Continuous wave Doppler flow velocity waveforms from the umbilical artery in normal pregnancy.

The objective was to establish a normal reference range of the indices of umbilical artery waveforms using continuous wave doppler ultrasound. A prospective longitudinal study was made of 85 uncomplicated singleton pregnancies between 16 and 40 weeks gestation (783 recordings). Pulsatility Index (P.I.), Resistance Index (R.I.) and A/B ratio were measured at each visit. The mothers all delivered normal healthy infants weighing more than the 10th centile for gestational age. All three indices have a positively skewed frequency distribution, a factor not generally appreciated until recently and the ranges are therefore described using non-parametric statistics. Placental resistance decreases as term approaches, the 50% PI at 16 weeks was 2.06, at 32 weeks was 1.08 and at 40 weeks was 0.85. The results show a comprehensive reference of normal values and provides a sound basis for the further study of abnormal pregnancy. It represents the largest published study of its type using continuous wave Doppler.

Flow cytofluorometric analysis of choriogonadotropin-like material on the surface of human and mouse malignant cells.

Quantitation by flow cytofluorometry of the distribution of human choriogonadotropin (hCG)-like material on the surface of various human and mouse tumor cells grown in tissue culture and as solid tumors has been done using fluorescein-tagged rabbit antisera (IgG fraction) to intact hCG and, in one experiment, by use of two monoclonal antibodies specific for hCG. Fibroblasts were used as a negative (nontumorigenic) cell control, and a rabbit antiserum to human hemoglobin was used as reagent control. All malignant cells tested stained more intensely with the anti-hCG serum than with the antihuman hemoglobin serum. Positive reaction with the monoclonal antibodies specific for hCG provided strong evidence that the material stained was identical to hCG. Heterogeneity of the expression of the hCG-like material was notable both within each cell line and between different cell lines. This heterogeneity was not associated with cell-cycle phase. 3T3 fibroblast-like cells in vitro were originally negative for hCG but acquired reactivity with anti-hCG serum after ten passages.

Improved clonal growth of human solid tumors by isolation of viable cells.

The potential for the establishment of predictive assays for cancer chemotherapy was generated recently by the development of a soft agar clonogenic assay for human tumor cells. Though the assay technique is promising, certain technical problems remain. One is the difficulty of preparing viable cell suspensions from solid human tumors. A method of separating viable and nonviable cells by Ficoll-Hypaque centrifugation was used. When this method was used to plate isolated viable single cells, an improvement in cloning was observed when compared with a nonseparated mixture of viable and nonviable cells.

Faucial bonorrhoea.

Nine hundred patients (250 females and 650 males) attending the Clinic for the Treatment of Venereal Diseases had throat swabs taken as a routine; 1-1% (1-6% of females and 0-92% of males) were found to have faucial gonorrhoea. This figure appears to be similar to that found by Odegaard and Gunderson (1973) in Norway. The treatment regime used in the clinic in Perth appeared to be satisfactory, as there were no failures. It is stressed that all patients suspected of suffering from venereal disease should have their throats swabbed as a routine. Information as to whether or not oral sex is practised should be obtained.

Functional evolution of the Myb oncogene family.

Three Myb-related genes (A-Myb, B-Myb, and c- Myb) have been found in all vertebrates examined thus far including mammals, birds, and amphibians. Two invertebrates, the sea urchin and the fruit fly, have only one Myb-related gene. Our laboratory has used Drosophila as a model system to explore the function of its sole Myb gene. We have also reintroduced the three different vertebrate Myb genes into Drosophila in order to begin to understand how their different functions may have arisen following gene duplication during evolution.

Clonal growth in the Hamburger-Salmon assay improved by rat RBCs.

This study was devoted to finding ways to improve the colony forming efficiency of the Human Tumor Colony Assay. Rat erythrocytes incorporated into the feeder layer of the Hamburger and Salmon assay showed notable improvements in colony growth by three different tumor cell lines.
