Quantification by image analysis of immunocytochemical reactions: application for determination of lysozyme content in individual smeared cells.

The objective of the present study was to develop a cytophotometric technique to quantitate immunocytochemical reactions. Cell antigens were detected after immunophosphatase alkaline staining procedure. The amount of reaction product was quantitated by computerized scanning cytophotometry. The technical conditions (dilution of primary antibody; incubation time of the three antibodies; volume and pH of the enzyme substrate reaction; storage of the slides) required for optimal cytophotometric determination of the reaction product were determined. Under these optimally defined conditions, a linear relationship between cell protein content (lysozyme) and microdensitometric measure of the colored reaction product was found. This method could be used for other cells, antigens, and enzymatic indicators.

[Spasm in a single coronary artery].

A 52 year old man presented with effort and resting angina with positive exercise stress testing and myocardial scintigraphy. The initial coronary angiogram showed a single coronary vessel arising from the right coronary ostium dividing into a right coronary artery with a normal trajectory lined with a few non stenotic atheromatous plaques and a left coronary artery which passed between the aorta and pulmonary artery to reach the left atrioventricular groove. At first, it was suggested that the angina was due to compression of the left coronary vessel by the great arteries during effort, but this mechanism could not explain attacks of resting angina and the failure of betablocker therapy. An ergometrine test performed during repeat coronary angiography induced almost complete occlusion of the right coronary vessel and Prinzmetal diaphragmatic ischaemia. The anginal attacks were completely suppressed by Calcium antagonist drugs. The association of coronary spasm and a single coronary artery is rare; only one case has been previously reported. We do not believe that there was a relationship between the two phenomena in our case: the spasm occurred at a distance from the aberrant course of the vessel. The authors emphasise the diagnostic problems and the potential danger of this association.

[Infarction with late disappearance of the Q wave. Apropos of 30 cases].

A selective coronary arteriography was performed in 30 subjects with myocardial infarction, after disappearance of the necrosis wave. The angiographic characteristics of this group when compared with a series of infarctions, unselected by the clinical course, wers: a decreased severity index; a better development of the compensatory circulation; and a lesser frequency of severe disturbances of ventricular dynamics. Disappearance of the Q wave might be explained by a very distal occlusion in 2 cases, by a recanalization in 4 cases and by a very intense development of the collateral circulation in 8 cases. One subject had a normal coronary arteriography.
