Active-site and membrane topology of the DD-peptidase/penicillin-binding protein no. 6 of Enterococcus hirae (Streptococcus faecium) A.T.C.C. 9790.

The membrane-bound 43,000-Mr penicillin-binding protein no. 6 (PBP6) of Enterococcus hirae consists of a 30,000-Mr DD-peptidase/penicillin-binding domain and a approximately 130-residue C-terminal appendage. Removal of this appendage by trypsin proteolysis has no marked effect on the catalytic activity and penicillin-binding capacity of the PBP. Anchorage of the PBP in the membrane appears to be mediated by a short 15-20-residue stretch at the C-terminal end of the appendage. The sequence of the 50-residue N-terminal region of the PBP shows high degree of homology with the sequences of the corresponding regions of the PBPs5 of Escherichia coli and Bacillus subtilis. On this basis the active-site serine residue occurs at position 35 in the enterococcal PBP.

Ascorbic acid, HDL cholesterol, and apolipoprotein A-I in an elderly Chinese population in Boston.

The relationships between plasma HDL cholesterol, apolipoprotein A-I, and plasma ascorbic acid were examined in 146 women and 92 men in a Chinese population in Boston, aged 60 and above. A significant partial correlation (r = 0.21, p = 0.015) between plasma ascorbic acid and plasma HDL cholesterol was observed among (predominantly nonsmoking) females after the effects of potentially confounding variables were taken into account. A similar relationship among 26 nonsmoking men was suggestive but inconclusive; there was no evidence of a relationship among 66 male smokers. The partial correlation between plasma apolipoprotein A-I and plasma ascorbic acid was marginally significant for women (r = 0.22, p = 0.057, n = 87) but was highly significant for men (r = 0.43, p = 0.007, n = 51). The correlation coefficient for men was highly dependent on smoking status, being 0.80 for those who never smoked and almost zero for current smokers.

Cytogenetic study of parthenogenetically activated bovine oocytes matured in vivo and in vitro.

Three experiments were performed to evaluate the potential for parthenogenetic activation of bovine oocytes in in vitro fertilization systems and to determine the chromosome complement of the resulting parthenogenotes. In the first experiment, immature oocytes from slaughtered cattles were matured in vitro in Defined Medium (DM) for 24 h to simulate in vitro fertilization conditions. Subsequently, a portion was fixed, and the remainder were transferred to rabbit oviducts. Oocytes were then cultured for 6-8 h or for 24 h with Colcemid present during the last 6 to 8 h and fixed on slides and examined. In the second experiment, mature oocytes were collected from the preovulatory follicles, and the oocytes were subjected to the same culture as in experiment I. In the third experiment, oocytes were treated as in experiment II, except that instead of transfer to rabbit oviducts, they were cultured an additional 48 h in vitro. In experiment I, 131 oocytes were fixed after culture in DM. Of the 79 oocytes analyzed in the pre-rabbit group, 71 (90%) were at the second meiotic metaphase (MII), and 8 (10%) were at pre-MII stage; none were activated. After transfer to rabbits, 291 were fixed. Of these, 80 were analyzed; 37 (46.3%) were MII, 7 (8.6%) were pre-MII, and 36 (45%) were activated. Of the 36 activated oocytes, 26 (72.2%) were haploid, 4 (11.1%) were diploid, 1 (12.8%) was tetraploid, and 5 (13.8%) were in the process of endoreduplication. In experiment II, 51 oocytes were fixed after culture in DM of which 36 (70.6%) could be analyzed; 30 (83.3%) were MII, and 6 (16.7%) were pre-MII. After culture in the rabbit, 68 were fixed of which 27 (39.7%) could be analyzed. Of these 27, 20 (74.1%) were MII, and 7 (25.9%) were activated; 6 were haploid, and 1 was endoreduplicating. In experiment III, 30 oocytes were fixed at the end of the culture period; only 10 could be analyzed of which 8 (80%) were MII and 2 (20%) were pre-MII. In all, 46% of in vitro and 26% of in vivo matured oocytes were activated, based on chromosomal analysis. Of those activated, the majority (74.4%) were haploid, suggesting that activation occurs at or after completion of MII. Endoreduplication appears to be one of the mechanisms leading to the formation of diploid and polyploid parthenogenotes.

Primary structure of the Streptomyces R61 extracellular DD-peptidase. 2. Amino acid sequence data.

