Class II histocompatibility antigens on human dendritic cells.

Histocompatibility antigens of the HLA D locus on the surface of human dendritic cells (DC) were visualized in the electron microscope using immunogold labelling. DC from peripheral blood expressed DR that was frequently concentrated at junctions between aggregating DC and lymphocytes or DC and macrophages. Labelling with an antibody to DQ was more diffuse and was not concentrated at points of cell-cell contact. The D locus antibody RFD1 labelled DC in distinct patches that were sometimes located at points of cell contact. Upon labelling DC with antibody to DR and incubating the cells at 37 degrees, some label remained on the cell surface but some was found in deep channels which appeared to be formed between veils at the surface of the cell and became internalized in membrane-bound structures. Under the same conditions, gold bound to DQ molecules remained on the surface of DC. Gold labelling RFD1 also remained mainly on the cell surface but there was occasionally internalization of patches into the cells through depressions in the cell membrane. The changes in distribution of the label on warming the cells suggests that materials bound to different D locus products may be 'processed' differently.

Sperm maturation associated with subfertility following hyperbaric exposure of mice.

Mice exposed to 50 ATA heliox pressure during one spermatogenic cycle subsequently exhibit subfertility which can recover over an 8-wk period. This study was designed to investigate the changes in spermatid and testicular histology associated with these phenomena. Electron microscopy of testicular tissue from hyperbaric-exposed animals revealed a significant effect on spermatogenesis including disorganization and patchy necrosis of the epithelium. Two weeks after exposure to pressure, abnormalities were more difficult to find and tissues taken from animals 8 wk after exposure were indistinguishable from normal. These data directly confirm our earlier hypothesis that chronic hyperbaric exposure has a deleterious effect on sperm maturation, which is subsequently manifest as a failure in fertilizing capacity.

The clinical features of homozygous alpha 2(I) collagen deficient osteogenesis imperfecta.

The detailed clinical features and progress of a child with homozygous alpha 2(I) collagen deficiency are described. Clinically, the disease presents as severe progressive Sillence type III osteogenesis imperfecta. The main biochemical defect is the synthesis of an abnormal pro alpha 2(I) chain which does not associate with pro alpha 1(I) chains and therefore is not incorporated into triple helical trimers of type I procollagen which can be used to assemble collagen fibres.
