Indolamines in the cockroach Blaberus craniifer Burm. nervous system--II. Fed and crowded young males in comparison with females.

1. Indolamines were assayed by HPLC-ECD in nervous tissue of fed and crowded young males Blaberus craniifer Burm. 2. In males, as in females housed in the same conditions, levels are depending on both age and region (= ganglia) of the central nervous system. 3. Registered sex differences are discussed in terms of anatomical, physiological and behavioral sexual dimorphism.

Biogenic amines in newly-ecdysed cockroaches.

1. Simultaneous quantification (HPLC and electrochemical detection) of biological extracts have shown dopamine, N-acetyl dopamine, tryptophan, 5-hydroxytryptamine, a 5-hydroxyindolacetic acid-like substance in nervous tissue and hemolymph of Blaberus craniifer and Periplaneta americana. 2. 5-Hydroxytryptophan was only detected in head and thoraco-abdominal nerve cord. 3. Octopamine, but not N-acetyl-5-HT was quantified in the hemolymph.

3,4-Dihydroxyphenylacetic Acid↗

Indolamines and onset of vitellogenesis in the imaginal molt-decapitated cockroach Blaberus craniifer Burm.

1. The effects of 5-hydroxytryptamine, 5-hydroxyindoleacetic acid and N-acetyl-5-hydroxytryptamine on oocytes of Blaberus craniifer, in which vitellogenesis was prevented by imaginal molt decapitation, were investigated. 2. Sites binding anti-egg-protein antibodies were detected in the periphery of basal oocytes of treated females, with individual variability. 3. In this ovoviviparous cockroach, the onset of vitellogenesis may thus not be triggered solely by juvenile hormone, and indolamines may play a role in the uptake of haemolymphatic proteins by oocytes.

Indolamines in the cockroach Blaberus craniifer Burm. nervous system--I. Fed and crowded young females.

1. Indolamine levels were determined in the cerebral ganglion, the thoraco-abdominal nerve cord (except the last ganglion), and the 6th abdominal ganglion of females of Blaberus craniifer. 2. Measurements were made at the imaginal molt and on fed and crowded imagos at 10, 20 and 30 post-imaginal days. 3. Indolamines were found in the nervous system of young females, but 5-hydroxytryptophan was only detected in the thoraco-abdominal nerve cord. 4. Amine levels were related to the age of the cockroach, particularly during this period, to post-ecdysis events and ootheca formation.

Antiparasitic activity of Annona muricata and Annona cherimolia seeds.

Methanolic extracts of Annona muricata and A. cherimolia (Annonaceae) seeds were tested for antiparasitic activity against E. histolytica, N. brasiliensis, M. dessetae and A. salina. The acetogenins isolated from these extracts are found to be responsible for the important activity on infective larvae of Molinema desetae.

A new in vitro model of Entamoeba histolytica adhesion, using the human colon carcinoma cell line Caco-2: scanning electron microscopic study.

The human colon carcinoma cell line Caco-2, which is widely used to study the adhesion and cytotoxicity of enterobacteria, was used to investigate the adhesion of the trophozoites of Entamoeba histolytica. We observed a high percentage of adhesion of amoebae to Caco-2 cells. Scanning electron microscopy showed that amoebial membrane structures were involved in adhesion and the cytolytic action. These differentiated cells should prove to be a useful model system for investigation of the pathogenic action of amoebae.

[Innovation in antiparasitic agents].

The emergence of new parasitic diseases has encouraged studies on the elucidation of the mode of action of new and old antiparasitic drugs, and has led to the development of new chemical series. Resistance occurs rapidly when a single drug is used massively in human and animals. A better knowledge of the mode of action allow synergistic combination of compounds, slow-release devices proposed for cattle. Another alternative is to improve bioavailability of antiparasitic drugs by targetting at the parasite molecular level with prodrugs, drug-carriers as liposomes, nanoparticles. The use of new compounds should be expected with relative optimism.

[Comparative data on biogenic amines and GABA in three Rodents Filariae].

Detection and quantification of biogenic amines and GABA were carried out on three adult Filariae Litomosoides carinii, Acantocheilonema viteae and Molinema dessetae obtained from Rodents. Catecholamines were under the level of detection. Indolamines and GABA levels depend on species and sex. Results are discussed in relation to the parasitism and localization in the host.

[Effects of Saccharomyces boulardii yeast on trophozoites of Entamoeba histolytica in vitro and in cecal amebiasis in young rats].

Lyophilized and rehydrated Saccharomyces boulardii yeasts were administered to young rats previously inoculated into the cecum with Entamoeba histolytica. The numbers of diseased young rats and the severity of the infection, assessed from the appearance of the cecum and its content of mucus and amebae, were significantly reduced by the treatment. The lesions observed resembled those seen in the untreated controls, although healing was faster in the treated animals. Saccharomyces boulardii had no intrinsic amebicidal action in vitro.

[The research of filaricides for the control of african onchocerciasis].

Amongst several other chemical series, the epoxi and ethylenesulfonamides (which are more stable) have been selected after that we have shown good filaricidal activities on the Filaria Molinema dessetae. This has been confirmed on other models. Good activity was observed with O. gutturosa adult male in vitro, while microfilariae of O. lienalis were insusceptible to the selected compounds. The future of these products was discussed, in the prospect of their development and in relation with their mode of action and the other available pharmacological data.