In order to confirm the Streptomyces codon usage, the Streptomyces R61 DD-peptidase was fragmented by cyanogen bromide cleavage of the carboxymethylated protein, trypsin digestion of the carboxymethylated protein and trypsin digestion of the protein treated with beta-iodopenicillinate and endoxo-delta 4-tetrahydrophthalic acid. The isolated peptides, which altogether represented more than 50% of the polypeptide chain, were sequenced. The data thus obtained were in excellent agreement with the primary structure of the protein as deduced from the nucleotide sequence of the cloned gene. Though a weak acylating agent, beta-iodopenicillanate reacted selectively with the active site of the DD-peptidase and formed an adduct which mas much more stable than that formed with benzylpenicillin, thus facilitating the isolation and characterization of the active-site peptide.

Dietary calcium intake and bone loss from the spine in healthy postmenopausal women.

Controversy exists over the relationship between calcium intake and axial bone loss. We measured bone density of the spine (L2-4) by dual photon absorptiometry (DPA) two times, 7 mo apart, in 76 healthy postmenopausal women aged 40-70 y. Ca intake was assessed by food-frequency questionnaire. Women with a Ca intake less than 405 mg/d lost spine density at a rate that was significantly greater than that of women with an intake greater than 777 mg/d, p = 0.026. There appears to be a threshold of Ca intake below which increased Ca in the diet is likely to be beneficial in reducing spine mineral loss.

Poststroke depression and lesion location. An attempted replication.

Recent evidence suggests that poststroke depression is associated with the location of the brain infarct, proximity to the frontal pole being associated with greater depression following left hemisphere injury, and an opposite relationship being seen with injury to the right hemisphere. This study was designed to replicate and extend previous investigation of this issue. Depression was assessed following recent stroke in patients with localized right (n = 16) or left (n = 19) hemisphere lesions. No overall differences between these groups were found on several measures of depression. Although there appeared to be a negative linear relationship between severity of depression and distance of the lesion from the frontal pole in left hemisphere patients, this association failed to attain statistical significance. A curvilinear relationship was evident in right hemisphere patients, with anterior and posterior lesions associated with high depression scores. These data support the notion that lesion location is associated with the severity of poststroke depression, although the nature of this association may be more complex than has previously been suggested.

Purification of Streptococcus faecium penicillin binding protein 5, a multifunctional penicillin-binding protein.

Penicillin-binding protein 5 of Streptococcus faecium has been solubilized and partially separated from other membrane proteins by covalent affinity chromatography. PBP 5 was successively purified to homogeneity by resolution on SDS-polyacrylamide gel, elution and renaturation of penicillin-binding activity. The purification procedure does not alter the properties that the protein exhibits in the membranous environment.

Phospholipase A2 activity in rat and human lymphocytes.

Partial characterization of phospholipase A from rat and human lymphocytes showed that it was much less active in man than in rat. The use of phosphatidylethanolamine labelled in the 2 position as substrate established that phospholipase A activity was 2 acyl-specific. It was maximal at pH 7.0 to 8.0, totally Ca2+ dependent and inhibited by detergents and Indomethacin.

Soluble polyglycans enhance recovery from cobalt-60--induced hemopoietic injury.

Six soluble polyglycans (glucan-C, glucan-F, glucan-S, krestin, lentinan, and schizophyllan), two soluble polymannans (mannan-A and mannan-R), and one soluble polyfructan (levan) were assayed for their ability to enhance hemopoietic recovery in C3H/HeN mice when administered either 1 h before or 1 h after a 6.5-Gy dose of cobalt-60 radiation. Hemopoietic recovery was measured by the endogenous spleen colony assay and was compared with recovery in both radiation control mice and irradiated mice treated with glucan-P (a particulate polyglycan previously shown to enhance recovery from radiation-induced hemopoietic injury). Compared with radiation controls, when administered before irradiation, mannan-A, glucan-F, and glucan-S enhanced endogenous colony formation 4.2-5.1-fold (equivalent to glucan-P), and levan and schizophyllan approximately 2.7-fold. Lentinan, krestin, mannan-R, and glucan-C did not enhance hemopoietic recovery above radiation controls under these conditions. When polyglycan administration was delayed until after irradiation, endogenous colony formation was enhanced 3.0-3.9-fold by mannan-A, schizophyllan, glucan-S, krestin, and glucan-F (at least comparable with glucan-P) but not at all by mannan-R, levan, lentinan, or glucan-C.

[Malignant mesenchymatous hamartoma of the liver in adults. Apropos of a case].

Clinical and pathological findings in a forty-eight-year-old woman with malignant mesenchymatous hamartoma of the liver (embryonic sarcoma) are reported. Special emphasis is put on the monomorphic leiomyosarcomatous aspect of the peritoneal metastases, and on the presence of pseudo-tumorous foci abounding in plasmocytes and centered by recent phlebitis.
