Transmission of non-A non-B hepatitis to chimpanzees by factor-IX concentrates after fatal complications in patients with chronic liver disease.

6 cases of non-A non-B hepatitis which followed administration of four different batches of concentrates of coagulation factor IX from commercial and non-commercial sources are described. Of 17 patients who received the concentrate on account of chronic liver disease, 4 developed hepatitis, and in 3 of these the illness proved fatal. The incubation periods ranged from 42 to 103 days (mean 65 days). 3 chimpanzees were inoculated with concentrate from the same batch used on the above patients, a further commercial batch upon which no adverse reactions had been reported, and plasma from a known non-A non-B carrier. All developed hepatitis after 10 weeks' incubation. Liver biopsy when serum-aminotransferase was at its highest level showed features consistent with acute hepatitis. As in the patients, viral markers for hepatitis A and B, cytomegalovirus, and Epstein-Barr virus were unchanged.

Studies on the mechanism of action of Albizzia lebbeck, an Indian indigenous drug used in the treatment of atopic allergy.

Studies were conducted on the decoction of the bark of Albizzia lebbeck which has been in use by Ayurvedic physicians for bronchial asthma and eczema. The effect of A. lebbeck was studied on the degranulation rate of sensitized peritoneal mast cells of albino rats when challenged with antigen (horse serum). Triple vaccine was used as adjuvant. Disodium cromoglycate (DCG) and prednisolone were used for comparison. Drugs were given during the first or second week of sensitization and the mast cells studied at the end of the second or third week. Serum from these rats was used to passively sensitize recipient rats whose peritoneal mast cells were then studied. The in vitro effects of A. lebbeck and DCG on the degranulation rate of the sensitized mast cells were also studied. The results show that A. lebbeck has a significant cromoglycate-like action on the mast cells. In addition, it appears that it inhibits the early processes of sensitization and synthesis of reaginic-type antibodies. If A. lebbeck is given during the first week of sensitization it markedly inhibits the early sensitizing processes, while if given during the second week it suppresses antibody production during the period of drug administration. The active ingredients of the bark appear to be heat-stable and water-soluble.

Modification of intramuscular pH oscillations in the isolated perfused rat heart by different interventions.

Changes in the intramuscular pH oscillations were examined by the use of an antimony electrode upon perfusing the isolated rat heart under different experimental conditions. The pH oscillations were decreased upon perfusing the hearts with Na+- or Ca2+-free medium and increased upon perfusing with K+-free medium. Increasing the temperature of perfusion medium from 25 to 40 degrees C or omitting glucose from the perfusing medium decreased the magnitude of oscillations. On the other hand, complete interruption of the perfusion flow resulted in an increase in the amplitude of pH oscillation. An initial increase followed by a decrease in the pH oscillation was seen when hearts were perfused with medium containing lactic acid at pH 6.6. These results suggest that pH oscillations reflect fluctuations in myocardial metabolism.

Detection and genetic counselling of subclinical and carrier states in Duchenne muscular dystrophy.

23 potential carriers of Duchenne muscular dystrophy (DMD) and 20 of their apparently healthy brothers were studied for evidence of any subclinical form of the disease. The results of the study confirmed that it was possible to confirm the diagnosis of a 'high genetic risk' carrier by integrating the results of clinical studies, estimation of basal serum CPK, steroid-CPK test, EMG and ECG observations. Similarly, the subclinical state of DMD could also be detected with certainty in 10% of the brothers of DMD carriers. The results may be applied for genetic counselling to bring down the incidence of the disease in the community.
