[Identification of latent forms of Chlamydia trachomatis in the conjunctiva signifying "new interpretation of ocular chlamydia infections"].

Chlamydia trachomatis (CT) sérovar D through K was demonstrated with electron microscopy and biology in 114 patients with bilateral dry eyes. Long time ago latent CT was recognised in conjunctival epithelium of vernal kerato conjunctivitis patients. CT was found in conjunctival section of patients with an extinct trachoma. Aberrants forms of chlamydia bear the responsibility of this long lasting infection. Reticuled body does not product elementary bodies. It becomes an aberrant body which could remain latent during years. DIF and others classical investigations are negatives because abberrant chlamydiae have lost most part of their MOMP albeit keeping immunopathogenicity.

[In situ hybridization of cells infected by Chlamydia trachomatis].

Hybridization in situ of chlamydia trachomatis allows identification of the bacteria. Moreover exact chlamydia trachomatis localization at tissue level can be demonstrated. It seems to us very important to know for instance in conjunctiva were chlamydia trachomatis are located. We used DNA DIGOXIGENIN labelled plasmidic probe of 503 b.p. Detection of chlamydia trachomatis on paraffin embedded cells becomes possible. Specificity is 100% as plasmidic sequence is not found in other bacteria. Sensitivity seems to be good quality on experimental model. At present time, it is under clinical evaluation. This method could complete PCR for chlamydia detection in case of controversial diagnosis.

[Permanence of Chlamydia of different serotypes in tissues (methodology)].

Immunofluorescence detection of C. trachomatis requires trained personnel because the threshold between positive and negative reaction is not obvious. Patients antibodies must cover EB and thus prevent binding with Mab. This problems have stimulated research on Chlamydiae probes DNA hybridization technology was applied and the specificity of the detection of CT was improved. Moreover, a polymerase chain reaction amplification of a fragment of about 1,200 bp of the M.O.M.P. gene followed by restriction endonuclease digestion with selected enzymes allows differentiation of serovars. This method seems to be promising for epidemiologic studies.

[Diagnosis of ocular chlamydiosis by gene amplification (polymerase chain reaction or P.C.R.)].

Identification of conjunctival chlamydioses could be very difficult since this disease is extremely latent. Moreover previous instillation of topical antibiotic provokes a negativation of D.I.F. and of cultures. P.C.R. is very useful, it was demonstrative in 41 cases out of 47 cases; D.I.F. was positive only in 18 cases. P.C.R. is a very promising method to recognize chlamydiae; identification of different serovars is processing.

Modification of the selected method for the determination of serum iron. Substitution of bathophenanthroline by ferene S.

The previously selected method (1977) for the measurement of iron in human serum has been modified by the working group on Iron and Iron-Transport Proteins (Sociéte Française de Biologie Clinique). Ferene S is used as the chromogen, it is more sensitive and cheaper than bathophenanthroline. The sample size has been reduced from 2 to 0.5 ml. No interference could be shown with high concentrations of bilirubin, copper or hemoglobin.

[Permanence of chlamydia in trachomatous tissue].

Electron-microscopic examination of conjunctival and corneal biopsies from six apparently healed trachomatous patients demonstrated chlamydiae in situ. Nine previously trachomatous patients were submitted to the same biopsies; all biopsies were positives. After two months specific anti-chlamydial treatment, we observed the same result before anterior segment surgery. Nevertheless surgery was performed without consequences except for the two corneal grafts which failed. We must ask for two questions: how long chlamydial particles persist in trachomatous tissue? What is the real efficacy of topical anti-chlamydial drugs?

Frequency of chlamydia in patients with vernal keratoconjunctivitis.

Ten years ago, we surmised that VKC could be provoked by chlamydia. 34 patients without any contamination of trachomatous origin were recently studied. We performed examinations of biopsies (histology and ultrastructure), serology, tissue group and specific allergen skin tests. Our findings indicate that Chlamydia could be an allergen provoking VKC in atopic patients.

Long-term mucosal alterations by sennosides and related compounds.

Sennosides and related compounds are presumed to be severe cell poisons after prolonged ingestion. Some histological and ultrastructural studies in animals and man with such laxative misuse have revealed myenteric plexus and colonic epithelium injuries; but others have failed to point out identical data. In a first histological and ultrastructural study in mouse, we were unable to find any intestinal mucosa injury after long-term sennoside ingestion. In a second long-term experiment, we compared the effects of sennosides and 1,8-dihydroxyanthraquinone (synthetic anthracene derivative) on the mouse jejunum and colon. Electron microscopic observations showed nervous myenteric plexus abnormalities only in 1,8-dihydroxyanthraquinone-treated animals. These results suggest that sennosides have a good intestinal mucosa tolerance as opposed to aglycosidic-related compounds.

[Microscopic analysis of the effect of mercuric salts on the parasite concentration in feces].

The fixation of mercuric ions on the uterin layer of the Ascaris lumbricoides egg wall is proved by light microscopic histochemical technics and electron microscopic microanalysis. These observations support the hypothesis of a possible reaction between the egg surface components and those of the dilution reagents. This reaction is used in the methods of concentration in coprology. In the case of mercuric salts, the free thiol components which are missing in all the Ascaris egg wall layers are not the factors of the reaction.

Ultrastructure of mouse intestinal mucosa and changes observed after long term anthraquinone administration.

In an attempt to study the relative toxicity of anthraquinonic laxatives on intestinal mucosa, we compared in mice the effects of fruit pulp containing sennosides A and B with those of a free anthraquinone, 1-8 dihydroxyanthraquinone. Observations have been made with transmission electron microscopy (EM) after 16 weeks of treatment with the two drugs. Although the doses used in this study were equipotent in terms of laxative activity, no damage to the intestinal tissue was observed with the sennosides. A number of changes, however, were detected in intestinal nervous tissues of all the animals treated with 1-8 dihydroxyanthraquinone, mainly in the form of vacuolisation of the axons, formation of lysosomal structures and in some cases appearances of fibrillar degeneration.
