Triterpene and steroid saponins isolated from two Melilotus species.

Four new triterpene saponins of the oleanan series-melilotosides I, II, III, IV and nonglycosylated soyasapogenol B have been isolated from roots of Melilotus albus Medik. (Leguminosae) and four steroid saponins were isolated from M. tauricus (Bieb.) ser. seeds. The chemical structure of the saponins and of their aglycone was proved based on chemical transformations and spectral data. Melilotoside I has the structure of 3-O-[a-L-arabinopyranosyl] soyasapogenol B, melilotoside II that of 3-O-[beta-D-galactopyranosyl-(1-->2)-O-alpha-L-arabinopyranosyl] soyasapogenol B, melilotoside III that of 3-O-[alpha-L-rhamnopyranosyl-(1-->2)-O-beta-D-galactopyranosyl-(1-->2)-O -alpha-L-arabinopyranosyl] soyasapogenol B, and melilotoside IV that of 3-O-[alpha-L-rhamnopyranosyl-(1-->2)-[alpha-L-arabinopyranosyl-(1- ->3)]- O-beta-D-galactopyranosyl-(1-->2)-O-alpha-L-arabinopyranosyl soyasapogenol B. The steroid saponins have been identified as 3-O-beta-D-glucopyranoside-diosgenin (V); 3-O-alpha-L-rhamnopyranosyl (1-->2)-[alpha-L-rhamnopyranosyl(1-->3)-O-beta-D-glucopyranoside- diosgenin (VI),26-O-beta-D-glucopyranoside (25R)-furost-5-ene 3beta,22alpha, 26-triol (VII), and 3-O-alpha-L-rhamnopyranosyl(1-->4)-alpha-O-beta-D- glucopyranoside(25R)-furost-5-ene 3beta,22alpha,26-triol-26-O-beta-D- glucopyranoside (VIII).

Steroidal glycosides from Nicotiana tabacum L. seeds and their biological activity.

Steroidal glycosides of the spirostane and furostane series have been isolated from the methanolic extract of Nicotiana tabacum L. seeds by using gel filtration on silica gel and chromatography on a silica gel column. They were tentatively named nicotianosides A (1), B (2), C (3), D (4), E (5), F (6), and G (7). It has been shown that nicotianosides A (1), B (2), C (3), and D (4) are 3-O-beta-D-glucopyranoside-; 3-O-alpha-L-rhamnopyranosyl (1-->2)-O-beta-D-glucopyranoside-; 3-O-¿[alpha-L-rhamnopyranosyl (1-->2)].[alpha-L-rhamnopyranosyl (1-->4)]-beta-D-glucopyranoside¿: 3-O-¿[alpha-L-rhamnopyranosyl (1-->2)-alpha-L-rhamnopyranosyl (1-->3)]-alpha-L-rhamnopyranosyl (1-->4)]-beta-D-glucopyranoside¿: neotigogenin, respectively. The structures of nicotianosides E (5), F (6), and G (7) have been established as: 3-O-alpha-L-rhamnopyranosyl (1-->2)-O-beta-D-glucopyranoside; 3-O-¿[alpha-L-rhamnopyranosyl-(1-->2)]-[alpha-L-rhamnopyranosyl (1-->4)]-beta-D-glucopyranoside¿: 3-O-¿[alpha-L-rhamnopyranosyl (1-->2)-[alpha-L-rhamnopyranosyl (1-->3)]-alpha-L-rhamnopyranosyl (1-->4)]-beta-D-glucopyranoside): (25S)-5-alpha-furostane-3 beta, 22 alpha, 26-triol [-26-O-beta-D-glucopyranoside]. Growth-stimulating activities, prevention of virus attacks on tobacco, and a possible mechanism of action were established for the nicotianosides.

Steroidal glycosides from Petunia hybrida L. seeds and their biological activity.

