of a meshwork stained positively with the PA-TCH-SP method immersed in a light background. The particular morphological features of the naso-labial glands and their histochemical features suggest that these glands can be classified as sero-mucous in bovines and mucous in ovines.

Identification of lymphocytes extracted from bovine hemal nodes.

Lymphocyte populations extracted and separated from hemal nodes (HN) and peripheral blood of 20 male healthy bovines were characterized by surface markers and the rosette-forming test. After treatment with FITC- or peroxidase-conjugated antibovine IgG, about 22% of cells from HN and peripheral blood showed superficial fluorescence (B cells) and about 13% were able to form "E rosettes" (T cells) with sheep erythrocytes pre-treated with neuraminidase. Nearly equal percentages were obtained from peripheral blood lymphocytes. On the basis of the data shown, we can conclude that the function of HN is, at least in healthy animals, analogous with that of lymph nodes. In this study we also used the SEM to evaluate the possibility of classifying the lymphoid cells as B and T lymphocytes on the basis of their surface morphology. Some authors have stated that B and T lymphocytes have a rough and smooth surface, respectively. In our experience, however, because of cells with a moderate number of surface microvilli exist, this method used on its own is not suitable for lymphocyte identification.

[Screening for aminoacidopathies in newborns by means of an aminoacid analyzer. Reference values and statistical determinations (author's transl)].

Plasmatic concentrations of amino acids valine, methionine, isoleucine, leucine, tyrosine and phenylalanine, in 4100 newborns of age varying between 1 and 18 days have been analyzed. The samples were randomly taken among the newborns of Lazio, Abruzzi and Molise subjected to neonatal screening for aminoacidopathies at the Centro delle Oligofrenie Dismetaboliche, Istituto di Neuropsichiatria Infantile. Average concentrations in mumoles/100 ml of the aminoacids and their correlations with weight and age have been studied. A comparison of the results of this research with the already published data is made, and the cut off points in mass screening for aminoacidopathies are calculated.

[Analysis of the discriminative capacity of hematological tests used in the diagnosis of sideropenia and microcythemia].

The significance of haematological tests has been measured on a statistical base. The purpose of such an inquiry is the simplification of diagnosing thalassemia and sideropenia. A pilot research has been performed on a sample of 10 thalassemic male subjects and 14 female subjects, 10 male subjects and 14 sideropenic female subjects compared with the same number of "normal" subjects of two sexes. This research has pointed the particular sensibility of some tests. There are reasons to consider best tests, in the order, HbA2, Hcv, MCHb for discrimination between "normal" and "thalassemic" and "sideropenic" subjects; Sideremia, Hb, GR for that between "normal" and "sideropenic" subjects. By combining such tests one obviously obtains more reliable results. Combination of tests Hb, Sideremia, GR appears a useful test for the classification of the subjects. Particularly we propose for the "double cases" the consideration of the test HbA2.

Cardiac S-100a0 protein: purification by a simple procedure and related immunocytochemical and immunochemical studies.

A simple procedure is described for the purification of the alpha alpha isoform of S-100 proteins (S-100a0) from porcine heart. Purification steps include the following: i) extraction of the tissue with a hypotonic medium containing EDTA; ii) ammonium sulfate fractionation (0-50%) of the extract; iii) Ca2+-dependent affinity chromatography of the supernatant obtained through the preceding step on phenyl-sepharose and elution of absorbed proteins through a two-chamber gradient of 1.0-0.0 mM CaCl2 and 0.0--1.0 mM EGTA, respectively; and iv) chromatography of the resultant S-100-containing fractions on Sephadex G-200. The yield is 20 mg S-100a0/kg porcine heart. The whole procedure takes five days and is highly reproducible. Data obtained from the phenyl-sepharose step suggest that the affinity of Ca2+ for S-100a0 increases by several orders of magnitude once the protein had interacted with that matrix. This observation is discussed in relation to the role of S-100 proteins in amplification of the Ca2+ signal. Immunocytochemical and immunoblotting analyses indicate that S-100a0 is exclusively found at the level of the sarcolemmal membranes, the membranes of the sarcoplasmic reticulum, the external mitochondrial membranes, and in the adjacent sarcoplasm. No evidence of S-100a0 being associated with the nuclei or with myofibrils has been obtained. Finally, the cardiac tissue does not contain the Triton X-100-extractable fraction of S-100 normally detected in the brain and in adipocytes. Our data suggest that S-100a0 behaves as a peripheral membrane protein in cardiac tissue.
