[Pharmaceutical technology of Tensiomin].

The physical chemical properties, stability and incompatibility of captopril (the active ingredient of Tensiomin tablets) have been discussed. Captopril has two polimorphic crystal modifications, the form I. of higher melting point is applied in the therapy. Captopril has a better stability in solutions below pH 4, the degradation is accelerated by metallic ions (Cu, Fe). In solid phase the degradation is accelerated by the humidity of the air. No incompatibility was found with the tabletting excipients but stearic acid and sodium-carboxymethyl-starch. Under compaction at higher pressure captopril itself shows tendency for lamination. The dissolution rate of the Tensiomin tablets can meet the requirement of USP if direct tabletting method was used. In this case the breaking strength of the tablets has no influence on the dissolution rate. The main properties (weight uniformity, content uniformity, tensile strength, friability, disintegration time, dissolution time) of the Tensiomin tablets of 12.5 mg, 25 mg, 50 mg and 100 mg of captopril meet the requirement of USP not only after manufacturing but after the accelerated stability test of three month as well.

[Pharmaceutical technology of Grandaxin].

The physical chemical properties, stability and incompatibility of tofizopam (the active ingredient of Grandaxin tablet) were investigated. Tofizopam has low water solubility therefore experiments were made to increase it. The behaviour of tofizopam in acidic medium was studied and the degradation rates of it were determined at different pH. The composition of Grandaxin tablet contains well established excipients. The granulation can be made by high shear and fluidized bed process as well. The properties of tablets (strength, friability, disintegration, dissolution) are independent from the granulation process. The scaling up of the technology and the commercial production of the tablets are managed successfully.

Cytology of primary versus secondary genital herpesvirus infection.

Certain authors state that primary genital herpes infections mainly produce cells with ground-glass nuclei, whereas secondary infections produce a higher percentage of multinucleated or single cells with prominent intranuclear inclusions. The cervicovaginal smears of 11 patients with primary genital herpes infection and 16 patients with secondary infection were studied to determine the validity of this differentiation. The percentage per patient of virally infected cells containing distinct inclusions ranged from 0% to 35% in the primary herpes group and 0% to 100% in the secondary herpes group. In eight additional patients, the initial herpes simplex virus (HSV)-positive smear was compared with recurrent positive smears taken from 2 to 120 mo later. No statistical difference in the incidence of distinct inclusions in the infected cells was detected. Finally, primary rabbit kidney and the heteroploid A549 cell cultures were inoculated with herpes viruses and followed by fixation with several agents at various time intervals. Alcohol and Bouin's fixative favored the recognition of distinct inclusions, as did the duration of the infection.

Fibroadenomas with stromal cellularity. A clinicopathologic study of 21 patients.

Data on clinical and pathologic features of tumors from 21 patients who had breast tumors compatible with juvenile (cellular) fibroadenomas (JCF) were reviewed. All patients were black females ranging in age at time of presentation from 10 to 39 years, with a median age of 15 years. Follow-up (median period, six years) was obtained for 20 patients. Patients were categorized into two groups; 13 had solitary lesions and eight had multiple and successive lesions. No histologic differences were observed between JCF occurring as solitary or multiple tumors or between lesions occurring in patients younger and older than age 20 years. The JCF was shown to be a fairly distinct clinicopathologic entity that characteristically occurred in adolescents, but which was occasionally seen in young adults. It had a benign course, characterized by synchronous and metachronous multicentricity, rather than local recurrence.

A rapid technique for alkaline phosphatase enzyme activity in tissues.

A rapid method is described for serial determination of alkaline phosphatase enzyme activity in the rat kidney and the developing avian vertebra by using an improved paraffin embedding process and a slide staining rack. Tissue samples fixed 12 to 24 h in 80% ethanol at 0 to 4 degrees C are dehydrated in absolute acetone at 0 to 4 degrees C, and then embedded in paraffin of 50 degrees C via benzene at room temperature. The alkaline phosphatase enzyme activity is essentially demonstrated with the method of GOMORI and TAKAMATSU but modified for the use of the slide staining rack. With the new technique, the enzyme activity is completely preserved and reproducible for years in the paraffin embedded tissues. The good quality of the sections enables quantitative evaluations by video scanning.

Application of factorial design in the examination of Tofizopam microcapsules.

The authors studied the parameters of sustained released Tofizopam microcapsules, prepared by a melt-dispersion method. The parameters examined were the flowing rate, strength and dissolution characteristics. Dissolution curves were linearized by the RRSBW distribution. The factorial design method proved to be very useful for examination of microcapsules.