A new plant source containing steroidal glycosides of the furostane and spirostane series has been shown. Ten individual steroidal glycosides (petuniosides) of the spirostane and furostane series have been isolated from Petunia hybrida L. seeds. They differ in the structures of aglycone and oligosaccharide chain, four of them are novel compounds, and their chemical structures have been established. Petuniosides A, C, and E are 3-O-beta-D-galactopyranoside spirostane; 3-O-beta-D-glucopyranosyl (1-->4)-beta-D-galactopyranoside spirostane; and 3-O-beta-D-glucopyranosyl-(1-->2)-beta-D-glucopyranosyl (1-->4)-beta-D-galactopyranoside of (25R)-5 alpha-spirostan-3 beta-ol, respectively. Petuniosides B, D, and F are 3-O-beta-D-galactopyranoside spirostane; 3-O-beta-D-glucopyranosyl (1-->4)-beta-D-galactopyranoside spirostane; and 3-O-beta-D-glucopyranosyl(1-->2)-beta-D-glucopyranosyl (1-->4)-beta-D-galactopyranoside of (25R)-5 alpha-spirostane-2 alpha, 3 beta-diol, respectively. Petuniosides E and F are new compounds, and B, C, and D have been detected in the Petunia genus for the first time. Glycosides of the furostane series, petuniosides I and K, are derivatives of (25R)-5 alpha-spirostane-3 beta, 22 alpha, 26-triol, and K and M of (25R)-5 alpha-spirostane-2 alpha, 3 beta, 22 alpha, 26-tetraol. Petuniosides I and L are new compounds, and K and M are known, but have been detected in the Petunia genus for the first time. Growth-stimulating activity has been established for petunioside M. This glycoside has been shown to enhance cucumber resistance to cucumber green eye-sport mosaic virus.

Chemistry and biological activity of steroid saponins from Moldovian plants.

The main point is that these glycosides which act as bioregulators differ fundamentally from produces currently used in agriculture: all of them are natural products causing neither environmental pollution nor any damage to useful organisms. Their rates of application are milligrams per hectare instead of kilograms and tons of chemicals traditionally used in agriculture. Also, the glycoside bioregulators are very inexpensive products, being recovered from wastes of canning and pharmaceutical industries. Their application allows the pesticide pressure to be considerable lowered. Steroid glycoside bioregulators can be used as supplements to nutrient media for in vitro culture. The morphogenesis stimulation by glycosides is conditioned by their interaction with phytohormones and depends on the basic medium content. The regulatory effect of glycosides directs the morphogenesis to the increase of shoot formation in the latter stages of growth or to the further activation of callus formation. The stimulation of morphogenesis in vitro under the influence or glycosides and phytohormones of interaction can be explained by using two mechanisms; one involves connection with the permeability of cell membranes, and the other one is connected with the regulation of reactions during the subsequent morphogenetic stages.

[Structural traits of the antioxidant and fungicidal activity of steroid glycosides].

To investigate the origin of antioxidant and antifungal properties of steroid glycosides (SG), a logico-structural analysis of the structure-activity relationships has been performed for 70 compounds using a set of computer programs STRAC. The following structural features responsible for antioxidant activity are revealed: 1) furostanol type of aglycon; 2) the presence of more than 4 branched monosaccharides in the carbohydrate chain; 3) the presence of glucose at C26 and OH-groups in the genin part of SG. Most characteristic for antifungal activity is the fragment genin-C3-Gal-Glc-Glc-Rha..., as well as the spirostanol type of aglycon. The computer analysis provided the basis for predicting a type of biological activity for 12 new compounds of the SG class.

[Specific binding of cholesterol by enterosorbents].

Soluble polymers, modified by steroid glycosides digitonine or tomatonine (the "enterosorbents") bound cholesterol dissolved in alcohol. Rates of the binding depend on amount of the glycosides introduced into the sorbent. The digitonine or tomatonine enterosorbents decreased the cholesterol content in blood serum of hypercholesterolemic rats after per oral administration. At the same time, the enterosorbent containing digitonine decreased simultaneously both the content of liver tissue cholesterol and the content of atherogenic lipoproteins in rat blood serum.
