Structure of the N-terminal region of Haemophilus influenzae H10017: implications for function.

Haemophilus influenzae is a gram-negative pathogen that causes infections ranging from asymptomatic colonization of the human upper respiratory tract to serious invasive diseases such as meningitis. Although the genome of Haemophilus influenzae has been completely sequenced, the structure and function of many of these proteins are unknown. H10017 is one of these uncharacterized proteins. Here we describe the three-dimensional solution structure of the N-terminal portion of H10017 as determined by NMR spectroscopy. The structure consists of a five-stranded antiparallel beta-sheet and two short alpha-helices. It is similar to the C-terminal domain of Diphtheria toxin repressor (DtxR). The C-terminal portion of H10017 has an amino acid sequence that closely resembles pyruvate formate-lyase--an enzyme that converts pyruvate and CoA into acetyl-CoA and formate by a radical mechanism. Based on structural and sequence comparisons, we propose that the C-terminus of H10017 functions as an enzyme with a glycyl radical mechanism, while the N-terminus participates in protein/protein interactions involving an activase (iron-sulfur protein) and/or the substrate.

The occurrence of Dirofilaria immitis in dogs in South Australia.

Heart, lung and samples of blood from 230 dogs were examined for infections of filarial parasites. Dirofilaria immitis worms and microfilariae were detected in one dog. Blood samples from a further 1428 dogs were examined for microfilariae and 22 were found to be infected. Eighteen dogs were infected with D immitis microfilariae and four with Dipetolonema reconditum microfilariae. The histories were available for 18 of the dogs infected with heartworm and only seven dogs had not travelled outside South Australia. It was concluded that heartworm infection was endemic in South Australia but the apparent prevalence was only about 1%.

Effects of capture on hematological values and plasma cortisol levels of free-range koalas (Phascolarctos cinereus).

Eight free-range koalas (Phascolarctos cinereus) were captured on Kangaroo Island (South Australia) and later transported to the Adelaide Zoological Gardens. Blood samples were collected at the time of capture and at 6 hr, 24 hr and 7 days later. Routine hematological analyses were performed and plasma cortisol levels determined. Significant differences were observed between sampling times for erythrocyte numbers, hemoglobin concentrations, packed cell volumes, mean cell volumes, mean cell hemoglobin concentrations, numbers of leukocytes, neutrophils, lymphocytes and lymphocyte:neutrophil ratio (P less than 0.05). No significant differences were detected between sampling periods in the numbers of monocytes (P = 0.707), eosinophils (P = 0.174) or plasma cortisol levels (P = 0.192).

Effects of prolonged administration of triazolam on adenosine A1 and A2 receptors in the brain of rats.

Continuous subcutaneous administration of triazolam, a benzodiazepine with short plasma half-life, for 10 days either decreased (31%, 2 mg/day) or increased (15%, 0.5 mg/day) radioligand binding to adenosine A2 receptors in the rat striatum. In a similar manner, we have shown previously that diazepam (5-10 mg/day), a benzodiazepine with a long plasma half-life attenuated radioligand binding to adenosine A2 receptors in the rat striatum by 45-25%.