Drug Evaluation, Preclinical↗

Synthesis and anthelminthic evaluation of 2-amidino-1,3,4-oxa and 1,3,4-thiadiazoles, structurally related to tetramisole.

N-(1,3,4-thiadiazol-2-yl) 1a-1k and N-(1,3,4-oxadiazol-2-yl-) amidines 2a-2g, N-(1,3-thiazol-2-yl) 3a, 3b and N-(1,3-benzothiazol-2-yl-)amidines 4a, 4b were synthesized and tested as anthelminthics, in vitro, against a free nematode (Rhabditis pseudoelongata), against infecting larvae of an intestinal parasite of rats (Nippostrongylus brasiliensis) and against infecting larvae of a filaria (Molinema dessetae).

[A new experimental model of cecal amebiasis in rats].

The young rat was used as an experimental model of caecal amebiasis by implanting, without injury of the caecum, a suspension of E. histolytica trophozoites gnotoxenic strain Rahman of high virulence. The degree of pathogenicity is demonstrated by the macroscopic aspect of caecum, by the density of trophozoites and by an anatomopathologic examination. As a correlation exits between all these macroscopic parameters and histology, a degree of illness is fixed which include several macro- and microscopic criteria. This model enables to observe the different phases of cicatrization and of recovering of young rats and so to appreciate the activity of antiamebic agents.

In-vitro evaluation of filaricidal activity of GABA and 1,3-dipalmitoyl-2-(4-aminobutyryl)glycerol HCl: a diglyceride prodrug.

A diglyceride ester of gamma-aminobutyric acid (GABA) has been synthesized and its filaricidal activity compared with GABA, and progabide in-vitro, on infective larvae and microfilariae of Molinema dessetae, a rodent filaria. GABA induced paralysis in infective larvae but was inactive on microfilariae. There were interactions between the culture medium and GABA. The ester drug at 0.1 mmol L-1 (1,3-dipalmitoyl-2-(4-aminobutyryl)glycerol HCl) was as active as progabide on infective larvae and hundredfold more potent than GABA. Its microfilaricidal activity at 1 mmol L-1 was lower than that progabide at 0.1 mmol L-1 but a delayed effect was observed. The data confirm filariae sensitivity to GABA derivatives.

[Synthesis of 2-aryl benzothiazoles and research on its antiparasitic activity].

Several 2-aryl benzothiazoles (with phenyl, naphthyl, thienyl and some oxygenated heterocycles) were synthesized. The anti-parasitic properties of these compounds were evaluated, in vitro and in vivo, against one Nematode Nippostrongylus brasiliensis and, in vitro against two Protozoaires Entamoeba histolytica and Trichomonas vaginalis. Only two compounds exhibited, in vitro a low nematicidal activity.

Macrofilaricidal activity of metabolites of diethylcarbamazine.

The antifilarial compound diethylcarbamazine (N,N-diethyl-4-methyl-2-piperazine, DEC) is known rather for its micro- than macrofilaricidal activity. But in some human filariasis i.e. loaiasis, lymphatic filariasis, the spectrum of DEC activities extends to adult filaria. The potential role of the metabolites of DEC in the action of the parent drug once it had been metabolized in the body of infected animals was investigated. The metabolites were evaluated in a new experimental model on which DEC is active: Molinema dessetae in its natural host, the rodent Proechimys oris. Experimental studies were carried out in vivo and in vitro, on microfilariae, infective larvae and adult filaria. Several other nematodes were also used. The metabolites were DEC itself, N-ethyl-4-methyl-1-piperazine-carboxamide (MEC) and their N-oxides, 4-methyl-piperazine-carboxamide and N,N-diethyl-1-piperazine-carboxamide. In vivo most of the metabolites were found active on microfilariae and both N-oxides active on adults and infective larvae. In vitro, the activity of the metabolites was observed only with high concentrations; the in vitro test could not be used as a screening method for antifilarial chemotherapy with piperazine derivatives. Infective larvae were the most sensitive stage. In the rodent and in man, the antifilarial action of DEC is swift and of short duration. This action is prolonged by the activity of metabolites, especially the N-oxides.

[After ivermectin, is it necessary to look again for a macrofilaricide?].

The use of ivermectin for mass treatment in human onchocerciasis is justified, because no other drug was available. But, as this compound is ineffective on adult filariae and as the emergence of drug resistance is quite possible, the evaluation of new chemical compounds for their macrofilaricidal activity should be continued. Several new compounds of interest are presented.

[A new series of antiparasitic organic arsenicals: the spiroarsoranes. Experimental trypanocidal activity].

Arsenical compounds are the main therapeutic way, effective in the neurological phase of trypanosomiasis. Unfortunately, their important toxicity prevents their easy administration. Structural cyclization of arsonic acid derivatives has led to a class of non-toxic compounds: spiroarsoranes. A 47 compound primary screening on an in vivo murine model of Trypanosoma brucei brucei resulted in the isolation of a very effective derivative after a single subcutaneous injection of 30 mg.kg-1 body weight (Chemotherapeutic Index = 21). In this study, trypanocidal activity of this compound was evaluated on an in vivo sheep model of trypanosomiasis by T. brucei brucei, according to experimental model of the Institute of Neurological Epidemiology and Tropical Neurology of Limoges (France). A single subcutaneous injection of 30 and 100 mg.kg-1 b.w., and 30 and 60 mg.kg-1 b.w. was given respectively during first and second phase of trypanosomiasis. Ovine pharmacokinetics of this compound will be evaluated in a further study.
